The authors have been given ample opportunity to present their thoughts and speculation on membrane fluidity and related phenomena in a more expanded form than is usually possible in reviews of this type. The chapters in this book are arranged in three sections, the first of which covers physical studies of membrane fluidity and related phenomena on the molecular level. Cell biological approaches, advanced genetics, biophysics and biochemistry have greatly contributed to an increase in our knowledge in this field.lt is obvious however, that the three major membrane constituents - lipids, proteins and carbohydrates- are studied, in most cases separately and that a coherent overview of the various aspects of membrane biogenesis is not readily available. The NATO Advanced Study Institute on "New Perspectives in the Dynamics of Assembly of Biomembranes" intended to provide such an overview: it was set up to teach students and specialists the achievements obtained in the various research areas and to try and integrate the numerous aspects of membrane assembly into a coherent framework. The articles in here reflect this. Statting with detailed contributions on phospholipid structure, dynamics, organization and biogenesis, an up to date overview of the basic, lipidic backbone of biomembranes is given. Extensive progress is made in the research on membrane protein biosynthesis. In particular the post- and co-translational modification processes of proteins, the mechanisms of protein translocation and the sorting mechanisms which are necessary to direct proteins to their final, intra - or extracellular destination have been characterized in detail. Modern genetic approaches were indispensable in this research area: gene cloning, hybrid protein construction, site directed mutagenesis and sequencing techniques elucidated many functional aspects of specific nucleic acid and amino acid sequences.

The elucidation of the pathways of fatty acid biosynthesis and oxidation, the delineation of the biogenesis of cholesterol from small-molecular weight precursors, the structure proof of simple and complex lipids from plants, animals, and microorganisms, are excellent examples of the spectacular advances made during the golden era of lipid biochemistry. The multifaceted discoveries in these diverse areas of study could be attributed to development of highly sophisticated column chromatographic techniques for separation and purification of simple and complex lipids. The advent of thin-layer chromatography as well as gas liquid chromatography provided an explosive impetus to research developments in this field. Concomitant advances in mass spectrometry allowed an interface with gas-liquid chromatography which spawned even greater insight into the structure of lipids. These eventful days of lipid chemistry nearly 25 years ago led to a relatively quiescent period wherein scientists applied these newly available techniques to investigation of the behavior of isolated (lipid) enzyme systems and to unraveling the intricacies of the metabolic behavior of lipids in the intact cell or whole organisms. Then, in the early 1960s, a decided change in research emphasis developed with the advent of a simple, reproducible procedure for the isolation of cell membranes.

This was the first time the Symposium was held in the United States. The list of previous host cities reads, Norwich, Karlsruhe, Goteborg, Paris, Groningen, Neuchatel. The addition of Davis to this distinguished list was made by the organizers with the doubts of people who give invitations to parties - will anybody come? In fact 155 participants registered and there were 21 spouses in attendance. The scientific program was composed of nine sessions: biochemistry of isoprenoids and sterols, function of isoprenoids and sterols, structure and function of lipids, biosynthesis of complex lipids, fatty acid oxygenases and desaturases, medium and long chain fatty acids, interaction of university, government and industrial research, algal lipids, and genetics and biotechnology. In addition to these sessions of plenary lectures, there were four poster sessions in which about 140 posters were presented. All of this was packed into four days, and there was some comment about the scarcity of time to ask questions of the speakers, discuss the posters and even to eat lunch. The compression of the program was a result of the continued desire of the organizing committees to avoid concurrent sessions. The congregation of participants into a single session increases interaction and generates a feeling of unity at these symposia.
