Book excerpt: The phenomenon of neural plasticity has been postulated to reflect changes in central nervous system circuitry in order to optimize the collection and/or interpretation of environmental information. The expression of immediate early genes (EEGs) is the first genomic response to environmental cues for change. Many EEGs act as transcription factors, in which case they could regulate the genetic mechanisms involved in more stable changes. Generally, long-term changes in circuit performance are mediated by proteins that result from the expression of late genes. The IEGs, nerve growth factor induced gene-A (NGFI-A) and activity regulated cytoskeletal protein (Arc) are currently under investigation as candidate-plasticity genes. NGFI-A is a nuclear protein that serves as a transcription factor for the Synapsins, a family of genes that has been strongly implicated as markers for neural plasticity. Arc has been hypothesized to mediate locally executed dendritic reconfigurations in response to changes in activity levels. In the present thesis, the response profiles for both these IEGs, along with the late genes growth associated protein-43 (GAP-43) and synapsin I, were evaluated for their role in mediating neural plasticity.

Book excerpt: Neuronal plasticity in the adult central nervous system is essential for dealing with environmental stimuli and injury-related changes. Kindling refers to the process whereby repeated, electrical stimulations of the brain leads to long-lasting or permanent changes in brain function. Kindling is typically established over several weeks using once daily stimulation, so called traditional kindling. It has been hypothesized that kindling may also be established within hours using different stimulation parameters, so called rapid kindling. However, studies investigating this possibility have provided conflicting results. In addition, while there has been intensive investigation into the mechanisms that mediate the pathological plasticity associated with kindling, much still remains to be elucidated. The expression of immediate-early genes (IEGs) is considered the first genomic response to underlie plastic changes in the CNS. Nerve growth factor induced gene-A (NGFI-A) and activity regulated cytoskeletal protein (ARC) are IEGs whose expression has been implicated in the transition from short-term to long-term synaptic plasticity. Based on these findings, the ability of rapid kindling to establish a kindled state and the response profiles of NGFI-A and ARC associated with rapid kindling were assessed.
