Natriuretic peptides are a family of three structurally related hormone/paracrine factors. Atrial natriuretic peptide (ANP) and B-type natriuretic peptide (BNP) are secreted from the cardiac atria and ventricles, respectively. 1. ANP signals in an endocrine and paracrine manner to decrease blood pressure and cardiac hypertrophy. 2. BNP acts locally to reduce ventricular fibrosis. 3. C-type natriuretic peptide (CNP) primarily stimulates long bone growth but likely serves unappreciated functions as well. ANP and BNP activate the transmembrane guanylyl cyclase, natriuretic peptide receptor-A (NPR-A). CNP activates a related cyclase, natriuretic peptide receptor-B (NPR-B). Both receptors catalyze the synthesis of cGMP, which mediates most known effects of natriuretic peptides. A third natriuretic peptide receptor, natriuretic peptide receptor-C (NPR-C), clears natriuretic peptides from the circulation through receptor-mediated internalization and degradation. However, a signaling function for the receptor has been suggested as well. A few mutations in these proteins have been reported in humans. Synthetic analogs of ANP (anaritide and carperitide) and BNP (nesiritide) have been investigated as potential therapies for the treatment of decompensated heart failure and other diseases. · Anaritide and nesiritide are approved for use in acute decompensated heart failure. New clinical trials are examining the effect of nesiritide and novel peptides, like CD-NP, on these critical parameters. Book excerpt: Astrocytes were the original neuroglia that Ramón y Cajal visualized in 1913 using a gold sublimate stain. This stain targeted intermediate filaments that we now know consist mainly of glial fibrillary acidic protein, a protein used today as an astrocytic marker. Cajal described the morphological diversity of these cells with some ast- cytes surrounding neurons, while the others are intimately associated with vasculature. We start the book by discussing the heterogeneity of astrocytes using contemporary tools and by calling into question the assumption by classical neuroscience that neurons and glia are derived from distinct pools of progenitor cells. Astrocytes have long been neglected as active participants in intercellular communication and information processing in the central nervous system, in part due to their lack of electrical excitability. The follow up chapters review the “nuts and bolts” of ast- cytic physiology; astrocytes possess a diverse assortment of ion channels, neu- transmitter receptors, and transport mechanisms that enable the astrocytes to respond to many of the same signals that act on neurons. Since astrocytes can detect chemical transmitters that are released from neurons and can release their own extracellular signals there is an increasing awareness that they play physiological roles in regulating neuronal activity and synaptic transmission. In addition to these physiological roles, it is becoming increasingly recognized that astrocytes play critical roles during pathophysiological states of the nervous system; these states include gliomas, Alexander disease, and epilepsy to mention a few.

Peptides and peptide-like compounds are found among toxins and antibiotics. It is unlikely that this process, an almost explosive broadening of the field, will come to a sudden halt. By now it is obvious that Nature has used the combination of a small to moderate number of amino acids to generate a great variety of agonists with specific and often highly sophisticated functions. Thus, peptide chemistry must be regarded as a discipline in its own right, a major branch of biochemistry, fairly separate from the chemistry of proteins. Because of the important role played by synthesis both in the study and in the practical preparation of peptides, their area can be considered as belonging to bio-organic chemistry as well. The already overwhelming and still increasing body of know ledge renders an account of the history of peptide chemistry more and more difficult. It appears therefore timely to look back, to take stock and to recall the important stages in the development of a new discipline.
