An understanding of the neural basis of hearing is thus a starting point for elucidating the mechanisms that are essential to human communication. The cochlear nucleus is the first region of the brain to receive input from the inner ear and is therefore the earliest stage in the central nervous system at which auditory signals are processed for distribution to higher centers. Clarifying its role in the central auditory pathway is crucial to our knowledge of how the brain deals with complex stimuli such as speech, and is also essential for understanding the central effects of peripheral sensorineural hearing loss caused by, for example, aging, ototoxic drugs, and noise. Ambitious new developments to assist people with total sensorineural deafness, including both cochlear and cochleus nuclear implants, require a detailed knowledge of the neural signals received by the brainstem and how these are processed. Recently, many new data have been obtained on the structure and function of the cochlear nucleus utilizing combinations of anatomical, physiological, pharmacological and molecular biological procedures. Approaches such as intracellular dye-filling of physiologically identified neurons, localization of classical neurotransmitters, peptides, receptors and special proteins, or gene expression have opened the door to novel morphofunctional correlations.

Book excerpt: The spiral ganglion neurons (SGN) in the mammalian inner ear play a key role in transferring the neural activity transduced by the hair cells to the central auditory system. The neurophysiological properties of the SGN are determined in large part by the voltage-gated and ligand-gated ion channels. There is a growing awareness that the voltage-gated and ligand-gated ion channels which are expressed on neurons throughout the brain undergo significant modifications during early postnatal life and following damage and recovery. This is a process often referred to as neural plasticity. The SGN are anatomically difficult to access, therefore relatively little is known about the changes in the SGN's voltage and ligand gated ion channels during early postnatal life. Recent studies indicate that the survival of SGN can be profoundly altered by neurotrophic factor 3 (NT-3) and brain derived neurotrophic factor (BDNF) released by hair cells of the inner ear. However, it is unclear how NT-3 and BDNF affect the development of voltage and ligand gated channels expressed on SGN. To address the lack of knowledge in this area, the whole-cell patch clamp recording was utilized in order to study the early postnatal changes which occur in several voltage and ligand gated channels of SGN. Since preliminary studies have shown that NT-3 and BDNF can influence the survival and function of SGN, I also studied their affects on the developmental changes. Using whole cell patch clamp recording, I found that the amplitude of the voltage-gated potassium current increased significantly during early postnatal development (P1-P5). However, when isolated SGN were maintained in culture from 1 to 28 days (C1-C28), the voltage-gated potassium current remained relatively constant while the voltage-gated sodium current increased significantly. During C1-C28 the firing pattern of action potentials evoked by suprathreshold current shifted from fast adapting to slow adapting. The developmental changes seen in the voltage gated potassium channels are likely to contribute to this shift in SGN action potential firing pattern. Glutamate receptors are believed to be the dominant excitatory neurotransmitter receptors of SGN. To investigate the development of glutamate receptors, I applied kainic acid (KA), a non- N -methyl-D-aspartic acid (non-NMDA) glutamate receptor agonist to the SGN.
