Book excerpt: Cell Biology of the Major Histocompatibility Complex documents the proceedings of a symposium on ""Cell Biology of the Major Histocompatibility Complex"" held at Arden House on the Harriman Campus of Columbia University from June 8 -10, 1984. The meeting was the ninth of the P & S Biomedical Sciences Symposia. The book is organized into five parts. Part I on the structure of MHC molecules includes papers on human histocompatibility antigens; the cloning of human MHC; and organization of the genes of the H-2 complex. Part II on alternate forms of MHC molecules includes studies on the expression of a secreted form of the MHC class I antigen and alternative splicing in the H-2 multigene family. Part III deals with MHC biosynthesis, intracellular transport, and membrane expression. It includes studies on the manipulation of glycans on antigens of the MHC and dynamics of MHC molecules in lymphoid cells. Part IV on homologues of class I MHC molecules covers topics such as Tla expression in normal and malignant cells and the Qa series of antigens. Part V on the association of MHC with antigens includes studies such as histocompatibility molecules as immune response gene products and molecular nature of T-cell recognition of antigen.

Book excerpt: From molecules to populations and back In biology, the most vigorous organisms often ensue from a union of two disparate, pure lines. In science, too, laws of hybrid vigor seem to operate at the interface between two disciplines, an interface that often proves to be fertile ground for germinating concepts and new outlooks. The fringes of research into the major histocompatibility complex (Mhc) have provided such an interface several times in the past and the encounters have invigorated fields such as transplantation biology, cellular immunology, and immunogenetics. In the last few years, a new interface has been emerging between Mhc and evolutionary genetics, and particularly the branch of evolutionary genetics dealing with molecular evolution. Mhc research relies upon molecular evolutionary genetics, with its grand superstructure of mathematical formulations, to come to grips with the events leading to and maintaining the Mhc polymorphism. Without the armament of rigorous statistical procedures developed by evolutionary geneticists, the intricate relationships among Mhc genes cannot be resolved. It will undoubtedly be a molecular geneticist who is the final arbiter in the dispute concerning the nature of the selection pressure molding the Mhc genes. And it is doubtful whether the true function of Mhc can ever be comprehended without the vantage point afforded by the elucidation of its evolutionary history.

Available in PDF, EPUB and Kindle. Book excerpt: Every biological system is the outcome of evolution and has a history all its own. This history dictates how the system works and why it has certain properties and not others. This is why we need to study not only the structure and function, but also the history of the system. This argument undoubtedly applies to the study of the immune system and also to the study of the major histocompatibility complex (MHC). Since 1989, researchers of various scientific disciplines who share a deep inter est in MHC evolution have held a meeting every two years to discuss their latest research developments, exchange ideas, and foster friendship. Together with my colleagues Drs. Naoyuki Takahata and Yoko Satta, I organized the Sixth Interna tional Workshop on MHC Evolution in Hayama, Japan, May 25-29, 1999. This volume is the proceedings of that conference. It covers diverse topics pertinent to MHC evolution, including the origin of the adaptive immune system, the organi zation of the MHC in humans and other model vertebrates, MHC-parasite co evolution, and the nature and origin of MHC polymorphism. I hope that this book will be of interest not only for MHC researchers and immunologists, but also for other specialists who are interested in the evolution of biological systems in gen eral.

Book excerpt: Antigen processing and presentation, as a field, explores a broad range of protein interactions and functions, both intracellular (in the cytoplasm and in the endoplasmic reticulum) and at the cell surface (between T cells and MHC molecules). To investigate such a diverse array, it is necessary that biochemical, cell biology, and immunological techniques all be employed. The purpose of Antigen Processing and Presentation Protocols is therefore to detail the most up-to-date techniques being used in this burgeoning field. Such techniques include those used to question how MHC-binding peptides are generated, to test how peptides are delivered to MHC molecules, to analyze MHC peptide-binding patterns, and to assay the T-cell response to MHC/peptide. Antigen Processing and Presentation Protocols should aid both those new and those experienced in this area of research in extending the questions that can be asked and answered by the application of these current methods. For editorial assistance, I would like to thank Angela Beninga and Rachael Turnquist.

Available in PDF, EPUB and Kindle. Book excerpt: The major histocompatibility complex (MHC) is a highly polymorphic and diverse multigene locus in all jawed vertebrate species that has an integral role in adaptive/innate immune systems, transplantation, and infectious and autoimmune diseases. The MHC supra-locus in mammalian vertebrates is usually partitioned into three distinct regions, known as classes I, II, and III, which, to varying extents, can be found conserved in nonmammalian jawed vertebrates, such as bony fish, amphibians, and bird lineages. The MHC gene region is characterized particularly by the expression of class I and class II glycoproteins that bind peptides derived from intracellular or extracellular antigens to circulating T-cells. While this expressed antigenic specificity remains the predominant interest with respect to MHC function and polymorphism in a population, a broader concept has emerged that examines the MHC as a multifunctional polymorphic controller that facilitates and regulates genome diversity with a much greater array of functions and effects than just MHC-restricted antigen recognition. This volume of 19 reprints presented by various experts and collected from the Special Issue of Cells on “MHC in Health and Disease” covers a broad range of topics on the genomic diversity of the MHC regulatory system in various vertebrate species, including MHC class I, II, and III genes; innate and adaptive immunity; neurology; transplantation; haplotypes; infectious and autoimmune diseases; fecundity; conservation; allelic lineages; and evolution. Taken together, these articles demonstrate the immense complexity and diversity of the MHC structure and function within and between different vertebrate species.

Available in PDF, EPUB and Kindle. Book excerpt: This volume documents our growing understanding of the human major histocompatibility complex. The application of this information is ever more important as the limits of transplantation continue to be reduced, including the recent success of bone marrow transplantation between unrelated but closely matched individuals. In addition, the need to transfuse platelets in the face of immunologic barriers continues to challenge transfusion services. Thus, the serologic information summarized in this volume is essential for optimal patient care. At the same time, recombinant DNA technology has led to a revolution in our understanding of many aspects of basic biology. Among the advances has been the initial characterization of the structure of some HLA loci. While this will ultimately improve clinical services, constant reference to serologic data is essential so that the powerful new techniques can be applied in the most effective ways. The timing of the First Red Cross International Histocompatibility Workshop is fortunate as it brings together experts from around the world to address the state of the art. We are all grateful to Dr. John Lee and his colleagues for organizing the workshop, and for bringing together in this volume the material to be presented in Beijing during October 17-23, 1990. Book excerpt: The Role of Products of the Histocompatibility Gene Complex in Immune Responses documents the proceedings of a conference held on 3-7 November 1975, which brought together an international group of scientists spanning three independent disciplines—genetics and immunogenetics, molecular biochemistry, and immunobiology—with clinical medicine overlapping these disciplines. This volume contains 42 papers organized according to the eight sessions held at the conference. The papers in Session I examined the genetics of the major histocompatibility complex. Session II presented studies on the biology of mixed lymphocyte interactions and cell-mediated cytotoxicity reactions. Session III discussed immune response gene systems while Session IV dealt with the genetic control of cell interactions. The papers in Session V covered idiotypic determinants on T cell receptors. Session VI investigated the properties of histocompatibility gene products involved in regulation of immune responses. Session VII focused on the biochemistry and immunocytology of cell surface products of the major histocompatibility complex. Finally, Session VIII discussed interrelationships between products of the major histocompatibility complex and their relevance to disease.
