Book excerpt: The Platelets: Physiology and Pharmacology is a critical review of the basic platelet systems, methodologic critiques, and platelet involvement in major disease states. This book is composed of 20 chapters that specifically deal with the physiology and biochemistry of the platelet system, along with possibilities for, and consequences of, pharmacologic manipulation. The opening chapters examine the basic processes of platelet aggregation and adhesion. These topics are followed by discussions on the types and functions of platelet a-granule proteins; the aspects of adenine nucleotide levels, regulation, and storage; and receptors for catecholamines and consequences of their agonism. Other chapters are devoted to the metabolism of arachidonic acid, its manipulation, consequences, and metabolite receptors. The discussion then shifts to the second-messenger entities, specifically calmodulin, calcium, and cyclic adenosine monophosphate. A chapter presents the comparisons of man and commonly used laboratory animals concerning platelet behavior variability. Another chapter looks into the use and appropriateness of the use of platelets as models of neurons, with a special emphasis on serotonergic neurons. The remaining chapters contain critical reviews of platelet involvement in diabetes, hypertension and peripheral vascular disease, atherogenesis/atherosclerosis, and ischemic heart disease. These chapters also include specific techniques for platelet analysis, including lumiaggregometry, eicosanoid measurements, and serotonin uptake and release. This book is an invaluable source for pharmacologists, biochemists, researchers, and clinicians who are interested in platelet involvement in disease.

Book excerpt: The Platelet Amine Storage Granule presents a comprehensive examination of the dense granule of human platelets and the development of dense granule precursors in megakaryocytes. Fundamental aspects addressed in the book include a description of the amine-nucleotide storage complex, the process of granule secretion (with focus on inositol phosphate metabolism), modulation of secretion by epinephrine, and a description of platelet storage pool disease. The book's broad coverage of the topic makes it a valuable resource to hematologists, cell biologists, platelet biologists, medical and veterinary geneticists, neurobiologists, endocrinologists, physiologists, and pharmacologists.

Available in PDF, EPUB and Kindle. Book excerpt: Immunopharmacology as a field of scientific endeavor had its origins more than thirty years ago in the application of antibody-based techniques to assays of hormones and drugs in tissues and body fluids. More recently, the field has been redefined to include a primary focus on the immune system as a target of xenobiotic action. Advances in the field of immunology have made it apparent that the immune system, like other organ systems, declines in its function as a result of aging, viral infections like AIDS, and other immunotoxic influences, giving rise to secondary immunodeficiency. Deficiencyof the immune system in turn leads to infections, autoimmune diseases, and an increased incidence of certain cancers. The notion of treating the failing immune system is relatively new; however, more than a decade of research on cancer and AIDS has created the burgeoning new clinical field of immunotherapy. Immunopharmacology then stands as the preclinical and clinical science of immune manipulation. As such, like its parent field of pharmacology, it includes within its scope basic studies of immune mechanisms as they relate to the pathogenesis of inflammation and immunologic disturbances. As with pharma cology, the perspective is always a therapeutic one. Studies of immune and inflammatory processes emphasize the use of pharmacologic probes and drugs to elucidate the underlying biochemical pharmacology.

Available in PDF, EPUB and Kindle. Book excerpt: Advances in Pharmacology and Therapeutics, Volume 4: Prostaglandins-Immunopharmacology contains the proceedings of the 7th International Congress of Pharmacology held in Paris, France, in 1978. The papers explore advances in the understanding of prostaglandins, their immunopharmacology, and their therapeutic applications. Topics covered range from the biochemistry of prostaglandins, thromboxanes, and prostacyclin to the pharmacology of blood platelets and pharmacological approaches to enhancement of immunity. This volume is comprised of 23 chapters and opens with a discussion on the discovery of prostaglandins and their possible biological functions, including platelet aggregation and adrenergic neurotransmission. The reader is then introduced to unstable metabolites of arachidonic acid in relation to thrombosis; pharmacological interference in biotransformation of arachidonic acid; participation of prostaglandins in inflammatory pain; and factors affecting the formation and actions of cyclic AMP in blood platelets. The following chapters explore the role of thymosin and the endocrine thymus in the maintenance of immune balance; effects of thymosin fraction 5 in cancer patients; and the immunoregulatory activities of non-toxic synthetic polyribonucleotides. This book will be of interest to practitioners in biosciences, pharmacology, physiology, and medicine.
