Recent work has provided new isolates from different geographical locations. Prior to detailed biochemical investigations, laboratory studies were conducted on ten different isolates representing geographical locations around the coast of Florida. The investigation of the physiological parameters for the ten different isolates provided a foundation for the proper selection of a single isolate that would closely represent the species in the field. Each isolate was capable of a range of responses with primary dependence on cell counts within the cultures. Evidence suggests that either carbon limitation or bacterial negative feedback caused changes in the physiological parameters as cell counts increased. The Apalachicola isolate was selected for further biochemical investigations because it was representative of the group response. A nutrient-replete intermediate-light mesocosm experiment investigated the taxis patterns, and the biochemical composition of vertically migrating K. brevis populations. This experiment confirmed the internal biochemical status of the cell controlled migratory behavior, growth and reproduction. Young cells fixed carbon at the surface while older cells limited vertical migration toward the middle depth in anticipation of cellular division. The cells were capable of performing complex lipid control during vertical migration. Quick turnover of these lipids indicated K. brevis was more than capable of taking advantage of changing physical conditions. Finally, nutrient replete and deplete mesocosms investigated how K. brevis responded to field-simulated high light conditions. Results from high light and nitrogen-limited exposure suggested K. brevis may utilize the benthos by minimizing exposure to oxidative stress and as a nitrogen source supplied from benthic pore-water flow in permeable sediments. High light also caused K.

Book excerpt: Coral cover is in decline on a global scale, with increased mortality events being attributed to a number of global and regional stressors. While the impacts of global stressors (e.g. sea surface temperature rise, ocean acidification) are well documented, there is growing interest in identifying and understanding the impacts of regional stressors. The reason for this change in focus is that regional stressors can often work in combination, sometimes synergistically, with global stressors and that stressors on a regional scale tend to be more easily mitigated by management practices. One regional stressor that impacts a myriad of marine organisms in the southeastern United States is the annual red tide blooms produced by the dinoflagellate, Karenia brevis. Their impacts, along with the lipid soluble polyether neurotoxins they produce, termed brevetoxins, are well studied in economically important organisms, such as bivalves. However, little is known of their impacts on organisms that possess ecological importance, such as species of scleractinian coral. To address this gap in knowledge, a multifaceted study is discussed herein, which evaluated the effects of ecologically relevant concentrations of K. brevis and associated brevetoxins on different coral life history stages and coral species. The second chapter addresses the impacts of red tide on larval behavior, settlement and survival of the coral species Porites astreoides, as well as impacts of photochemical efficiency and oxidative stress within different coral species (P. astreoides larvae, P. astreoides adults, Acropora cervicornis, Cladocora arbuscula, and Phyllangia americana). The third chapter confers the use of broad-scale proteomic analysis to identify the cellular response of the non-model coral species, P. astreoides, following exposure to red tide. Coral larvae actively avoided both medium and high bloom conditions of K. brevis and brevetoxins, while percent larval settlement and survival were impacted following exposure to high bloom concentrations of K. brevis. Photochemical efficiency of in hospite Symbiodinium was reduced following exposure to both K. brevis and brevetoxin in P. astreoides larvae, as well as exposure to K. brevis in P. astreoides adults, while being unimpacted in A. cervicornis. Compared to controls, high bloom conditions resulted in an increase in biomarkers of lipid peroxidation in C. arbuscula. This was also seen in P. astreoides larvae at 24 hours; however, this difference was indistinguishable following 48 hours. Surprisingly, no other biomarker of oxidative stress analyzed were impacted. Broad-scale proteomic analysis of P. astreoides following exposure to red tide conditions revealed variable changes in proteome expression depending on if the corals were exposed to K. brevis or brevetoxins. Exposure to brevetoxins resulted in differential expression of proteins related to DNA organization, chromatin formation and transcription expression; while exposure to K. brevis resulted in differential expression of proteins related to redox homeostasis, protein folding, energy metabolism, and production of reactive oxygen species. The results of this study demonstrate the potential for annual red tide blooms to act as a regional stressor on coral species. They highlight the ability of red tide conditions to negatively impact coral at multiple life history stages and that the extent of these effects may be species specific. They also provide further incite of coral's response to red tide exposure at the cellular level.

