Book excerpt: The surface enhanced Raman scattering spectra of crystal violet and R6G and a number of colourless molecules adsorbed on silver colloid aggregates were studied as a function of power and position using a Raman microscope. The anti-Stokes signals of the dye molecules show line intensities much greater than the expected on the basis of the Boltzmann equilibrium population of the excited vibrational state. The large anti-Stokes intensities are attributed to the difference in Stokes and anti-Stokes Raman cross-sections resulting from pre-resonance or resonance Raman processes in the adsorbate-substrate complex rather than to optical pumping of molecular vibrational excited states as proposed by Kneipp and co-workers [Phys. Rev. Lett., 76, 2444, 1996]. Far-field imaging with a Raman microscope shows no significant inhomogeneity of the Raman enhancement in the images of the deposited self-affine silver colloid clusters down to spatial resolutions of I [mu]m2. A near-field scanning optical microscope (NSOM) operating with a shear-force feedback distance-control mechanism was employed to study the optical response of self-affine, deposited colloidal silver films. In the near-field region, NSOM is capable of an optical resolution down to 100 nm which is beyond the traditional diffraction limit of far field optics. Near-field Rayleigh microscopy with simultaneous topographic imaging indicates that the electromagnetic fields on the surface of the self-affine films are localized in regions smaller than the excitation wavelength. The pattern of high-field regions varies greatly with excitation wavelengths and does not correlate with topography. Local spectra of self-affine film showed distinct resonances arising with highly location-dependent frequencies. The most highly resolved of these resonances correspond to individual surface plasmon normal modes. Specific vibrational bands from the SERS spectra of plithalazine adsorbed on self-affine, silver colloid films were mapped over portions of the film surface in the near-field. The near-field Raman intensity maps display patterns of field localization in subwavelength regions. The pattern of field localization varies greatly with scattered wavelength. Observations in the near-field validate recent theories of the optical response of self-affine fractal objects [Phys. Report, 61, 272, 1996].

Book excerpt: Localized surface plasmon resonance resulting from the concerted oscillations of conduction-band electrons in noble-metal (Au, Ag) nanostructures greatly induces enhanced electric ([italic E]) fields in confined nanoscale locations, such as on the tips of nanorods or in the junctions of nanodimers. These [italic E]-field enhanced locations are called hot spots. In the vicinity of hot spots, Raman scattering spectra of biochemicals can be substantially amplified with an [italic E]4 dependence due to the [italic E]-field enhancement of both the incident light and Raman spectra. This is called surface enhanced Raman scattering (SERS). SERS is known for its high sensitivity in providing fingerprint vibrational information of molecules. It has triggered intense interest because of its potential applications for label-free and multiplex biochemical detection relevant to medical, environmental and defense purposes. However, the tremendous potential of SERS for ultrasensitive detection has still not materialized because of four major obstacles: (1) it is extremely difficult to obtain a large number of hotspots for sensitive and reproducible detection due to the stringent requirement of hot spots of only a few nanometers; (2) it is arduous to achieve ultrasensitivity for the detection of a single/few molecules; (3) it is challenging to assemble the hot-spots at designated positions for location predicable sensing; and (4) it is even more difficult to change the state-of-the-art static/passive sensing schemes into dynamic/robotized schemes and also to incorporate multi-functionality into a single SERS nanostructure. In this research, we addressed the aforementioned problems by rational design, fabrication and robotization of ultrasensitive SERS nanomotor sensors. A nanomotor sensor consists of a tri-layer structure with a three-segment Ag/Ni/Ag nanorod as the core, a thin layer of silica in the center, and uniformly distributed Ag nanoparticles as the outer layer. The inner metallic nanorod core is the key structure in realizing the concept of the robotization of nanosensors, which can be electrically polarized and thus efficiently manipulated by electric tweezers. The presence of the Ni segment in the metallic nanowire core also allows manipulation and assembling by magnetic interactions. The