Available in PDF, EPUB and Kindle. Book excerpt: During an immune reaction, T cell clones must be expanded by mitogenic cytokines such as interleukin-2 (IL-2). IL-2 is able to induce proliferation, protection from apoptosis, and differentiation of T cells. These cellular phenomena are mediated through signaling events initiated at the IL-2 receptor (IL-2R) and are associated with changes in the mRNA levels that encode necessary proteins. The following work examined the molecular signals initiated at the IL-2R and sought to understand their linkage to increases in mRNA levels and subsequent cellular responses, such as proliferation and protection from apoptosis. Understanding these links is vital to understanding the molecular events that control the immune response, as well as providing insight into the cell cycle and activated transcription. To study this problem, an IL-2 responsive system based on thymic blast cells (TBCs) was developed and characterized. This system has the advantage of yielding a large number of IL-2 responsive cells from primary human tissue. Using the TBCs it was shown that transcription of important cell cycle genes is linked to the IL-2R via the c-Ras/MAPK pathway. Additionally, this pathway is responsible for IL-2 mediated proliferation. A second pathway, mediated through PKC, is responsible for upregulation of anti-apoptosis genes and protection from apoptosis. TBCs were also used to show that IL-2 can increase mRNA levels of specific genes by releasing transcriptional attenuation. Together these results show that specific signaling pathways can be linked to increases in specific mRNAs and suggest at least one mechanism of this upregulation. These results also show that increased expression of certain mRNAs may explain specific cellular responses. Finally, this work provides direction for future research that should yield more interesting insights into IL-2 mediated control of transcription.

Book excerpt: Cells sense and respond to chemical and physical stimuli through signal transduction pathways, which mediate cell proliferation, differentiation, migration, and survival. The cytokines interleukin-2 (IL-2) and interleukin-4 (IL-4) are key regulators of the adaptive immune system, particularly influencing the clonal expansion and differentiation of T cells. At least in culture, both synergistic and antagonistic effects of IL-2 and -4 co-stimulation have been reported; the antagonism, when observed, is thought to arise from the utilization of a common subunit shared by IL-2 and IL-4 receptors. We have sought to characterize IL-2 and IL-4 signaling at the level of intracellular pathways activated by these receptors. IL-2 receptors are known to activate the Ras/extracellular signal-regulated kinase (Erk) and phosphoinositide (PI) 3-kinase pathways as well as the STAT5 transcription factor. IL-4 is unique among cytokines in that it does not activate Ras⁄Erk; it does activate PI 3-kinase⁄Akt as well as a distinct STAT, STAT6. The HT-2 mouse T cell line responds to both IL-2 and -4. We found that IL-4 initially antagonizes, and later synergizes with, IL-2-stimulated HT-2 cell proliferation in a dose-dependent manner. IL-4 also stimulates cell adhesion in static cultures. At the level of intracellular signaling, IL-4 antagonizes IL-2-stimulated activation of Akt, possibly through competition for limiting amounts of common receptor subunit and/or PI 3-kinase. Because IL-4 alone does not promote any Erk activation, we were surprised to find that IL-4 enhances IL-2-stimulated activation of Erk. IL-2⁄IL-4 co-stimulation provokes transient activation of STAT5 and prolonged activation of STAT6. This extended STAT6 activation may be critical in the IL-2⁄IL-4 induced synergy in T cell growth. Currently, we are investigating the crosstalk between these pathways and their functional roles in IL-2 and IL-4-stimulated T cell responses.

Multiple mechanisms work in concert to ensure self-tolerance. Apart from purging the T cell repertoire from auto-reactive T cells via negative selection in the thymus dominant tolerance exerted by regulatory T cells plays a major role in tolerance imposition and maintenance. Among the various regulatory/suppressive cells hitherto described, CD4+CD25+ regulatory T cells (Treg) and interleukin-10 producing T regulatory 1 (Tr1) cells have been studied in most detail and are the subject of most articles in this issue. Treg, also called "natural" regulatory T cells, will be traced from their intra-thymic origin to the site of their action in peripheral lymphoid organs and tissues. The repertoire of Treg is clearly biased towards recognition of self-antigens, thereby potentially preventing autoimmune diseases such as gastritis and oophoritis. Regulatory T cells, however also control infections, allergies and tolerance to transplanted tissues and this requires their induction in the periphery under conditions which are not yet fully understood. The concept of dominant tolerance, by far not novel, will offer new insights and hopefully tools for the successful treatment of autoimmune diseases, improved cancer immunotherapy and transplant survival. The fulfillment of these high expectations will, however, require their unambiguous identification and a better understanding of their mode of action.
