Book excerpt: During the last decade a considerable body of knowledge has come into existence concerning a class of carcinogenic molecules chiefly represented by 4-nitroquinoline i-oxide. Original papers on this subject are numerous and widely scattered over many branches of science; it was felt that these papers should be reviewed and the knowledge brought together in one volume before it became too unwieldy. This we have attempted to do in this monograph. Our aim has been to include all relevant papers published to date, so that it may serve as an epitome of the present status of knowledge on this important subject. We have been fortunate in securing the cooperation of several colleagues who have contributed chapters, each dealing with one aspect of the subject. We have been doubly fortunate in that these contributors, like ourselves, were at one time or another members of the group belonging to the scientific staff of Dr. WARO NAKA HARA, Director of the National Cancer Center Research Institute, Tokyo, Japan.

Available in PDF, EPUB and Kindle. Book excerpt: This series of books attempts to present, in a comprehensive manner, the field of oncology divided into three major areas; etiology, biology, and therapy. These books should serve as landmarks in the rapidly expanding experimental and clinical "universe" of this field. To some, they will be introductory; to others, a summary; for all, critical comments on the futuI'e of research. In l'ecognition of the difficulties inherent in attempting to pause and reflect while experimental data emerge with ever-increasing rapidity, the presentations take the form of overviews rather than reviews. Where possible, an historical perspective on observations and experimentation which led to our present understanding is presented, the state of the art in technique and approach is reviewed, and the gaps in knowledge and in technique are indicated. The aim throughout is integration-using the findings from one approach for comparison with others. The tremendous expansion of interest in oncology as a medical-biological discipline stimulated the publication of these volumes. This expansion, well warranted in terms of the impact of oncology on human morbidity, has been characterized by at least three phenomena. First, there has been an enormous increase in money and manpower devoted to the investigation and treatment of malignancy. That the research has become more and more "directed" or program-oriented signals the interest of those beyond the scientific community in the management of the eHort. Second, increasing numbers of students are entering the field of oncology as their major training program.

Available in PDF, EPUB and Kindle. Book excerpt: Metabolic Basis of Detoxication: Metabolism of Functional Groups considers the possible fates of the relatively circumscribed number of functional groups that xenobiotics bear. An understanding of the possible reactions, and the chemical and biological factors influencing them, will contribute to the overall predictability of the fate of "real" molecules. This approach attempts to knit together the understanding of metabolic pathways with that of the enzymes that catalyze the specific steps. The book contains 18 chapters and begins with a discussion of the biological oxidation of carbon atoms. This is followed by separate chapters on the metabolism of halogenated aliphatic hydrocarbons, aryl halides, heterocyclic rings, alcohols, aldehydes, and ketones. Subsequent chapters cover oxidative processes such as metabolic dealkylations and biological oxidation at nitrogen centers; the reduction of nitro and azo compounds and tertiary amine N-oxides; the oxidation, alkylation, acylation, and glycosylation of mercaptans; epoxide metabolism; and conjugation of phenols. The book aims to inform and interest the pharmacologist and toxicologist concerning the biochemical aspects and to orient the biochemist to the pharmacological insights required in dealing with the metabolism of xenobiotics.

Available in PDF, EPUB and Kindle. Book excerpt: Free Radicals in Biology, Volume V covers the mechanisms for the generation of free radicals. This volume contains eight chapters that discuss the biology and chemistry of oxy-radicals in mitochondria and the radical-mediated metabolism of xenobiotics. The opening chapter describes the mechanisms of free radical production in enzymatically promoted lipid peroxidation, generally in microsomes or microsomal lipids. The subsequent chapters explore the biochemistry and biology of plant and animal lipoxygenases; the production of superoxide and hydrogen peroxide in mitochondria; and the biological role of these species in mitochondria and related systems. The discussions then shift to the effects of superoxide production in white blood cells, with an emphasis on an evaluation of the oxygen-dependent reactions of the important phagocytic cells, the monocytes, and the polymorphonuclear leukocytes. This volume further covers the formation and the role of oxy-radicals in the red blood cell, which is a very useful system for studying the protection of biological tissue against radical-mediated damage. A chapter presents a comprehensive review of the production of free radicals during the metabolism of xenobiotics. The last chapters provide an overview of the enzymology, biological functions, and free radical chemistry of glutathione peroxidase. These chapters also examine a number of gerontological principles and the effect of antioxidants in aging. Chemists, biologists, and physicists will find this book of great value.

Book excerpt: The Role of Chromosomes in Cancer Biology provides a description of the molecular organization and function of chromosomes and the consequences of chromosomal aberrations in human development. The book presents accounts on the structure and function of the chromosome; the cellular features of primary tumors and ascetic fluid; the cytological actions of radiation and drugs and their relevance to therapy. Developmental disorders caused by chromosomal anomalies; chromosome aneuploidy in human malignancies; and viral oncogenesis are discussed as well. The book will prove to be very insightful to those involved in cancer research, oncologists, cytologists, and molecular biologists.

Book excerpt: Scientists are coming under increasing pressure from activist groups to stop animal experimentation, branded as cruel and unnecessary for improving human health. This attitude, however, stems from an unrealistic evaluation of the situation and distorted information about present scientific knowledge. There is no question that most medical progress - perhaps all, in fac- has been attained through knowledge derived initially from experiments in various animal species. There is practically no way of replacing animals in these investigations and so-called 'alternative methods' are in reality merely complementary. Tissue cultures, cell, microorganisms, enzymes, membranes, mathematical models - all are useful for preliminary screening tests and for testing hypotheses, but the complexity of a living organism is such that in vivo studies are essential before any test can responsibly be made in man. This book presents the proceedings of an international symposium organized in Strasbourg (October 24-25, 1988), with the aim of assessing present-day requirements as regards animal experimentation in research related to major medical and toxicological problems still awaiting solutions.
