Equally impressive is our appreciation of the cause of tumors based on observation. Even in the earliest era of the study of infectious diseases, it was proposed that tumorous growth in animals and birds resulted from "minute" infectious particles. Experiments then supported the hypothesis, that the etiologic agent in many animal tumors was viral. The development of molecular biology, supported by technical advances and conceptual understanding of macromolecular action, led to an explosive increase in studies of animal oncogenic viruses. For a decade, new findings emerged from research laboratories revealing the enormous variety of such agents, the complex ity of their interactions with cells, and the tantalizingly possible mechanisms by which they might cause malignant transformation of the cell. Repeatedly, clues emerged which suggested the intervention of viral agents in human tumors. A breathless excitement pervaded both the scientific and public communities as highly publicized findings rapidly followed one another. The excitement was no less scientific than it was practical, for implicit in the concept of the viral oncogen is the possibility of specific virostatic or virotoxic agents or of immunization.

This book then addresses the nature and origin of transforming RNA viruses and gives a detailed review of knowledge concerning the genomic structure of type C viruses. Translational products encoded by the type C viral genome are examined in ensuing chapters, emphasizing the viral reverse transcriptase. Other mammalian retroviruses, including the mouse mammary tumor virus and type D isolates of primates, are also described. The book concludes by evaluating the possibility of direct etiologic involvement of either endogenous or exogenous RNA tumor viruses in human cancers. This book will be of value both to graduate students and to established investigators with specific interest in other aspects of molecular biology.

Book excerpt: It has long been suspected, and recently confirmed, that there is an etiologic relationship between several viruses and naturally occurring neoplasias. Virus precursors in the form of nucleic acids or antigens have consistently been associated with certain neoplasias. However, the role of these virus-specified precursors in etiology remains obscure. Recent studies of virus-associated neo plasias have led to advances in molecular techniques, which have yielded increas ingly sensitive assays for detection of virus-specific nucleic acids, and which have enabled the disruption of virus particles without concomitant loss in antigenicity of the components. These procedures have, in turn, resulted in molecular probes that allow more definitive evaluation of the host response to its virus and to the tumor cell with which the virus or its precursors are associated. Evaluations of the immune response and status of the host have provided important informa tion about carcinogenesis and the tools for seroepidemiological studies of a variety of cancers. These seroepidemiological studies have demonstrated that several human cancers, e. g. , Burkitt's lymphoma and nasopharyngeal carcinoma, are probably virus-induced, and that antibodies that are diagnostic and prog nostic for these diseases are detectable. The conclusion that feline leukemia is a disease transmitted horizontally by a virus resulted primarily from immunologi cal experiments.

The purpose of the symposium was to present the current status of the challenging cancer problem, breast carcinoma, to the staff and students of this medical center and to students and in terested physicians from other institutions in the Chicago area. In a fashion similar to the other teaching symposia held in 1964, 1966, 1968,. and 1970, this symposium attracted over 450 physicians and scientists. In the course of one and one half days the audience had the opportunity to listen to 18 invited speakers and to the lively discussions. The formal presentations are recorded in these pages.
