Book excerpt: The endothelium, a monolayer of endothelial cells, constitutes the inner cellular lining of the blood vessels (arteries, veins and capillaries) and the lymphatic system, and therefore is in direct contact with the blood/lymph and the circulating cells. The endothelium is a major player in the control of blood fluidity, platelet aggregation and vascular tone, a major actor in the regulation of immunology, inflammation and angiogenesis, and an important metabolizing and an endocrine organ. Endothelial cells controls vascular tone, and thereby blood flow, by synthesizing and releasing relaxing and contracting factors such as nitric oxide, metabolites of arachidonic acid via the cyclooxygenases, lipoxygenases and cytochrome P450 pathways, various peptides (endothelin, urotensin, CNP, adrenomedullin, etc.), adenosine, purines, reactive oxygen species and so on. Additionally, endothelial ectoenzymes are required steps in the generation of vasoactive hormones such as angiotensin II. An endothelial dysfunction linked to an imbalance in the synthesis and/or the release of these various endothelial factors may explain the initiation of cardiovascular pathologies (from hypertension to atherosclerosis) or their development and perpetuation. P2Y2 receptors increase intracellular calcium, as a result of an activation of the phospholipase C pathway, involving the production of inositol-1,4,5-triphosphate. Diacylglycerol is also produced during this process which activates protein kinase C. In order to investigate further the mechanism of P2Y2 receptor activation and desensitization, four mutants were created within the cytoplasmic tail of the P2Y2 receptor and expressed in 1321N1 human astrocytoma cells that did not previously contain P2Y2 receptors. The cells expressing the altered receptors were then tested in order to determine the mobilization of intracellular free calcium. The concentration response curves of intracellular free calcium increased in all four sets of P2Y2 mutant cell lines, leading to signaling and desensitization similar to that found with the wild type receptor. As a result, these particular locations within the cytoplasmic tail of the P2Y2 receptor are not vital to the activation of the phospholipase C pathway. Further investigation is necessary in order to determine whether other possible locations within the cytoplasmic tail of the P2Y2 receptor play a role in the regulation of the signaling activity.

How do GPCR signalling complexes get assembled to generate such specificity? In order to answer this question, we need to understand how receptors and their signalling partners are synthesized, folded and quality-controlled in order to generate functional proteins. Then, we need to understand how each partner of the signalling complex is selected to join a complex, and what makes this assembly possible. GPCRs are known to be able to function as oligomers, what drives the assembly into oligomers and what will be the effects of such organization on specificity and efficacy of signal transduction. Once the receptor complexes are assembled, they need to reach different locations in the cell; what drives and controls the trafficking of GPCR signalling complexes. Finally, defects in synthesis, maturation or trafficking can alter functionality of GPCRs signalling complexes; how can we manipulate the system to make it function normally again? Pharmacological chaperones may just be part of the answer to this question.
