This barrier is present in arterioles, capillaries and venules and is formed at the level of adjacent endothelial cells, which are coupled to astrocytes, microglia, neurons and pericytes. The structure of this endothelial barrier is unique among endothelia of other organ systems and is composed of complexes made up of tight, gap and adherens junctions. In addition, it is the responsibility of the surrounding cellular elements to maintain the integrity of the junctional complexes and restrict the entry of substances from the blood into the brain. Changes in permeability of the BBB during physiologic and pathophysiologic conditions involve alterations in specific transporters at the level of the endothelium, activation of specific cellular second messenger pathways and/or the dissolution of the junctional complexes composing the BBB. This book focuses on various aspects that account for the formation and maintenance of the BBB, and on disease states that compromise this barrier.

Central nervous system (CNS) vasculature plays a second critical role as a barrier system, limiting brain entry of bloodborne ions, molecules, and cells. Indeed, CNS blood vessels are endowed with a series of unique properties that allow them to tightly regulate the extracellular environment of the brain, and together these properties are termed the "blood-brain barrier" (BBB). The barrier properties of CNS vasculature are critical for proper brain function; BBB dysfunction during disease or injury can lead to a cascade of consequences resulting in neuronal death. Several questions remain regarding how BBB properties are maintained in health and the mechanisms underlying their dysfunction in neuropathologies. The brain uses an extraordinary amount of the body's energy, and without constant blood supply from the vasculature, its functions quickly deteriorate. Indeed, neuronal activity leads to a temporary increase in local blood flow to provide neurons with sufficient energy and oxygen, and this phenomenon is known as neurovascular coupling (NVC). NVC has been shown to involve various cell types and signaling molecules, but it is still unclear to what extent endothelial cells, the cells that form the innermost walls of the vasculature, participate in NVC. This dissertation explores brain vasculature in health and disease. Chapter I focuses on BBB dysfunction in disease, identifying PDLIM1 as a marker for BBB dysfunction and characterizing its role in CNS endothelial cells. Chapter II focuses on the neurovascular unit, specifically whether microglia play a role in maintaining barrier properties in healthy vasculature. Chapter III demonstrates the dynamic nature of neurovascular cholesterol metabolism and how this metabolic pathway influences NVC.

When we first had the idea of afestschrift to mark Mike's retirement as Professor of Physiology at King's College London and the beginning of a new vie libre as scientist and yachtsman the plan was initially for a gathering of all of the many colleagues, collaborators and students who had worked with him over the years. However what we had not taken into account was the closeness of the international community of workers in the field, the widespread influence that Mike had wielded and the speed with which word would spread. We should have anticipated all three. The final outcome was an excellent symposium with the majority of the world's key workers either contributing or attending. It is a central feature of numerous post-embryonic processes, including tissue and organ growth and regeneration. It is also part of the pathology of tumour formation and certain inflammatory conditions. In recent years, comprehension of the molecular genetics of blood vessel formation has progressed enormously and studies in vertebrate model systems, especially the mouse and the zebrafish, have identified a common set of molecules and processes that are conserved throughout vertebrate embryogenesis while, in addition, highlighting aspects that may differ between different animal groups. The discovery in the past decade of the crucial role of new blood vessel formation for the development of cancers has generated great interest in angiogenesis (the formation of new blood vessels from pre-existing ones), with its major implications for potential cancer-control strategies. In addition, there are numerous situations where therapeutic treatments either require or would be assisted by vasculogenesis (the de novo formation of blood vessels). In particular, post-stroke therapies could include treatments that stimulate neovascularization of the affected tissues. The development of such treatments, however, requires thoroughly understanding the developmental properties of endothelial cells and the basic biology of blood vessel formation. While there are many books on angiogenesis, this unique book focuses on exactly this basic biology and explores blood vessel formation in connection with tissue development in a range of animal models. It includes detailed discussions of relevant cell biology, genetics and embryogenesis of blood vessel formation and presents insights into the cross-talk between developing blood vessels and other tissues. With contributions from vascular biologists, cell biologists and developmental biologists, a comprehensive and highly interdisciplinary volume is the outcome.
