It covers the "state of the arts" with respect to the use of hemopoietic stem cells collected from the peripheral blood for the reconstitution of hematopoiesis after myeloablative therapy. If is is accepted that hematopoietic function in the mammalian organism is the result of stem cell seeding of an appropriate stromal matrix, then the use of blood derived stem cells for hematopoietic reconstitution represents the "physiological form" of the (re) establishment of a hematopoietic bone marrow. All observations to date are compatible with the assumption that stem cells migrate via the blood stream from extraembryonic hematopoietic tissue to the fetal liver to establish there a first intraembryonic site of blood cell formation and especially of stem cell replication and proliferation. This fetal liver tissue appears then to be the major source for the seeding offetal bone marrow stroma as it develops sequentially in all the bones of the skeleton - in other words during most of the entire embryonic development. There is a very high concentration of stem cells in the blood of the embryo (more than 20000 CFU-GM per ml in the 22nd week) and the stem cells in cord blood seem to be the "tail end" of a dramatic "stem cell traffic" in the embryo to establish the hemopoietic as well as lymphopoietic tissue.

The first volume, published in 1980, stressed the relationship of development and cancer as reflected in the production of markers by cancer that are also produced by normal cells during fetal development. The concept that cancer represents a problem of differentiation was introduced by Barry Pierce in describing differenti ation of teratocarcinomas. Highlighted were lymphocyte markers, alphafetoprotein, carcinoembryonic antigen, ectopic hormones, enzymes and isozymes, pregnancy proteins, and fibronectin. The second volume, published in 1982 and coedited with Britta Wahren, focused on the diagnostic use of oncological markers in human cancers, which were systematically treated on an organ by organ basis. At that time, the application of monoclonal antibodies to the identification of cancer markers was still in a very preliminary stage. A general introduc tion to monoclonal antibodies to human tumor antigens was given there by William Raschke, and other authors included coverage of those mark ers then detectable by monoclonal antibodies in their chapters.

It is the most common extracranial solid tumor occurring in children and exhibits a marked variability in outcome when the disease is categorized by clinical (e.g. age or stage) and biologic characteristics. This book gives an introduction into the clinical features of progressive neuroblastoma and focuses on molecular-targeted therapies and immunotherapies of this disease. It has become increasingly clear that MYCN (v-myc avian myelocytomatosis viral oncogene neuroblastoma derived homolog) holds a key position in neuroblastic transformation and gene expression in normal and transformed cells. In the 14 chapters important topics such as genomic alterations in neuroblastoma and strategies for indirect molecular targeting of MYCN are discussed.
