Available in PDF, EPUB and Kindle. Book excerpt: Over the last decade the volume Membrane Fusion. edited by Poste and Nicholson, has probably served as one of the major sources of review in formation on fusion in membrane systems. Since its publication much new information has been collected. New methods of inducing fusion have been invented or discovered, and new applications for fusion have been found. The need for an up-to-date monograph that covers and in tegrates these subjects, reviews established material, and rationalizes and integrates the old and the new is thus obvious. This book is the product of efforts to meet this need. Most of the current work in the field of membrane fusion takes place within the context of intact or modified cells. Hence this book emphasizes the plasma membrane. Each chapter is either a review, a report, or a short historical overview, depending, respectively, on whether the subject is large in scope and has a long history, or the subject is in such an early stage of development that most of what is known is still in the hands of a relatively small number of investigators and is best covered in report form.

Book excerpt: Although cell fusion is an omnipresent process in life, to date considerably less is still known about the mechanisms and the molecules being involved in this biological phenomenon in higher organisms. In Cell Fusion in Health and Disease Vol 1 & Vol 2 leading experts will present up-to-date overviews about cell fusion in physiological and patho-physiological processes, which further covers the current knowledge about cell fusion-mediating molecules. Volume 1 deals with Cell Fusion in Health and will cover aspects of cell fusion in fertilization, placentation, in C. elegans, in skeletal muscle development and tissue repair, and the use of cell fusion for cellular reprogramming and cancer vaccine development. Volume 2 focuses on Cell Fusion in Disease with a particular emphasis on the role of cell fusion in cancer development and progression. Thus, Cell Fusion in Health and Disease Vol 1 & Vol 2 represents a state-of-the-art work for researchers, physicians or professionals being interested in the biological phenomenon of cell fusion and beyond.

Available in PDF, EPUB and Kindle. Book excerpt: The American Anti-Vivisection Society (AAVS) petitioned the National Institutes of Health (NIH) on April 23, 1997, to prohibit the use of animals in the production of mAb. On September 18, 1997, NIH declined to prohibit the use of mice in mAb production, stating that "the ascites method of mAb production is scientifically appropriate for some research projects and cannot be replaced." On March 26, 1998, AAVS submitted a second petition, stating that "NIH failed to provide valid scientific reasons for not supporting a proposed ban." The office of the NIH director asked the National Research Council to conduct a study of methods of producing mAb. In response to that request, the Research Council appointed the Committee on Methods of Producing Monoclonal Antibodies, to act on behalf of the Institute for Laboratory Animal Research of the Commission on Life Sciences, to conduct the study. The 11 expert members of the committee had extensive experience in biomedical research, laboratory animal medicine, animal welfare, pain research, and patient advocacy (Appendix B). The committee was asked to determine whether there was a scientific necessity for the mouse ascites method; if so, whether the method caused pain or distress; and, if so, what could be done to minimize the pain or distress. The committee was also asked to comment on available in vitro methods; to suggest what acceptable scientific rationale, if any, there was for using the mouse ascites method; and to identify regulatory requirements for the continued use of the mouse ascites method. The committee held an open data-gathering meeting during which its members summarized data bearing on those questions. A 1-day workshop (Appendix A) was attended by 34 participants, 14 of whom made formal presentations. A second meeting was held to finalize the report. The present report was written on the basis of information in the literature and information presented at the meeting and the workshop.

The book presents the basic biological mechanisms of how cloning works and progresses to discuss current and potential applications in basic biology, agriculture, biotechnology, and medicine. Beginning with the history and theory behind cloning, the book goes on to examine methods of micromanipulation, nuclear transfer, genetic modification, and pregnancy and neonatal care of cloned animals. The cloning of various species—including mice, sheep, cattle, and non-mammals—is considered as well. The Editors have been involved in a number of breakthroughs using cloning technique, including the first demonstration that cloning works in differentiated cells done by the Recipient of the 2012 Nobel Prize for Physiology or Medicine – Dr John Gurdon; the cloning of the first mammal from a somatic cell – Drs Keith Campbell and Ian Wilmut; the demonstration that cloning can reset the biological clock - Drs Michael West and Robert Lanza; the demonstration that a terminally differentiated cell can give rise to a whole new individual – Dr Rudolf Jaenisch and the cloning of the first transgenic bovine from a differentiated cell – Dr Jose Cibelli. The majority of the contributing authors are the principal investigators on each of the animal species cloned to date and are expertly qualified to present the state-of-the-art information in their respective areas. Additionally, cell fusions participate in tissue repair and may be important to cancer development, progression and therapy. A large number of factors regulate cell fusions, including receptors and ligands, membrane domain organizing proteins, proteases, signaling molecules and fusogenic proteins that bring membranes close together. In the present volume, we interrogate what mechanisms that may be important to cell fusions in mammals and what mechanisms that may be shared with lower organisms and viruses. Additionally, the importance of cell fusions to the pathogenesis as well as treatment of diseases is addressed.

Book excerpt: In order to study the genetics of somatic animal cells, Henry Harris and his colleagues invented the powerful method of viral-induced animal cell fusion. This volume evaluates the impact of cell hybridization on the study of cell differentiation, gene mapping, gene regulation, and the development of monoclonal antibodies. The book details important studies on nuclear structure and function, intracellular transport, membrane protein mobility, and nuclear-cytoplasmic interactions in heterokaryons and other cells. Tumor-suppressor genes and the role of radiation-hybrid mapping in the analysis of genetic tumors are also discussed. The book concludes with a chapter by Henry Harris which addresses a number of unsolved questions that remain regarding the link between cell growth and differentiation. Students and researchers in molecular genetics and cell, molecular, and cancer biology will welcome this fascinating overview of cell fusion research.
