Available in PDF, EPUB and Kindle. Book excerpt: This volume deals with the structure and function of molecules that have, during the last decade, turned out to have a central role in immune responses. Trans plantation antigens were discovered and characterized by Gorer about 50 years ago, and the biological basis for the unequalled complexity of their variability between individuals within a species, in spite of extreme conservation between species, was the subject of intense research and discussion for many years. During the days of belief in "immune surveillance" against spontaneously developing tumors, it was suggested that histoincompatibility between members of one species would prevent cancer from being a contagious disease and thus a threat to the species. Immunologists involved in human transplantation had to learn and care about the complexity, especially after 1967, when it was found that HLA antigens were the products of the human MHC. Rejection of HLA-identical sib kidney grafts was so rare, even in those days, that cases of rejection were described in scientific papers.

Book excerpt: This publication covers the recent veritable explosion in knowledge regarding the genetic organization, structure, and function of the HLA class II region. It deals with the nature of diversity of HLA class II alleles, the molecular basis of this diversity, and the mechanisms by which HLA class II expression is regulated. Also addressed is the relationship of class II polymorphism to susceptibility to autoimmune diseases such as rheumatoid arthritis, insulin dependent diabetes mellitus, and pemphigus vulgaris. The book presents information on allele specific oligonucleotide (ASO) probes and their use in identifying susceptible individuals. It includes chapters which describe how cells transfected with HLA class II genes can be used for functional analysis. An appendix representing a compilation of all the known DR?? DQ?, DQ? sequences is included. This serves as an excellent source of comparison of newly derived sequences. This publication provides information on the enormous progress in the HLA field. It is intended for both the established investigator and the beginner.

Available in PDF, EPUB and Kindle. Book excerpt: Many common human diseases have a multifactorial origin: they are influenced by a person's genetic predisposition as well as by factors in the environment. This volume deals with the application of recombinant DNA techniques to the identification of diseases that have more than one inherited component. Focus is on the polygenic factors responsible for coronary atherosclerosis. Several other disorders having a polygenic origin are also discussed, including hypertension, diabetes mellitus, psychiatric diseases, and autoimmune (HLA-related) disorders. Problems raised by the study of different families or different populations are covered, as well as the possibility of applying molecular techniques to disease prevention--for example, through gene therapy. Also, some of the ethical issues that relate to human gene mapping are briefly explored.

Book excerpt: This symposium was established in 1976 for the purpose of bringing to gether once every two or three years, active investigators in the fore front of contemporary immunology, to present their findings and to discuss their significance in the light of current concepts and to identify important new directions of investigation. The founding of the symposium was stimulated by the achievement of major breakthroughs in the under standing of the immune recognition of proteins and peptides. We believed that these breakthroughs will lead to the creation of a new generation of peptides which should have enormous potential in biological, therapeutic and basic applications. This anticipated explosion has finally occurred and many applications of these peptides are now being realized. The main symposia topics of the fourth symposium were: T-cell recognition of proteins, structure and function of the T-cell receptor, presentation of protein antigens, recycling and activation of membrane receptor molecules, Ir-gene control of T-cell responses and methods of cell separation. The molecular features recognized by antibodies on proteins were the first immune recognition sites to be local ized and confirmed by synthetic peptides. The complete antigenic structures of several proteins have been defined, and individual antigenic sites have been described on many more proteins. More recently, major breakthroughs have been reported in the immune recognition of proteins by T cells.

Available in PDF, EPUB and Kindle. Book excerpt: The Second International Symposium on Narcolepsy was held at Fairchild Auditorium, Stanford University, on 6-7 July 1985 under the presidency of Drs. William C. Dement and Christian Guillemi nault. It succeeded the First International Symposium on Narco lepsy held in La Grande Motte, France, organized by Pierre Pas souant in July 1975 in commemoration of the 100th anniversary of the publication of Jean B. E. Gelineau's paper which proposed the naming of narcolepsy. At the second narcolepsy symposium, many important research reports on both basic and clinical aspects of narcolepsy were given by investigators from many countries of the world. Audience inter est was particularly attracted by the section on the relationship be tween HLA and narcolepsy, in which recent evidence that almost all narcoleptic patients are HLA-DR2 positive was reported by in vestigators from Japan, England, France, Canada, and the United States. The close relationship between the HLA antigens, hitherto considered as immune-related genetic markers encoded by genes on human chromosome 6, and narcolepsy appeared to open a new approach not only for the research of narcolepsy but also for the mechanism of sleep in general. Publication of all these new findings on the association of HLA and narcolepsy was considered; an outline was worked out and all the groups agreed to prepare a contribution covering the various aspects of this topic.
