Book excerpt: Mesoporous materials with engineered surface properties are of interest for molecular separations, catalysis, drug delivery, and chemical sensing. One of the longstanding chemical challenges in the engineering of nanomaterials is to control the placement of different chemistries in spatially distinct regions on a nanoscale object. This thesis focuses on discovering and understanding processes to prepare such spatially differentiated chemistries on porous silicon. For the porous silicon system, the ability to prepare pores of average diameter anywhere from 1 to 200 nm allows the harnessing of surface tension and capillary forces to promote or obstruct the infiltration of reagent for selective modification. The first process investigated involves placing different chemistries on the pore walls by means of microdroplet patterning. In this method, a chemical resist is drop-coated on a porous silicon sample to mask distinct regions across the plane of the chip for subsequent chemical modification. Two chemistries, silicon oxide and silicon-methyl, are demonstrated here. The differential partitioning of test molecules on the resulting hydrophilic/hydrophobic film is achieved by simultaneous optical reflectance measurement of both regions, where the reflectance spectrum contains a convolution of the Fabry-Pérot interference spectrum for both types of surface chemistries. A second approach to engineer porous Si nanostructures that is investigated uses a hydrophobic organic liquid as a chemical resist; it is infiltrated into the pores to mask the interior of the porous silicon film, while the exterior surface and the pore mouths of the film are subjected to an aqueous chemical reaction with HF and subsequent chemical modification by thermal hydrosilylation. When chemically modified with a hydrophobic dodecyl species, the resulting film has a hydrophilic interior and a hydrophobic outer surface. The ability of these core-shell porous nanostructures to admit and release small molecules is assessed and exploited. The last portion of the thesis focuses on an evaluation of chemically modified porous silicon particles as oral drug delivery material, with an emphasis on the ability of the interior of the porous silicon nanostructure to protect physiologically unstable drugs. Partially oxidized porous silicon particles show no toxicity to nematodes at particle concentration up to 1 mg/mL in culture media. In vitro experiments show the particles protect anthelmintic protein-based drug Cry5B from hydrolytic degradation in simulated gastric fluid, and the bioactivity against nematodes is maintained. However, much reduced bioactivity of the therapeutic particles is observed In vivo on hookworm-infected hamsters. The lack of effectiveness in treating the disease In vivo is attributed to the short residence time of the particles in the gut of the animals.

This thesis begins with an overview on current separation techniques and progress in the field of optical sensors and nanomaterials. The second half of the introduction discusses recent development in porous silicon material with the focus on biosensing and molecular filtration applications. The objective of this thesis is to explore porous silicon as a multifunctional material with the ability to separate, process, and detect biomolecules at low concentration and in real-time with minimal sample preparation. Interrogation of porous silicon material as a multifunctional nanostructure involved three major aspects: 1) manipulation of its optical and spectral information for encoding and signal processing applications, 2) examination of the effect of its physical properties on molecular transport within its porous structure, 3) investigation of analyte-pore surface interaction for enhanced selectivity or better separation based on analyte surface moieties. The last chapter of this thesis provides an example of exploiting porous silicon as a multifunctional matrix that is capable of capturing and concentrating analyte while processing the signal, providing a new strategy for bioanalytics.

This thesis describes cargo loading chemistry, surface chemistry, molecularly targeted delivery and bioimaging applications using porous silicon nanomaterials. After a brief introduction to porous silicon materials for biomedical applications, Chapter 2 describes a single-step procedure to simultaneously load and protect a model siRNA therapeutic in porous silicon nanoparticles (pSiNPs). Exogenous calcium ions precipitate with locally generated silicic acid to form calcium silicate, which serves to encapsulate the siRNA payload in pSiNPs. The target gene knockdown efficiency in vitro and target tissue accumulation of delivered siRNA in vivo are demonstrated. Chapter 3 presents a facile chemical modification of the surface of the hydroxylated silicon nanostructure. The reaction, a ring-opening heterocyclic silane "click" reaction, is a rapid and efficient means to obtain high surface coverage while preserving the open pore structure and intrinsic photoluminescence of the original silicon nanostructure. This chemistry is sufficiently mild to maintain the activity of payload proteins. Chapter 4 presents the example of pSiNPs as an imaging agent, which are targeted to tumor tissues in vivo using an iRGD peptide targeting probe, and the nanoparticles are imaged by two-photon microscopy. Superior photostability and low systemic toxicity are observed. Chapter 5 discusses enhanced photoacoustic signals that can be obtained from indocyanine green (ICG) when it is encapsulated in pSiNPs. The photoacoustic response from ICG is enhanced 17-fold when it is sealed in pSiNPs. The substantially improved performance is attributed to the low thermal conductivity of pSiNPs and their ability to protect loaded ICG from photolytic degradation.

