Chromatin modifications are therefore often required to allow transcriptional activation. Here, we addressed the question whether global histone acetylation is involved in the regulation of transcriptional activation of class II promoters. We show that global histone acetylation has a role in activation of RNA polymerase II dependent transcription by modulating the inherent inhibitory effects of chromatin on the formation of the pre-initiation complex. In a second project we have developed a chromatin immunoprecipitation based assay that allows the study of transcriptional reinitiation "in vivo". Our results suggest the formation of a reinitiation scaffold on the GAL1, but not the ACT1 promoter. In a third project assessed whether TFIIB reaches promoters in the form of the holoenzyme "in vivo". Our data indicate that TFIIB does not assemble in a holoenzyme but associates with another, yet unidentified, factor(s) prior to promoter binding.

Book excerpt: Viral Polymerases: Structures, Functions and Roles as Antiviral Drug Targets presents in-depth study information on the structure and functions of polymerases and their roles in the lifecycle of viruses, and as drug targets. Viral polymerases constitute a vital component in the lifecycle of many viruses, such as human immunodeficiency virus (HIV), hepatitis viruses, influenza virus, and several others. They are essentially required for the replication of viruses. Thus, the polymerases that can be found in viruses (called viral polymerases) represent favorable targets for the design and development of antiviral drugs. Provides comprehensive, state-of-the-art coverage on virus infections, the virus lifecycle, and mechanisms of polymerase inhibition Analyzes the structure-activity relationships of inhibitors of each viral polymerase Presents a consistent and comprehensive coverage of all aspects of viral polymerases, including structure, function and their role as antiviral drug targets

Book excerpt: This report considers the biological and behavioral mechanisms that may underlie the pathogenicity of tobacco smoke. Many Surgeon General's reports have considered research findings on mechanisms in assessing the biological plausibility of associations observed in epidemiologic studies. Mechanisms of disease are important because they may provide plausibility, which is one of the guideline criteria for assessing evidence on causation. This report specifically reviews the evidence on the potential mechanisms by which smoking causes diseases and considers whether a mechanism is likely to be operative in the production of human disease by tobacco smoke. This evidence is relevant to understanding how smoking causes disease, to identifying those who may be particularly susceptible, and to assessing the potential risks of tobacco products.

Book excerpt: Maintenance of the information embedded in the genomic DNA sequence is essential for life. DNA polymerases play pivotal roles in the complex processes that maintain genetic integrity. Besides their tasks in vivo, DNA polymerases are the workhorses in numerous biotechnology applications such as the polymerase chain reaction (PCR), cDNA cloning, next generation sequencing, nucleic acids based diagnostics and in techniques to analyze ancient and otherwise damaged DNA (e.g. for forensic applications). Moreover, some diseases are related to DNA polymerase defects and chemotherapy through inhibition of DNA polymerases is used to fight HIV, Herpes and Hepatitis B and C infections. This book focuses on (i) biology of DNA polymerases, (ii) medical aspects of DNA polymerases and (iii) biotechnological applications of DNA polymerases. It is intended for a wide audience from basic scientists, to diagnostic laboratories, to companies and to clinicians, who seek a better understanding and the practical use of these fascinating enzymes.

There are a number of ways in which this state can be achieved and the majority of papers in this volume de scribe the effects of nucleotide pool imbalance on DNA replication and repair, and the induction of gene mutations in various organisms. This book grew out of a Conference on "Genetic Consequences of Nu cleotide Pool Imbalance" which was held at the National Institute of Environmental Health Sciences on May 9-11, 1983. The Organizing Com mittee consisted of Drs. Diana Anderson (BIBRA), Robert Haynes (York University), Bernard Kunz (NIEHS), Barry Glickman (NIEHS), and Frederick J. de Serres (NIEHS), Chairman. The meeting was org- ized by the staff of the Office of the Associate Director for Genetics and was financed solely by the National Institute of Environmental Health Sciences. This group of viruses includes human simplex type 1 and 2, Epstein–Barr virus, Kaposi's Sarcoma-associated herpesvirus, cytomegalovirus, HHV6A, 6B and 7, and varicella-zoster virus. The viral diseases and cancers they cause are significant and often recurrent. Their prevalence in the developed world accounts for a major burden of disease, and as a result there is a great deal of research into the pathophysiology of infection and immunobiology. Another important area covered within this volume concerns antiviral therapy and the development of vaccines. All these aspects are covered in depth, both scientifically and in terms of clinical guidelines for patient care. The text is illustrated generously throughout and is fully referenced to the latest research and developments.
