Book excerpt: European corn borer, Ostrinia nubilalis, is one of the most destructive insect pests of corn in the Midwest corn belt of the United States. The crystal protein toxin (Cry1Ab) expressed by the bacterium, Bacillus thuriginesis (Bt), specifically targets O. nubilalis gut and functions as "stomach poison". Transgenic corn expressing Cry1Ab can effectively control O. nubilalis larval infestation. However, O. nubilalis has the potential to develop resistance to Bt toxins which prompts concerns that transgenic corn will lose its control efficacy. Previous studies found that O. nubilalis gut serine proteases and membrane proteins were involved in Bt toxicity and resistance. Therefore, this study was to identify and characterize gut transcripts potentially involved in Bt toxicity and resistance, and to compare their transcriptional responses to the ingestion of Cry1Ab protoxin and transgenic corn leaves expressing Cry1Ab toxin. We identified and characterized 34 cDNAs encoding putative trypsins, chymotrypsins, and trypsin- and chymotrypsin-like protease homologs from O. nubilalis gut-specific expressed sequence tags (ESTs). Blast and phylogenetic analysis of their deduced amino acid sequences indicated that 15 were putative trypsins belonging to Try-G2 and Try-G3 groups (none of them was grouped in Try-G1), another 15 were putative chymotrypsins in one large group (CTP-G1), and the remaining four were serine protease homologs in Try-G4 and CTP-G2 groups, respectively. The existence of diverse trypsins, chymotrypsins and serine protease homologs in O. nubilalis could be an adaptation to different food sources and also a defense mechanism against plant-specific protease inhibitors and Cry toxins from transgenic corn. The expressions of four putative trypsins (OnTry4, OnTry5, OnTry6 and OnTry14) were up-regulated in O. nubilalis larvae after the ingestion of Cry1Ab protoxin. The differential expressions of these protease transcripts may implicate a link to Cry1Ab intoxication. To better understand the basic physiology of insect gut and Bt toxin interactions, we developed a high-resolution 8x15K cDNA microarray chips based on the larval gut specific ESTs. Each microarray contains 12,797 probes representing 2,895 unique larval gut transcripts. The expressions of 174 transcripts were differentially regulated at least 2-fold (P-value [equal to or less than]0.05) after the larvae fed Cry1Ab protoxin for 6 hours. Among them, 13 transcripts, putatively encoding eight serine protease, three aminopeptidase, one alkaline phosphatase, and one cadherin-like protein, were identified and further validated their expression ratios by quantitative PCR (qPCR). Three trypsin transcripts were up-regulated by more than 5-fold in larvae fed Cry1Ab protoxin. Sequence analysis suggests that they may have role in protoxin activation and toxin degradation. The transcriptional responses of laboratory-selected Cry1Ab resistant (R) and susceptible (S) strains of O. nubilalis to the ingestion of transgenic corn (MON811) leaves expressing Cry1Ab toxin were also examined. Even though R-strain larvae showed 200-fold resistance to Cry1Ab protoxin as compared with S strain, the larvae from both strains eventually died after fed transgenic corn leaves. However, the survival time of R-strain larvae was significantly different from that of S-strain larvae. The median lethal time (LT50) for the early third-instar larvae of R- and S-strains were 5.4 and 3.6 days, respectively. Furthermore, we identified 398 and 264 transcripts from the larvae of the S and R strains, respectively, with a significantly increased or decreased expression (expression ratio cut off [equal to or greater than]2.0 fold with p-value [equal to or less than]0.05) as compared with those in the larvae fed on non-transgenic corn leaves. The number of transcripts and their expression ratios of S-strain larvae are larger than these of R-strain larvae. These significantly differentially expressed transcripts may play important roles in influencing Cry1Ab toxicity from toxin degradation, toxin binding, to intracellular defense. Seventeen transcripts including serine protease and aminopeptdiase in S strain and nine in R strain were further analyzed by qPCR to validate their expression ratios. This study not only revealed information about the difference in the transcriptional responses of these genes to Cry1Ab between Bt-resistant and susceptible strains of O. nubilalis, but also provided new insights into potential interactions of the protoxin, toxin from transgenic corn with important proteins in the gut of O. nubilalis larvae.

