Book excerpt: There is a critical need for new environmentally friendly processes in the United States chemical industry as legislative and economic pressures push the industry to zero-waste and cradle-to-grave responsibility for the products they produce. Carbohydrates represent a plentiful, renewable resource, which for some processes might economically replace fossil feedstocks. While the conversion of biomass to fuels, is still not generally economical, the selective synthesis of a commodity or fine chemical, however, could compete effectively if appropriate catalytic conversion systems can be found. Oxygenated organics, found in a variety of products such as nylon and polyester, are particularly attractive targets. We believe that with concerted research efforts, homogeneous transition metal catalyzed reactions could play a significant role in bringing about this future green chemistry technology.

Book excerpt: Transition metal catalysis belongs to the most important chemical research areas because a ubiquitous number of chemical reactions are catalyzed by transition metal compounds. Many efforts are being made by industry and academia to find new and more efficient catalysts for chemical processes. Transition metals play a prominent role in catalytic research because they have been proven to show an enormous diversity in lowering the activation barrier for chemical reactions. For many years, the search for new catalysts was carried out by trial and error, which was costly and time consuming. The understanding of the mechanism of the catalytic process is often not very advanced because it is difficult to study the elementary steps of the catalysis with experimental techniques. The development of modern quantum chemical methods for calculating possible intermediates and transition states was a breakthrough in gaining an understanding of the reaction pathways of transition metal catalyzed reactions. This volume, organized into eight chapters written by leading scientists in the field, illustrates the progress made during the last decade. The reader will obtain a deep insight into the present state of quantum chemical research in transition metal catalysis.

Book excerpt: Carbohydrates are synthetically challenging molecules with vital biological roles in all living systems. To better understand the biological functions of this fundamentally important class of molecules, novel methodologies are urgently needed in two different directions: building block synthesis and selective glycosylation. During my Ph.D. study, I developed two different methodologies for building block synthesis, which could simplify the assembly of oligosaccharides.Differentiation of hydroxy groups on the carbohydrate is usually considered challenging as a result of the almost identical chemical environment. 1,2-trans diols are even more difficult to deal with compared to 1,2-cis diols. In my first project, I developed a method for regio- and stereo-selective OH carbene insertion together with my collaborator. Under very mild conditions, a series of 1,2-trans diols can be selectively functionalized. The regioselectivity originated from interactions between substrates and transition-metal catalysts, while the stereochemistry came from intramolecular proton delivery. The newly introduced moiety can work as a protection group, function group for the glycoconjugate mimic synthesis, or participate in the following glycosylation reactions and impact the anomeric selectivity. Inspired by the regio-selectivity of first projects, I developed another transition metal-catalyzed reaction, which could successfully epimerize one hydroxyl group in 1,2-trans diols, thus many rare sugars can be accessed easily starting from readily available common sugars. Compared to the traditional route, the method I developed can simplify the synthesis significantly and is more environmentally friendly. Synthesis of aminosugars through direct amination of 1,2-trans diols is also investigated. Bromodomain (BRD) is an evolutionarily conserved protein-protein interaction module that can recognize and bind acetylated lysine residues in histones and many other proteins. BRD4 protein, which is considered a transcriptional factor and epigenetic regulator, can bind to acetylated histone and recruit protein complexes to stabilize the binding of RNA polymerase II to histone. The dysregulation of BRD4 can cause cancer and other diseases such as immune and inflammatory diseases. Because of this, the BRD4 protein has been long considered an important therapeutic target. In the third project, I worked on the development of tissue-selective BRD4 degraders, aiming to provide a novel therapy for prostate cancer. By attaching tissue-selective ligands to BRD4 degraders, we successfully achieved selectivity among different cell lines, which proved our initial hypothesis. The tri-functional degraders will be tested in vivo in the future.

