Available in PDF, EPUB and Kindle. Book excerpt: The elucidation of the cellular and molecular bases underlying the inte grated function of the central nervous system, both in disease and in health, must ultimately come from the combined efforts of scientists from many disciplines, including biology, chemistry, histology, pathology, physiology, pharmacology, and psychology. Communication between scientists from these various disciplines-vital to the advancement of our understanding of the function of the nervous system-has become more and more difficult in recent years. Both increasing specialization and the incredible increases in publications pertinent to brain research in a wide spectrum of journals, in symposium volumes, in monographs, in abstracts, and in reviews contrib ute to the problems of cross-communication and even of communication within a scientific discipline. Research on the significance of cyclic nucleo tides to the function of nervous systems is particularly illustrative of the communication problem. Since the initial publications by Sutherland, Rall, and Butcher in the late fifties and early sixties on high levels of adenylate cyclase, phosphodiesterases, and cyclic AMP in brain, the ensuing litera ture of this field has expanded exponentially. At the present time, from five to ten publications relevant to cyclic nucleotides and the nervous system appear each week. Indeed, these are minimal numbers based mainly on examination of literature titles and key index words. Many articles concerned with some aspect of central function contain, buried within their text, experiments with or related to cyclic nucleotides.

Book excerpt: Prothoracicotropic hormone has been partially purified from whole heads of tobacco hornworm larvae, Manduca sexta. Activity was followed with a larval bioassay. Activity was purified 4400 fold from crude extracts. The hormone was found to be a heat-stable polypeptide with an approximate molecular weight of 25,000 Daltons. The hormone has an isoelectric point in the range pH 5.25-5.50. The most purified preparations were shown to possess activity in vitro with a prothoracic gland culture system. Cyclic AMP and cyclic GMP were quantitated in the central nervous system of Manduca sexta during the pre-metamorphic endocrine activity that leads to a change from feeding to wandering behavior and to the commitment to pupal development. Cyclic AMP and cyclic GMP accumulated in the brain in tandem during this period. Cyclic nucleotide levels remained unchanged in abdominal and thoracic ganglia and in the subesophageal ganglion. Proctolin was quantitated by radioimmunoassay in the central nervous system of the Maderae cockroach, Leucophaea maderae. Proctolin RIA activity was found throughout the CNS, with the sixth abdominal and thoracic ganglia being the richest sources. RIA activity was also found in the cockroach hindgut.

Book excerpt: This book is based on the Symposium "Metabolic Regulation and Functional Activity in the Central Nervous System" which was held on September 16 and 17, 1972, at Saint Vincent (Aosta)/Italy, and was sponsored by the Accademia di Medicina di Torino with the scientific cooperation of the Istituto di Farmacologia, Universita di Torino, and the Pharmakologisches Institut der Freien Universitat Berlin. Its purpose was to give a greater number of scientists from different countries an opportunity to report their latest results under a heading concerning general relationships between metabolism and function. We quite deliberately refrained from pursuing the partly heterogenous subjects into details. Thus, the organizers hoped to be able to interest a greater circle of readers for the manifold subjects from various fields dealing with the investigation of metabolic processes in the central nervous system. The discussion remarks to the lectures could not be considered for printing. The Accademia di Medicina di Torino and all others concerned do not only thank Dr. R. Di Carlo and other members of the Istituto di Farmacologia, Universita di Torino, for the excellent work they performed in preparing this Symposium, but also the Amministrazione Regionale della Valle di Aosta and SIT A V di Saint-Vincent for their generous support and their kind hospitality which made the stay at Saint-Vincent most agreeable for all participants.

Available in PDF, EPUB and Kindle. Book excerpt: PDEs are a family of enzymes that catalyze the hydrolysis of intracellular cyclic nucleotides. They are implicated in a number of disorders and dysfunctions and PDE inhibitors have already proven to be effective therapies for erectile dysfunction, COPD, and psoriatic arthritis. This family of enzymes also plays a role in diseases and disorders of the CNS such as depression, anxiety, schizophrenia, and Alzheimer's Disease. Unfortunately no effective PDE inhibitors have been developed for the treatment of these diseases. The proposed book will be a comprehensive overview of the current state of basic and translational research on PDE inhibitors written by internationally recognized experts. Authors will also discuss potential PDE subtypes and splice variants in the hopes that this will spur more creative approaches to PDE targeting drugs.

Available in PDF, EPUB and Kindle. Book excerpt: Biochemical Correlates of Brain Structure and Function deals with the biochemical correlates of brain structure and function, providing some examples of contemporary work interrelating structure with function of the nervous system. The developing brain provides a system for this kind of study, but broad correlates are also drawn between changing biochemistry and increasing physiological activity. This book is organized into nine chapters and begins with an overview of biochemical, morphological, and functional changes in the developing brain, as well as the underlying molecular basis of nerve differentiation and growth of the developing brain. An account of the concept of the cell cycle and its control is also given. The reader is methodically introduced to the properties of the developing retina and its functional biochemistry, with specific reference to the cyclic nucleotides; the use of selective lesioning to delineate GABA-ergic and cholinergic tracts as well as the catecholamine pathways; and cerebral blood flow alteration in concert with mental activity. The remaining chapters explore regions of the brain with altered glucose utilization in response to changes in local functional activity; the physiologically important factors regulating the supply of oxygen and glucose and the relation of metabolic rate to the metabolic state of the brain; and varying aspects of behavioral neurochemistry. This book is intended for chemists and biologists as well as students of biochemistry.

Available in PDF, EPUB and Kindle. Book excerpt: The original premise of the Editors in initiating this series was that there existed a readership ofneurochemists with considerable biochemical back ground who would make use of a series dedicated to both new develop ments and specialized reviews in neurochemistry. Having selected our authors, we have offered them virtually complete freedom to reflect and speculate in a field in which they have achieved prominence. The response to the first two volumes has been rewarding. The present one continues in this tradition. While we have not attempted to publish specialized volumes, the present volume contains two somewhat related chapters (Chapters 4 and 5, on the role of amino acid neurotransmitters). The first three chapters examine three diverse approaches, each of current interest, in neurochemi cal approaches to the molecular bases of neuronal and glial structure. B. W. Agranoff M. H.
