Eight mature beef cows, each cow serving as her own control, were injected intravenously (i.v.) with saline on days 2 and 8 of the cycle (day of estrus=day 0 of the cycle), then with 10Oug GnRH on days 2 and 8 of the subsequent cycle. Jugular blood samples were taken immediately prior to an injection and at 15, 30, 45, 60, 120 and 240 min postinjection to characterize changes in serum luteinizing hormone (LH). Blood was also collected on alternate days after an injection until day 16 of the cycle, to characterize serum progesterone levels. Treatment with GnRH did not significantly alter serum progesterone concentrations. In Experiment 1, 10 heifers were assigned randomly into two groups with each heifer serving as her own control. Heifers in Group I (n=5) were injected intravenously (iv) with vehicle (saline) on day 2 of the cycle (day 0=day of estrus) followed by an iv injection of 100 ug GnRH on day 2 of the subsequent estrous cycle. Group II (n=5) heifers were similarly treated except injections were given on day 10 of the estrous cycle. All heifers were bled via the jugular vein at 15 minute intervals beginning 30 minutes prior to injection and for 3 hours after injection. Blood samples were also taken on alternate days after injection through day 16 of the cycle. Gonadotropin releasing hormone caused a significant release of luteinizing hormone (LH) on both treatment days with the peak occurring at 15 to 30 minutes postinjection. Treatment with GnRH on either day 2 or 10 of the cycle caused a reduction in serum progesterone levels on days 12, 14 and 16 of the cycle (Group I, control 3.99, 3.97, 4.07 vs treated 2.63, 3.45, 2.87; Group II, control 3.18, 3.82, 4.13 vs treated 2.50, 2.82, 3.17 ng/ml, respectively, P

Book excerpt: The objective of this study was to determine if a pre synchronization gonadotropin releasing hormone (GnRH) injection would increase conception percentages in postpartum anestrous angus beef cows and heifers. Sixty-four Angus females (42 multiparous cows and 20 heifers) from the Angelo State University's Management, Instruction and Research Center were randomly assigned to two treatments groups at the beginning of the study. The control group were administered a two shot prostaglandin estrous synchronization protocol. The treatment group were administered a GnRH injection thirteen days prior to the two shot prostaglandin synchronization protocol. However, only 30 females showed signs of estrus and were artificially inseminated. Results in cows and heifers were similar, and no differences were found among the two treatment groups (P>0.05). Differences might be noted with a larger number of females that show signs of estrus. Conception was recorded by the use of ultrasound, and is a viable management tool.

Book excerpt: The objectives of this study were to determine whether conception, ovulation rates, presynchronization rates, or follicle and CL characteristics were altered after modifying the Double-Ovsynch (DO) protocol to include human chorionic gonadotropin (hCG) compared to the conventional DO protocol in dairy heifers and lactating dairy cows. We hypothesized that replacing the first injection of gonadotropin releasing hormone (GnRH) in the DO protocol with hCG would increase the proportion of females that ovulate, thus improving the presynchronization rate leading to increased conception rates. Ovulation rates were increased in cows administered hCG compared to GnRH, but subsequent synchronization, and conception rates did not differ between treatments. A greater proportion of hCG-treated cows tended to fail to have undergone luteolysis compared to those treated with GnRH. In conclusion, no improvement in overall fertility was achieved by replacing the first injection of GnRH in the DO protocol with hCG.
