Book excerpt: The mouse was first used in immunological research by Paul Ehrlich in 1891 in an extraordinary series of experiments on the maternal transfer of antitoxic immunity. A short 22 years later in 1913 Halsey Bagg acquired a stock of albino mice from a commercial dealer and used them in a series of experiments on learning. Because he was interested in the genetics of intelligence, Halsey Bagg began breeding a pedigreed line of these mice that were subsequently named for him - Bagg Albino. Though Halsey Bagg is not credited with initiating the inbred strains of mice, his stock curiously has played an indisputably important role. Bagg Albinos were progenitors of the present day BALB/c family of sublines - the subject of this book. They were also used as one of the parents in the development of inbred strains A, CBA and C3H, three other very famous strains. Today the BALB/c mouse is among the five most widely used inbred strains in biomedical research and a particular favorite in immunology and infectious disease research. The hallmark of the BALB/c response to so many kinds of infections is susceptibility and sometimes an exaggerated susceptibility, but this paradoxically is not associated with immunodeficiency as BALB/c is an excellent responder to immuni zation. These characteristics have made the BALB/c mouse a model for identifying genes that determine susceptibility to infectious and neoplastic diseases. In 1985 the laboratory BALB/c mouse became 72 years old. The current filial generations are somewhere around 350 generations [MURPHY].

That is, the physiological properties and functions of tra- planted human tissues and cells can be analyzed in the mouse instead of using a living human body. It should therefore be possible to study the pathophysiology and treatment of human diseases in mice with good reproducibility. Thus, the hum- ized mouse can be used as a potent tool in both basic and clinical research in the future. The development of appropriate immunodeficient mice has been indispensable in the creation of the humanized mouse, which has been achieved through many years of efforts by several laboratories. The first stage on the road to the humanized mouse was the report on nude mice by Isaacson and Cattanach in 1962. Thereafter, nude mice were studied in detail by Falanagan and, in 1968, Pantelouris found that these mice have no thymus gland, which suggested that the mice lack transplan- tion immunity against xenografts such as human hematopoietic stem cells. At the Nude Mouse Workshops (organized by Regard, Povlsen, Nomura and colleagues) that were held nine times between 1972 and 1997, the possibility of creating a humanized mouse using nude mice was extensively examined. The results, however, showed that certain human cancers can be engrafted in nude mice, but unfortunately engraftment of normal human tissue was almost impossible.

It is clearly beyond the limitations of each volume in this series to comprehensively cover progress in each of these fields. Three general areas of contemporary investigations are discussed in the current issue: cellular events of B cell differentiation, including antigen recognition and respon siveness of B lymphocytes; regulation of the immune response by T cells, and the roles of this regulatory system in allotype suppression and in autoimm unity; and the genetic control of immune responses and its possible relation to disease pathogenesis. Analysis of responsiveness of B lymphocytes has been greatly aided by the availability of athymic nude mice. The immunological responsiveness of these mice to a wide range of stimuli has been reviewed and analyzed by H. H. Wortis. This article concludes with the provocative thesis that in the future "nudes will supply new rather than confirmatory data . . . (especially regarding) the develop ment of neoplasia.