central silica layer provides a supporting substrate for the synthesis of the Ag nanoparticles and separates the Ag nanoparticles from the metallic nanorod core to eliminate the effect of plasmonic quenching. Finally, the outermost layer made of Ag nanoparticles with optimized sizes and junctions provides a large number of hot spots (~1200/ [mu] m2) for ultrasensitive SERS detection with single molecule sensitivity and an enhancement factor (EF) of 1.1×1010. Moreover, two additional SERS enhancement mechanisms were investigated, i.e., the optical management with nanophotonic devices and the near field effect, which can readily increase the EF by 10 and 2 times, respectively, to at least 1011. Finally, three applications of the SERS nanomotor sensors have been demonstrated: (1) the ultrasensitive SERS nanomotors were transported and assembled into a 3×3 array for location predicable sensing of multiplex molecules; (2) ultrasensitive SERS nanomotors were transported to individual living cells amidst many cells for single-cell bioanalysis; and (3) the SERS nanomotor sensors can be controlled to rotate by the electric tweezers for tunable biochemical release and detection. This research, exploring robotized nanosensors by integrating SERS and NEMS, is innovative in both material design and device concept, which could inspire a new device scheme for various bio-relevant applications.

Book excerpt: Nonlinear nanophotonics is a rapidly developing field of research that aims at detecting and disentangling weak congested optical signatures on the nanoscale. Sub-wavelength field confinement of the local electromagnetic fields and the resulting field enhancement is achieved by utilizing plasmonic near-field antennas. This allows for probing nanoscopic volumes, a property unattainable by conventional far-field microscopy techniques. Combination of plasmonics and nonlinear optical microscopy provides a path to visualizing a small chemical and spatial subset of target molecules within an ensemble. This is achieved while maintaining rapid signal acquisition, which is necessary for capturing biological processes in living systems.Herein, a novel technique, wide-field surface enhanced coherent anti-Stokes Raman scattering (wfSE-CARS) is presented. This technique allows for isolating weak vibrational signals in nanoscopic proximity to the surface by using chemical sensitivity of coherent Raman microspectroscopy (CRM) and field confinement from surface plasmons supported on a thin gold film. Uniform field enhancement over a large field of view, achieved with surface plasmon polaritons (SPP) in wfSE-CARSS, allows for biomolecular imaging demonstrated on extended structures like phospholipid droplets and live cells. Surface selectivity and chemical contrast are achieved at ~ 70 fJ/mum2 incident energy densities, which is over five orders of magnitude lower than used in conventional point scanning CRM.Next, a novel surface sensing imaging technique, local field induced metal emission (LFIME), is introduced. Presence of a sample material at the surface influences the local fields of a thin flat gold film, such that nonlinear fluorescence signal of the metal can be detected in the far-field. Nanoscale nonmetallic, nonfluorescent objects can be imaged with high signal-to-background ratio and diffraction limited lateral resolution using LFIME. Additionally, structure of the extended samples' surfaces can be visualized with a nanoscale axial resolution providing topographic information.Finally, a platform for coherently interrogating single molecules is presented. Single-molecule limit SE-CARS on non-resonant molecules is achieved by means of 3D local field confinement in the nanojunctions between two spherical gold nanoparticles. Localized plasmon resonance of the dimer nanostructure confines the probe volume down to 1 nm3 and provides the local field enhancement necessary to reach single-molecule detection limit. Nonlinear excitation of Raman vibrations in SE-CARS microspectroscopy allows for higher image acquisition rates than in conventionally used single-molecule surface enhanced Raman spectroscopy (SERS). Therefore, data throughput is significantly improved while preserving spectral information despite the presence of the metal. Data simultaneously acquired from hundreds of nanoantennas allows to establish the peak enhancement factor from the observed count rates and define the maximum allowed local-field that preserves the integrity of the antenna. These results are paramount for the future design of time resolved single-molecule studies with multiple pulsed laser excitations, required for single-molecule coherence manipulation and quantum computing.
