Greek anatomists were impressed by the ob servation that the pineal gland is an unpaired structure and they believed that it regulated the flow of thoughts. The philosopher Descartes proposed an important role for this organ in brain function. At the beginning of the 20th century experiments by several investi gators indicated that the pineal influenced sexual function and skin pigmentation and was also responsive to light signals. With the iso lation of melatonin from bovine pineal glands by Lerner and cowork ers in 1958 the modern era of pineal research was initiated. Within a few years the pathway for the biosynthesis of melatonin in the pineal was elucidated. Soon thereafter it was shown that the formation of melatonin was influenced by environmental lighting. Ana tomists found that the pineal was innervated by sympathetic nerves and that the gland had photoreceptor elements. It was also shown that the gonads were influenced by light via the pineal gland. Research on the pineal gland became of increasing interest to anatomists, bioche mists, pharmacologists and endocrinologists. With the expanding know ledge concerning the function of the pineal gland contributed by the wide variety of disciplines, it was thought that a study workshop would be timely.

The detection of new endocrine feed back loops and the availability of new classes of hormonal agents made hormonal intervention with predictable outcome possible. Besides the intellectual challenge of modulating the hormone system, an important aspect of recent research on hormones and cancer is the reduction of treatment-related morbidity achieved with the new hormonal strategies. Thus, controlled intervention in the hypothalamic-gonadotropic axis is increasingly apt to replace surgical removal of the relevant glands, i. e. , the pituitary gland or the gonads. In the same way as, for example, aromatase inhibitors are being used as a substitute for adrenalectomy. The concept that secretion of hypothalamic gonadotropin releasing hormone (GnRH), pituitary gonadotropins, and sex steroids are regulated via negative and positive feedback loops is based on the pioneering work of Hohlweg and Harris some 40 years ago. In 1971, a breakthrough was achieved with the isolation, structural analysis, and synthesis of the luteinizing hormone releasing hormone (LH-RH), or GnRH as it is now more appropriately termed, since it provokes the secretion of both gonadotropins, LH and FSH, and since then the progress made in this area of research has been remarkable. Both ago nists and antagonists of LH-RH have been synthesized and extensively studied in preclinical and clinical settings.

Carbonic anhydrase was first discovered in 1932 by Meldrum and Roughton. Inhibition by sulfanilamide was shown in 1940 by Mann and Keilin. Even Hans Krebs contributed to early studies with a paper in 1948 showing the relationship of 25 different sulfonamides to CA inhibition. It was he who pointed out the importance of both the charged and uncharged character of these compounds for physiological experiments. The field of study that focuses on carbonic anhydrase (CA) has exploded in recent years with the identification of new families and isoforms. The CAs are metalloenzymes which are comprised of 5 structurally different families: the alpha, beta, gamma, and delta, and epsilon classes. The alpha class is found primarily in animals with several isoforms associated with human disease. The beta CAs are expressed primarily in plants and are the most divergent. The gamma CAs are the most ancient. These are structurally related to the beta CAs, but have a mechanism more similar to the alpha CAs. The delta CAs are found in marine algae and diflagellates. The epsilon class is found in prokaryotes in which it is part of the carboxysome shell perhaps supplying RuBisCO with CO2 for carbon fixation. With the excitement surrounding the discovery of disease-related CAs, scientists have redoubled their efforts to better understand structure-function relationships, to design high affinity, isotype-specific inhibitors, and to delineate signaling systems that play regulatory roles over expression and activity. We have designed the book to cover basic information of mechanism, structure, and function of the CA families. The authors included in this book bring to light the newest data with regard to the role of CA in physiology and pathology, across phylums, and in unique environmental niches.

RECOMB 2010 was hosted by INESC-ID and Instituto Superior Tecnico, or- nized by a committee chaired by Arlindo Oliveira and took place at the Int- national Fair of Lisbon Meeting Centre. This year, 36 papers were accepted for presentation out of 176 submissions. The papers presented were selected by the Program Committee (PC) assisted by a number of external reviewers. Each paper was reviewed by three members of the PC, or by external reviewers, and there was an extensive Web-based discussion over a period of two weeks, leading to the ?nal decisions. RECOMB 2010 also introduced a Highlights Track, in which six additional presentations by senior authors were chosen from papers published in 2009. The RECOMB conferenceseriesiscloselyassociatedwiththeJournalofComputational Biology, which traditionally publishes special issues devoted to presenting full versions of selected conference papers.
