Book excerpt: Throughout the more than 20 years that have followed the beginnings of capillary electrophoresis (CE), its application to the analysis of proteins and peptides has continued to be reliable, versatile, and productive. Over time, CE has matured to become a superb complement to HLPC, and in many cases has also evolved as an automated and quantitative replacement for conventional slab gel electrophoresis methods such as SDS-PAGE and isoelectric focusing. Within Capillary Electrophoresis of Proteins and Peptides, we have assembled contributions from researchers who are applying state-of-the-art CE for protein and peptide analysis, including topics that we believe are of great potential both in the present and for the future. In comparison to traditional separation methods, CE represents a miniaturized analysis technique (especially in its microchip-based format) that is highly dependent upon the basic fundamentals of effective sample recovery and high sensitivity detection. With these issues in mind, Chapters 1–4 describe recently developed approaches for both capillary coatings and analyte detection via laser-induced fluorescence. Since the discipline of biotechnology has established itself as a primary platform for the application of CE to the analysis of proteins and peptides, Chapters 5–7 demonstrate a variety of examples of the specific techniques that have been applied for the development of biopharmaceuticals and their commercialization. The methods covered here include also the analysis of oligosaccharides from glycoproteins.

Available in PDF, EPUB and Kindle. Book excerpt: The purpose of the preface is to explain the book's objectives and how to use it; give warnings, disclaimers, and the like.* The main objective of Protein and Peptide Analysis by Mass Spec trometry is quite straightforward—to present authoritative, up-to-date, and practical accounts of the use of mass spectrometry in the analysis of pep tides and proteins. How to use it? Every reader will have their own particular interests and will surely be drawn toward the chapters that cover these interests. Within the remaining chapters, however, techniques are described with analytical possibilities that such a reader can then only guess at. So, read the book fully. Again, as is customary in the Methods in Molecular Biology series, the chapter format (Introduction, Materials, Methods, and Notes) allows the authors to introduce the techniques, to explain their relevance and applicability, and, above all, to provide detail—detail that represents each author's accumulated experience and enables the reader to use and benefit from these methods. So, read the book fully, and read it diligently. Warnings and disclaimers: Mass spectrometry today offers the pro tein chemist ready access to a wealth of information that is otherwise avail able only with great difficulty, or perhaps not at all. With this goal in sight, any warnings and disclaimers will almost surely be ignored. So, a warning anyway; the use of mass spectrometry might be habit forming.

Karger AG (Switzerland). This book was released on 2005 with total page 228 pages. Available in PDF, EPUB and Kindle. Book excerpt: With the latest advances in analytical technologies, most biological products can now be extensively characterized in terms of their identity, heterogeneity and impurity profile. The currently available analytical methods (both physicochemical and biological) can characterize the primary, secondary and to some extent, the higher order structure of proteins. The sensitivity and selectivity of these methods allow for the identification and characterization not only of the desired protein component, but also many product-related substances and impurities as well as process-related impurities present in the drug preparations. Using an appropriate selection of analytical tools, it may be possible to demonstrate physicochemical and functional comparability between protein products manufactured before and after a manufacturing change or during product development. However, the extent of demonstrable comparability depends on the tests used, on the nature of the product and on the basic understanding of structure-function relationships. Assessment of immunogenicity is also a critical component of product characterization and is highly dependent on the types of assays used.

Book excerpt: Reflecting the versatility of the author’s science and the depth of his experience, Application of Solution Protein Chemistry to Biotechnology explores key contributions that protein scientists can make in the development of products that are both important and commercially viable, and provides them with tools and information required for successful participation. One of the of the world’s most respected protein researchers, Roger Lundblad does not succumb to the notion that new is always better. The application of protein science to the practice of commercial biotechnology is traced to the underlying basic solution protein chemistry. It is only by achieving this understanding that the full potential of protein science may be obtained in the development and characterization of the diverse products of modern biotechnology. Dr. Lundblad also goes far beyond the biopharmaceutical applications that are often equated with protein science today to demonstrate the field’s unique versatility. From the making of bread and the invention of adhesives to the production of pharmaceuticals and the development of recombinant DNA products— in each of these products, the role of the protein chemist remains prominent. The important point is that classical protein chemistry is a critical part of the practice of biotechnology in the marketplace. Providing the direction and the foundational work needed by students as well as the details and hundreds of references needed by designers and developers, this remarkable work— Delves into the application of protein science for producing products as diverse as adhesives, drug delivery systems, and quality food products Explores chemistry of attachment of proteins and peptides to solid surfaces with regard to applications both for the improvement of steel and titanium and in DNA and protein microarrays Describes the development of bioconjugates used in antibodies Offers essential advice on guidelines required for producing licensed biopharmaceutical products While he does include a great deal of material not found in other sources, Dr. Lundblad makes a point to separate what is truly new from that which has merely been renamed. A reference unlike most, scientists and students eager to learn will find a text that is as practical as it is purposeful.

This book was released on 2010-08-28 with total page 237 pages. Available in PDF, EPUB and Kindle. Book excerpt: Protein hydrolysates, otherwise commonly known as peptones or peptides, are used in a wide variety of products in fermentation and biotechnology industries. The term “peptone” was first introduced in 1880 by Nagelli for growing bacterial cultures. However, later it was discovered that peptones derived from the partial digestion of proteins would furnish organic nitrogen in readily available form. Ever since, p- tones, which are commonly known as protein hydrolysates, have been used not only for growth of microbial cultures, but also as nitrogen source in commercial fermen- tions using animal cells and recombinant microorganisms for the production of value added products such as therapeutic proteins, hormones, vaccines, etc. Today, the characterization, screening and manufacturing of protein hyd- lysates has become more sophisticated, with the introduction of reliable analytical instrumentation, high throughput screening techniques coupled with statistical design approaches, novel enzymes and efficient downstream processing equipment. This has enabled the introduction of custom-built products for specialized appli- tions in diverse fields of fermentation and biotechnology, such as the following. 1. Protein hydrolysates are used as much more than a simple nitrogen source. For example, the productivities of several therapeutic drugs made by animal cells and recombinant microorganisms have been markedly increased by use of p- tein hydrolysates. This is extremely important when capacities are limited. 2. Protein hydrolysates are employed in the manufacturing of vaccines by ferm- tation processes and also used as vaccine stabilizers.
