This book, the Handbook of Traumatic Brain Injury and Neurodegeneration, has as its theme the marriage between neurodegenerative disease and neurotrauma through TBI surrogates such as sport, military service, and experimental models, and the legitimacy of that marriage. In the 32 contributions included here, this handbook not only explores the deleterious effects of genuine TBI, but also, and more importantly, the relationship between TBI and neurodegeneration. Controversy notwithstanding, there is much to be learned about the biological effects of TBI, substrates for long-term sequelae, the relationship between TBI and diverse neuropsychiatric disorders, and targets for therapy. The overall message to the neuroscience community from these papers may be a cautionary tale. The null hypothesis, that there is no causal relationship between TBI and progressive neurodegenerative disease, appears to be very much in play, and the book will be of interest to all those working in the field.

Book excerpt: Alzheimer's Disease (AD) and Parkinson's Disease (PD) are the two most common age-related chronic neurodegenerative diseases with significant global health burden. In addition to motor and cognitive impairments, these patients experience gut dysbiosis and gastrointestinal effects that may have a role in exacerbation of the neurological symptoms via the gut microbiome-brain axis. It has also been theorized that certain gut microbial-derived metabolites, indole metabolites of L-tryptophan are associated with neurodegeneration. Increasing evidence suggests that the bacteria that produce them are dysregulated in PD and AD patients. In this study, we used LCMS and Unifi analysis to measure and quantify the levels of these metabolites in fecal samples of rodents with PD or AD disease phenotype. In addition, we evaluated the levels of indole and its metabolites in PD animals treated with long-term L-DOPA which causes dyskinesia (LID) and LID rats treated with an experimental drug called S-SK609. The results showed decreases in the levels of indole-3-carbaldehyde and indole-3-acrylate with an increase in indole in PD animals; decreases in indole-3-carbaldehyde, indole-3-lactate and tryptophan in AD animals; and decreases in tryptophan, tryptamine, indole-3-lactate and indole-3-carbaldehyde with an increase in indole in LID animals. Treatment with the experimental drug S-SK609 had no ameliorating effect on metabolite levels in rodents with either LID or AD but it notably did not worsen the dysbiosis. Our findings validate that there is significant gut dysbiosis in neurodegeneration and measuring the levels of indole and its metabolites can serve as early biomarkers for neurodegeneration.