Book excerpt: Porous silicon (pSi), with its nanoscale architecture, acts as a promising resorbable biomaterial for a broad variety of applications: biosensors, orthopedic tissue engineering, and controlled drug delivery. The most conventional methods for preparing porous silicon particles are anodization of silicon wafers and metal-assisted chemical etching (MACE). These established techniques require elemental crystalline silicon feedstocks, and the use of corrosive hydrofluoric acid (HF) and organic solvents. Rather than employing these methods, we have developed an eco-friendly alternative of producing pSi from silicon accumulator plants/agriculture waste. Two different silicon accumulator plants Bambuseae (tabasheer) and Equisetum telmateia (great horsetail)have been investigated for this purpose, and high surface area porous silicon nanostructures prepared using a magnesiothermic reduction of silica isolated from these plants. This fabrication process of pSi includes extraction of biogenic silica from the ground plant by calcination, followed by reduction with magnesium, and purification of reduced Si product. Details of fabrication process and characterization of pSi powders by a combination of scanning electron microscopy (SEM), energy dispersive X-ray analysis (EDX), X-ray diffraction (XRD), transmission electron microscopy (TEM), and low temperature nitrogen gas adsorption measurements will be outlined in this presentation. As pointed out above, Bambuseae serves as a viable eco-friendly starting materials for fabricating porous silicon (pSi).At the same time, if the selected plant leaf components contain medicinally-active species (antibacterial) as well, then the single substance can provide not only the nanoscale high surface area drug delivery carrier, but the drug itself. With this idea in mind using a single plant source (Bambuseae) we have developed porous silicon based drug delivery carrier matrix for the sustained release of antibacterials. Nanoentrapment of vitamins, nutrients, and plant based therapeutics is another attractive area in food and pharmaceutical industries to address the challenges of stability and bioavailability of entrapped agents. One way to increase the bioavailability of the drugs is by enhancing their solubility. We have evaluated the ability of nanostructured plant-derived porous silicon particles (pSi) as potential candidates to increase the solubility of plant extracts rich in therapeutic polyphenolic compounds (Equisetum arvense) and a poorly water soluble vitamin (vitamin d3). We have also evaluated the ability of plant derived porous silicon nanoparticles (pSiNPs) as gene delivery vehicles. Following necessary reduction of pSiNP particle size and functionalization with primary amine moieties at the nanoparticle surface, the binding of a reporter plasmid DNA, pEF1-eGFP (endothelial factor 1 enhanced green fluorescent protein, 6429 bp) to the pSiNPs was investigated, followed by attempted transfection of HEK293 cells.

The second chapter discusses chemical modifications of nanostructured porous Si for stabilizing the material in biologically relevant media while providing an extended release of a therapeutic in vitro. This chapter also demonstrates the utility of the porous silicon optical signatures for effectively monitoring drug release from the system and its applications for development of a self-reporting drug delivery device. In chapter three, the concept of providing a triggered release of a therapeutic from porous silicon microparticles through initiation by an external stimulus is demonstrated. The microparticles are chemically modified, and the release is enhanced by a short application of ultrasound to the particulate system. The effect of ultrasound on the drug release and particle size is discussed. Chapter four presents a new method for sustaining the release of a monoclonal antibody from the porous matrix of porous SiO2. The therapeutic is incorporated into the films through electrostatic adsorption and a slow release is observed in vitro. Its tunable structural features, low toxicity profile and readily modifiable surface render this material extremely useful for a wide variety of applications. By chemically modifying surface species, such as silicon hydrides and silicon oxides with silanes, the properties of porous silicon can be enhanced for its use for chemical sensing, biomedical imaging, and drug delivery. After a brief introduction to porous silicon materials, the first part of this dissertation details surface-modified porous silicon photonic crystals for the chemical sensing of toxic vapors and nerve agents. Chapter 2 utilizes a dual-peak porous silicon photonic crystal embedded with specific for the selective detection of hydrogen fluoride (HF), hydrogen cyanide (HCN), and the chemical nerve agent diisopropyl fluorophosphate (DFP). The pore walls are rendered hydrophobic with octadecylsilane to aid with the loading of the colorimetric molecules while being insensitive to humidity fluctuations. This provides a robust means to develop a remote detection system for chemical agents. Chapter 3 employs the same photonic crystal, modified, however, with a specialized protein-based gatekeeper that is rendered semi-permeable only in the presence of HCN. This is one of the first novel designs of a bio-inorganic sensor capable of detecting chemical agents with high specificity and precision. The second portion of the dissertation describes how surface-modified porous silicon nanoparticles can be applied in biomedical applications. The first project details the use of Anti-KIT protein DNA-aptamers decorated onto a fluorescently labelled porous silicon nanoparticle for the in vitro and in vivo imaging of gastrointestinal stromal tumors. This work provides an effective platform in which aptamer-conjugated porous silicon nanoparticle constructs can be used for the targeted imaging of KIT-expressing cancers. The final project utilizes hydrophobic porous silicon nanoparticles for the delivery of erucamide, a highly hydrophobic fatty acid amide, within the retina. By harnessing the versatility of porous silicon, erucamide's target cells and mechanism of neurotrophic action can be identified.
