C'est actuellement la méthode de référence dans l'industrie pharmaceutique pour produire des microparticules. C'est un procédé qui permet de d'obtenir des particules de formes régulières et sphériques et une granulométrie resserrée. La granulométrie d'une poudre sèche a une importance capitale dans les procédés pharmaceutiques. Une poudre ayant une granulométrie dispersée est dite polydisperse. La polydispersité peut être problématique pour la formulation de formes sèches car des particules de diamètres différents peuvent entrainer des comportements différents au sein d'une même population. La polydispersité prend son origine dans le procédé même de fabrication de la poudre. Face à cela de nouvelles méthodes ont vu le jour pour produire de la poudre monodisperse. L'étude expérimentale menée sur la caléfaction a permis de montrer qu'il est possible de fabriquer des microgranules par caléfaction d'une solution de cellulose microcristalline associée ou non à de l'acide ascorbique. Ce procédé devrait permettre la fabrication de populations de granules dont la distribution de taille peut être considérée comme monodispersée.