Book excerpt: The dinoflagellate Karenia brevis causes harmful algal blooms, called red tides, that increasingly disrupt ecosystems and economic activity in the western coast of Florida. Strategizing for a future of imminent red tide demands interventive control options for severe blooms. Bio-inspired chemical control attempts to synergistically merge the concepts of chemical control and biotic control, incorporating the logistical advantages of the former and the environmental consciousness of the latter. Amine compounds identified in a dinoflagellate-specific bacterial algicide were assessed for their potential to control the abundance of K. brevis. First, dose-response of K. brevis to four selected compounds was measured. Three of those, ammonia, putrescine, and spermidine, controlled K. brevis and had lesser effects on a nontarget algal species. To improve dose efficiency, combinations of these compounds were screened for synergistic interaction. Three combined amine doses potentiated by synergisms proceeded to testing on natural microbial communities collected from a field site subject to red tide. The impact of these doses on nontarget microbes was compared. 0.9 ppm ammonium with 7.5 ppm putrescine, hereafter "NP", emerged as preferable with respect to effects on photosynthetic biomass and photophysiological stress. Next, NP was applied to a naturally-occurring red tide where it controlled K. brevis and restructured microbial communities. Dynamics in microbial communities and water chemistry parameters were then monitored over 7 days in NP-treated. NP demonstrated control of non-K. brevis dinoflagellates in non-red tide communities, with specificity over diatoms and other phytoplankton groups. Effects of NP on pH, turbidity, and dissolved oxygen of mesocosms revealed pertinent considerations for future field application. Putrescine had short residence times whereas ammonia accumulated to potentially toxic levels. Ultimately, NP, formulated from commercially-available and relatively inexpensive amine compounds produced by naturally-occurring algicidal bacteria, exhibited potent, taxonomically-specific control of K. brevis in complex natural microbial communities. Thus, efforts to replicate this result and further engineer NP for eventual field deployment are supported.

The blooms have adverse effects on humans and marine animals. Karenia brevis produces toxins that, in humans, can cause upper and lower respiratory problems as well as nausea and vomiting. For marine mammals, birds, fish, and other marine organisms, exposure to brevetoxins can result in morbidity and even death. Even with the technology and resources we have today, scientist do not fully understand the mechanisms behind toxin production and bloom initiation. Because cellular processes may play a major role in Karenia brevis blooms, attempts have been made to understand K. brevis at the molecular level, but studies are still ongoing due to lack of understanding gene organization and expression. Thus, our goal was to help guide the reconstruction or sequencing of the genome. The purpose of my research was to find if there are multiple copies of certain gene sequences, and what variability exists between and within the genes. I attempted determine the copy number of a suspected multi-copy gene and if any sequence variability existed within the gene and the intergenic regions. Focus was placed on the gene for proliferating cell nuclear antigen (PCNA) whose known function is to aid in DNA replication and repair, because the PCNA is present in multiple copies and (Zhang et al. 2006) and the EST collection of K. brevis contained several PCNA single nucleotide polymorphisms (Lidie et al, 2005; McLean, unpublished data).--P. 1.

Book excerpt: Diel vertical migration (DVM) can offer motile phytoplankton a competitive advantage over other phytoplankton species. DVM has been well documented in the harmful dinoflagellate, Karenia brevis; however, the extent to which changes in nutrient regimes and density stratification modify migration patterns has not been thoroughly investigated. A 2 m experimental column was constructed with sensors and sampling ports at four depths to obtain fluorescence and temperature measurements and discrete water samples. The environmental parameters of the experimental column represented temperature and nutrient conditions found on the West Florida Shelf (WFS), where Karenia blooms frequently occur. Results demonstrated a clear DVM pattern for K. brevis where surface aggregations occurred during light periods, and bottom aggregations occurred in dark periods. However, the rate and intensity of migration varied between experimental conditions. Injections of nutrient replete water at the surface and bottom did not appear to modify migratory behavior, in that a portion of the population resided at depth regardless of light or dark periods. Weak density stratification caused a delay in downward migration in dark periods, whereas strong density stratification caused a complete cessation of downward migration. In a weakly stratified environment with a bottom injected replete water mass, DVM patterns most closely replicate the pattern under weakly stratified conditions alone. Modifications in the natural DVM pattern of K. brevis with respect to varying environmental and nutrient conditions that occur on the WFS can have major implications current forecast model predictions, and for monitoring and mitigation strategies.
