Book excerpt: It is now apparent that several fields of investigation are converging upon each other as the goal of finding a practical way to prevent cancer is pursued. The present monograph represents the efforts of an interdisciplinary ESO Task Force that was convened to assess interactions of peptide growth factors and steroid-like molecules in the prevention of cancer. Each of the participants in the Task Force represents a differentscientific discipline, and the monogrph is intended to be a synthesis of their different approaches to the common problem of cancer prevention. It is hoped that the reader will find this monograph to be of use in the evaluation of both preclinical and clinical approaches to the major issues that face us in chemoprevention. It is clear that prevention of cancer is an idea whose time has come, and the problem now is to implement the most effective approaches toward achieving the desired goals.

Book excerpt: Recent years have seen a considerable emphasis on growth factors and the elucidation of their mode of function, which has led to the recognition that growth factors, their receptors as well as downstream elements of signalling associated with their function might be potential targets in therapeutic management of human diseases. Humanised monoclonal antibodies raised against growth factor receptors have proved to be valuable for targeted cancer treatment and in patient management. This book reviews the latest developments providing insights into the signalling processes involved in morphogenesis and pathogenesis with emphasis on using the elements of the signalling cascades as targets for therapeutic deployment. Provides a fundamental understanding of the basic functions of growth factors and their receptors, describing how they are linked in biological processes Aids the development of therapeutic treatments for cancer Focuses on the interrelationships and convergence of growth factors and their receptors in development and pathogenesis and encourages greater cooperation and integration in the areas of developmental, cancer and cancer therapeutic research

Book excerpt: This report considers the biological and behavioral mechanisms that may underlie the pathogenicity of tobacco smoke. Many Surgeon General's reports have considered research findings on mechanisms in assessing the biological plausibility of associations observed in epidemiologic studies. Mechanisms of disease are important because they may provide plausibility, which is one of the guideline criteria for assessing evidence on causation. This report specifically reviews the evidence on the potential mechanisms by which smoking causes diseases and considers whether a mechanism is likely to be operative in the production of human disease by tobacco smoke. This evidence is relevant to understanding how smoking causes disease, to identifying those who may be particularly susceptible, and to assessing the potential risks of tobacco products.

Available in PDF, EPUB and Kindle. Book excerpt: This book explores in depth the relation between physical activity and cancer control, including primary prevention, coping with treatments, recovery after treatments, long-term survivorship, secondary prevention, and survival. The first part of the book presents the most recent research on the impact of physical activity in preventing a range of cancers. In the second part, the association between physical activity and cancer survivorship is addressed. The effects of physical activity on supportive care endpoints (e.g., quality of life, fatigue, physical functioning) and disease endpoints (e.g., biomarkers, recurrence, survival) are carefully analyzed. In addition, the determinants of physical activity in cancer survivors are discussed, and behavior change strategies for increasing physical activity in cancer survivors are appraised. The final part of the book is devoted to special topics, including the relation of physical activity to pediatric cancer survivorship and to palliative cancer care.

Available in PDF, EPUB and Kindle. Book excerpt: The fifth of the annual research conferences of the American Institute for Cancer Research was held September l-2, 1994, at the L'Enfant Plaza Hotel in Washington, DC. Appropriately, in view of current directions in research, the theme was "Diet and Cancer: Molecular Mechanisms of Interactions". This proceedings volume contains chapters from the platform presentations and abstracts from the poster session held on the end of the first day. The subtopics for the tl;rree sessions held were "Retinoids, Vitamins A and Din Cancer Prevention and Therapy," "Choline and Lipids: Signal Transduction, Gene Expression and Growth Regulation," and "Dietary Factors and Regulation of Oncogenes, Growth and Differentiation. " A general overview on vitamins A and D emphasized that A and D, in addition to their established roles in vision, reproduction, and bone mineral homeostasis, may play significant roles in regulating cell function. Vitamin A metabolites, trans-retinoic acid and 9-cis-retinoic acid, regulate growth and differentiation. Furthermore, vitamin A deprived animals were more susceptible to both spontaneous and carcinogen-induced tumors. Epidemiological studies showed a correlation between low A intake and higher incidences of certain types of human cancers. Conversely, all-trans retinoic acid is useful in treatment and control of certain types of cancer. Physiologically, Vitamin D is converted to the active form, l ,25-dihydroxyvitamin D (VD). VD regulates hormone production and secretion, myocardial contractility, vascu 3 3 3 lar tone, and growth inhibition and differentiation.

Book excerpt: Cancer will remain a global major health problem unless new diagnostic, prognostic, and management approaches are discovered to address both loss of life and quality of life. Here we summarize the general physiology, pathology, heterogeneity, and evolution of cancer, current status, limitations and challenges associated with prevention, incidence, treatment, survival, and mortality, as well as future directions with regards to solid tumors. Perspectives are provided on how to improve pre-clinical understandings, outcomes, and patient care. Further, this comprehensive, timely overview of the literature has educational value as part of an academic course, seminar, or as a supplementary text.