Book excerpt: This authoritative reference work presents comprehensive information about one of the most important and most wide-spread classes of (bio)organic compounds: the polysaccharides. The comprehensive and thoroughly up-to-date handbook presents the sources, identification, analysis, biosynthesis, biotechnology and applications of important polysaccharides likes starches, cellulose, chitin, gum and microbial polysaccharides. Polysaccharides can exhibit complex structure and various functional activities. These bio macromolecules can therefore serve as raw materials for various different materials, e.g. rayon, cellulose acetate, celluloid and nitrocellulose; and they find multiple applications, for instance as surgical threads (chitin), as sources of energy, dietary fibers, as blood flow adjuvants, in cosmetics, emulsion stabilizers, film formers, binders, viscosity increasing agents or skin conditioning agenta, as food additives in gums, chewing gum bases and as vaccines. Polysaccharides form the basis for useful products, like xanthan gum, dextran, welan gum, gellan gum, diutan gum and pullulan. Some of the polysaccharide-derived products have interesting and useful properties and show biological activities, such as immunomodulatory, antibacterial, anti-mutagenic, radioprotective, anti-oxidative, anti-ulcer, antidepressant, anti-septicaemic or anti-inflammatory activities. All these applications and properties of polysaccharides are for the first time compiled in a thorough and comprehensive overview in the present work. This reference work is organized thematically in four parts: Part I. Polysaccharides: Occurrence, Structure, Distribution and Biotechnology. Part II. Methods. Part III. Bioactive Polysaccharides. Part IV. Polysaccharides as Food. This reference work is edited by experienced experts, all chapters are written by well recognized international specialists. It is useful to all those working in the field of botany, phytochemistry, pharmacy, drug delivery, molecular biology, metabolomics, forestry, environment, conservation, biotechnology and NGOs working for forest protection.

Available in PDF, EPUB and Kindle. Book excerpt: Soluble catalysts are used extensively in many branches of chemistry and are indeed a vital constituent of many natural processes. They find wide application throughout the chemical industry where they assist in the production of several million tonnes of chemicals each year. Since homogeneous systems, especially those incorporating transition metals, often function effectively under milder conditions than their heterogeneous counterparts, they are becoming increasingly important at a time when the chemical industry in particular, and society in general, is seeking ways of conserving energy and of making the best possible use of available resources. My principal objective in· writing this book is to engender sufficient enthusiasm for, and knowledge of, the subject in the reader that he or she will be encouraged to begin, or continue, to make their own contribution to advancing our knowledge of homogeneous catalysis. After attempting to acquaint the reader with some of the ground rules I have tried to describe the .present scope, and the future potential, of this fascinating field of chemistry by drawing both on academic and on industrial data sources. This approach stems from a personal conviction that future progress could be considerably hastened by a more meaningful dialogue between chemists working both in industrial and in academic research institutions. Wherever possible, examples of the commercial application of homogeneous catalyst systems have been included and no attempt has been made in any way to disguise the many unresolved questions and exciting challenges which still pervade this rapidly developing area.

Book excerpt: Transition metal-catalyzed cross-coupling reactions have proved to be powerful tools for carbon-carbon as well as carbon-heteroatom bond formation in the development of synthetic methodologies for applications ranging from pharmaceuticals to materials. This book, consisting of an editorial, two reviews and two articles, focuses on recent promising research and novel trends in the field of cross-coupling reactions, employing a range of different catalysts. A review by Kostas and Steele provides a survey of the research in the area of cross-coupling catalytic reactions with transition metal complexes based on the thiosemicarbazone unit and a discussion of the prospects for future developments. Another review by Polychronopoulou, Shaya and co-authors describes the progress made over the 21st century concerning the utilization of C(sp3)-organoboranes as partners in metal-catalyzed C(sp3)-C(sp2) cross-couplings, such as B-alkyl Suzuki-Miyaura reactions. The article by Waldvogel, Breinbauer and co-authors demonstrates for the first time the synthetic potential of combining the electro-oxidative dehydrogenative cross coupling of ortho-substituted phenols with Pd-catalyzed cross-coupling reactions. In the second article, Štĕpnička and co-workers describe the preparation of palladium catalysts deposited over silica gel-bearing composite amide-donor functional moieties on the surface, which were evaluated in the Sonogashira-type cross-coupling of acyl chlorides with terminal alkynes.
