Book excerpt: Fusiform rust, a disease caused by Cronartium quercuum f.sp. fusiforme, is the most economically important disease for loblolly pine (Pinus taeda L.) in the southern U.S. Identification and breeding of loblolly pines that are genetically resistant to fusiform rust are important for successful establishment of commercial plantations. This research developed a new analytical approach to detect major genes for rust resistance using complex segregation analysis in a diallel progeny population. Molecular markers were examined for association with the potential rust resistance genes. Loblolly pines from a six-parent half-diallel mating were planted in a randomized complete block field design at four test sites. Rust infection (gall presence / absence) was recorded annually through age 8. Time trends and genetic differences for rust infection were analyzed based on a polygenic model using a Bayesian logistic approach. For genetic control of rust infection among families, the parental general combining ability (GCA) due to additive effect was much more important than specific combining ability (SCA) due to none-additive effect in full-sib combinations. Large genetic differences among parents and full-sib families were found for rust infection. Among six parents, parent A showed consistent low infection rates over time and across four sites, which indicated high genetic resistance to fusiform rust due to strong polygenic effects and / or major gene effects. A Bayesian analysis of a threshold model was developed and used to make inference about a mixed inheritance model (MIM) that included both polygenic effects and major gene effects. The MIM was compared with a pure polygenic model. Marginalizations were achieved by means of Gibbs sampler. A parent block sampling has been implemented to improve mixing. Results showed that the MIM was a better model to explain the inheritance of rust-resistance than the pure polygenic model in the diallel population. A large major gene vari.

The goal of this study was to advance the understanding and applied use of genetic improvement by analyzing inter- and intra-provenances hybrids’ rust resistance and evaluating midrotation performance of varying levels of genetically improved stock types. The first study compares 16 seedlots at the USDA Resistance Screening Center and evaluates rust resistance of controlled-pollinated inter- and intra-provenances crosses, and openpollinated seedlots from three provenances: Western Gulf, Atlantic Coastal, and Interior Piedmont. Post inoculation, one Coastal OP seedlot was resistant and ten of the seedlots were susceptible to the disease. The second study compares three levels of improved stock types: second-generation open-pollinated, controlled pollinated, and varietal material. After the fifteenth growing season, all three improved stock types were not significantly different from one another in defects, height, diameter, volume, and exhibited site index.

The majority of the ethanol in the US is produced from corn starch. With the US Department of Energy’s target that 30% of the fuel in the US is produced from renewable resources by 2030, the anticipated demand for corn starch will quickly exceed the current production of corn. This, plus the concern that less grain will become available for food and feed purposes, necessitates the use of other feedstocks for the production of ethanol. For the very same reasons, there is increasing research activity and growing interest in many other biomass crops. Genetic Improvement of Bio-Energy Crops focuses on the production of ethanol from lignocellulosic biomass, which includes corn stover, biomass from dedicated annual and perennial energy crops, and trees as well as a number of important biomass crops. The biomass is typically pretreated through thermochemical processing to make it more amenable to hydrolysis with cellulolytic enzymes. The enzymatic hydrolysis yields monomeric sugars that can be fermented to ethanol by micro-organisms. While much emphasis has been placed on the optimization of thermo-chemical pretreatment processes, production of more efficient hydrolytic enzymes, and the development of robust microbial strains, relatively little effort has been dedicated to the improvement of the biomass itself.