Book excerpt: Genomic information for lepidopteran insects, particularly agricultural pest species, is very limited but urgently needed due to their economic importance and biodiversity. The huge economic losses ($ 1-2 billons / year) caused by the European corn borer (Ostrinia nubilalis, Hübner, ECB) makes this insect species one of the major pests of corn in the United States and western world. Management of ECB by conventional methods is limited but has had a great success by transgenic Bt (Bacillus thuringiensis) corn, which targets insect gut. However, the widespread use of Bt corn may lead to the development of Bt resistance in ECB. Knowledge of genes expressed in the insect gut is considered crucial for understanding basic physiology of food digestion, their interactions with Bt toxins and pathogens, and for discovering new targets for pest management. A large database of 15,000 expressed sequence tags (ESTs) was established from the ECB larval gut. To our knowledge, this database represents the largest gut-specific EST database from a lepidopteran pest. Analysis of 10 aminopeptidase-like genes between Cry1Ab-resistant and -susceptible ECB larvae revealed that aminopeptidase P-like (OnAPP) gene is a strong candidate for its role in Bt toxicity and resistance. The RNA interference mediated reduction in the transcript level of OnAPP gene in ECB larvae resulted in their reduced susceptibily to Cry1Ab. Analysis of the chitinase-like gene (OnCht) revealed its essential role in regulating chitin content of peritrophic membrane (PM). Our results suggest that OnCht may influence food digestion, nutrient absorption or movement of digestive enzymes through the PM and can be an important target for insect management. We also identified and characterized six genes involved in the innate immune defense response in ECB and showed that the expression of these genes were induced when challenged with bacteria. In addition to these results, this research generated significant genomic information for the development of microarray from the larval gut of ECB. The establishment of the feeding-based RNA interference technique could potentially help in delivering dsRNA orally to ECB for high throughput screening of effective genes to be targeted for insect pest management.

Native and transgenic variants of Bacillus thuringiensis (Bt) crystalline (Cry) toxins control larval lepidopteran feeding upon important agronomic crop plants. Invertebrate Bt resistance phenotypes arise via altered expression of midgut proteases involved in Cry toxin activation (trypsins) and degradation (chymotrypsins), reduced toxin binding to N-acetylgalactoseamine (GalNAc) modified peptide receptors (cadherin, aminopeptidase, or alkaline phosphatase), or receptor glycosylation pathway knockouts ([Beta]-1,3-galactosyltransferases; [Beya]3GalT5). Ostrinia nubilalis trypsin, chymotrypsin, cadherin, and [Beta]3GalT5 cDNA clones, transcript expression, and partial genomic genomic DNA copies were characterized. Mendelian inheritance of trypsin, chymotrypsin, and cadherin genomic DNA markers suggest association between allele segregation and Bt resistance phenotypes can be conducted. Movement of O. nubilalis moths was indirectly estimated by genetic similarity between North American subpopulations using mitochondrial and genomic microsatellite markers. Marker data suggested differentiation between two O. nubilalis ecotypes, univoltine and bivoltine, which may affect movement of Bt resistance phenotypes in northern regions of the Midwest United States.

Substantial progress will therefore be required in bridging the yield gap between what is currently achieved per unit of land and what should be possible in future, with the best farming methods and best storage and transportation of food, given the availability of suitably adapted cultivars, including adaptation to climate change. My book is divided into four parts: Part I is an historical introduction; Part II deals with the origin of genetic variation by mutation and recombination of DNA; Part III explains how the mating system of a crop species determines the genetic structure of its landraces; Part IV considers the three complementary options for future progress: use of sexual reproduction in further conventional breeding, base broadening and introgression; mutation breeding; and genetically modified crops.

After two decades of production, some groups and individuals remain critical of the technology based on their concerns about possible adverse effects on human health, the environment, and ethical considerations. At the same time, others are concerned that the technology is not reaching its potential to improve human health and the environment because of stringent regulations and reduced public funding to develop products offering more benefits to society. While the debate about these and other questions related to the genetic engineering techniques of the first 20 years goes on, emerging genetic-engineering technologies are adding new complexities to the conversation. Genetically Engineered Crops builds on previous related Academies reports published between 1987 and 2010 by undertaking a retrospective examination of the purported positive and adverse effects of GE crops and to anticipate what emerging genetic-engineering technologies hold for the future. This report indicates where there are uncertainties about the economic, agronomic, health, safety, or other impacts of GE crops and food, and makes recommendations to fill gaps in safety assessments, increase regulatory clarity, and improve innovations in and access to GE technology.

Book excerpt: Bacillus thuringiensis (Bt) has been used as a biopesticide in agriculture, forestry and mosquito control because of its advantages of specific toxicity against target insects, lack of polluting residues and safety to non-target organisms. The insecticidal properties of this bacterium are due to insecticidal proteins produced during sporulation. Despite these ecological benefits, the use of Bt biopesticides has lagged behind the synthetic chemicals. Genetic improvement of Bt natural strains, in particular Bt recombination, offers a promising means of improving efficacy and cost-effectiveness of Bt-based bioinsecticide products to develop new biotechnological applications. On the other hand, the different Bacillus species have important biotechnological applications; one of them is carried out by producing secondary metabolites, which are the study object of natural product chemistry. The amazing structural variability of these compounds has attracted the curiosity of chemists and the biological activities possessed by natural products have inspired the pharmaceutical industry to search for lead structures in microbial extracts. Screening of microbial extracts reveals the large structural diversity of natural compounds with broad biological activities, such as antimicrobial, antiviral, immunosuppressive, and antitumor activities that enable the bacterium to survive in its natural environment. These findings widen the target range of Bacillus spp., in special B. thuringiensis, besides insecticidal activity and help people to better understand its role in soil ecosystem.
