Available in PDF, EPUB and Kindle. Book excerpt: This book is about selectively toxic agents. That is to say, it is about those substances that affect certain cells without harming others, even when they are close neighbours. Toxicity need not be fatal. It can be made easily reversible, as is the case with general anaesthetics. Selective toxicity covers an immense field: most of the drugs used for treating illness in man and his economic animals, as well as all of the fungicides, insecticides, and weed killers that are used in agriculture. Essentially, this book is a discussion of the physical and chemical means which contribute to selectivity, and this is the basis of molecular pharmacology. _Selective Toxicity began as a course of lectures that Professor F. G. Young encouraged me to give in University College London, in 1948 and again in 1949. The first edition appeared in 1951, as a very small book because little was then known about the factors that provide selectivity. Since those early days, the subject has undergone tremendous development. At first, industry was un receptive to the word 'toxicity', however qualified! Yet the market was being supplied with biologically powerful substances of which several had the potential to cause harm. This aspect was brought to light by two events of the early 1960s. The first of these was the discovery that a sedative, thalidomide, administered to expectant mothers, after what was then considered to be adequate testing, had caused permanent deformities in about 10000 children.

Available in PDF, EPUB and Kindle. Book excerpt: Toxic Phosphorus Esters: Chemistry, Metabolism, and Biological Effects aims to become a source book on organophosphate research and show how the understanding of the events following organophosphate poisoning of animals can be understood in terms of events at the molecular level. The manuscript first offers information on nonenzymic reactions and the reaction with cholinesterase in vitro. Discussions focus on hydrolysis, isomerization, oxidation, phosphorylation of the enzyme, and selectivity for different cholinesterases. The text then examines enzymic degradation and activation in vitro, effects in mammals, and effects on isolated whole tissues. The publication ponders on the effects in insects and plants, including results of poisoning, metabolism, resistance, antagonism and synergism, and phytotoxicity. The text also reviews selective toxicity, as well as excretion and storage, metabolism, design of new selective compounds, and nature of the target. The manuscript is a dependable reference for readers interested in the composition, reactions, and effects of toxic phosphorus esters.

Book excerpt: Antimicrobial agents are essential for the treatment of life-threatening infections and for managing the burden of minor infections in the community. In addition, they play a key role in organ and bone marrow transplantation, cancer chemotherapy, artificial joint and heart valve surgery. Unlike other classes of medicines, they are vulnerable to resistance from mutations in target microorganisms, and their adverse effects may extend to other patients (increased risk of cross-infection). As a consequence, there is a constant requirement for new agents, as well as practices that ensure the continued effective prescribing of licensed agents. Public awareness and concerns about drug resistant organisms has led to widespread publicity and political action in the UK, Europe and worldwide. The control of drug resistance and the implementation of good prescribing practice are now legal requirements in the UK as a result of the UK Health Act (2008). These fundamental changes underscore the need for a thorough understanding of the advantages and risks associated with specific antibiotic choices. This sixth edition of Antimicrobial Chemotherapy continues to be a valuable resource for undergraduates and graduates requiring a thorough grounding in the scientific basis and clinical application of these drugs. This new edition is updated to include the most recently licensed agents, notably in the treatment of viral infections including HIV/AIDS, and contains new guidance on prescribing practice and infection control practices that limit the development and spread of resistant organisms.

The scope of this book includes a discussion of the major types of chemicals involved, their general properties and detrimental biologic effects, the methods used to demonstrate these effects, the basis for clinical diagnosis, and therapy for the harmful effects of chemicals on humans. Individual examples are used to demonstrate the principle discussed. Available in PDF, EPUB and Kindle. Book excerpt: A thorough understanding of cellular and molecular mechanisms involved in the individual expression of toxic effects provides an important tool for assessment of human health risk. New aspects, major advances, and new areas in molecular and cellular biology and toxicology demand updated sources of information to elucidate the functional mechanics of human toxicology. Mechanistic Toxicology: The Molecular Basis of How Chemicals Disrupt Biological Targets, Second Edition retains the accessible format of the original to present the general principles that link xenobiotic-induced toxicity with the molecular pathways that underlie these toxic effects. Extensively illustrated, this book forms a conceptual bridge between multiple events at the molecular level and the determinants of toxicity at the physiological and cellular level. Specific examples of drugs, environmental pollutants, and other chemicals are carefully chosen to illustrate and highlight the fundamental mechanisms of toxicity at different toxicokinetic and toxicodynamic levels. The book includes references and review articles at the end of each chapter, as well as boxed text for relevant review information on biological, biochemical, molecular, and toxicological background. Linking molecular pathways to more general biomedical contexts, the author ensures that the reader is not lost in the details and instead receives a broad understanding of the processes underlying xenobiotic toxicity. New in the Second Edition Updated chapters Types of toxic responses Disruption of signal transduction by xenobiotics Disruption of mitochondrial function Novel mechanisms derived from systems toxicology
