Book excerpt: Obesity has emerged as a major threat to public health in both the western and developing world. Essentially a disorder of energy balance, obesity occurs when energy intake and storage exceeds expenditure. Much of energy homeostasis depends on the activity and function of adipose tissue. Adipocytes in mammals fall into two categories classified by their primary functions: white fat cells that mediate energy storage and thermogenic fat cells that counteract hypothermia and obesity through adaptive thermogenesis. Whereas white fat and its function as an energy reservoir and endocrine organ have been studied for decades and are relatively well understood, until recently many aspects of the thermogenic fat biology have remained elusive. Accumulating evidence supports the hypothesis that thermogenic fat cells arise from at least two different developmental origins: the ones of a skeletal muscle-like lineage are now called “classical” brown fat cells, and the rest of the thermogenic fat cells are normally referred to as the beige fat cells. The last decade has witnessed an explosion of interest and studies focusing on the regulation of thermogenic fat cells and potential therapeutics targeting these adipocytes. Here we summarize the recent advancements in our understanding of these metabolically active fat cells.

Available in PDF, EPUB and Kindle. Book excerpt: Author of the popular Primal Body, Primal Mind and widely recognized Paleo expert and nutritionist Nora Gedgaudas presents a revolutionary and accessible high-fat diet for maintaining health, losing weight, and reversing some chronic diseases. Popular nutritionist Nora Gedgaudas returns with advice that may sound counterintuitive: Eat fat to burn fat. In Primal Fat Burner, she explains the benefits and science behind a ketogenic (or fat-burning) diet, which switches your metabolism from a dependence on sugar to running on healthy fats. As Gedgaudas reveals, numerous studies in recent years refute the long-promoted anti-saturated fat and anti-cholesterol agenda. Now, “with able, funny writing” (Booklist), Gedgaudas explains the science that fat isn’t always a bad thing—if you know the right kinds of fats to eat. In her comprehensible, enjoyable style, she also lays out a practical meal plan with recipes. Author of the bestselling Grain Brain, Dr. David Perlmutter writes in his foreword that Primal Fat Burner is, “Wonderfully actionable, compassionately taking the reader from ‘why’ to ‘how.’” On this diet, you efficiently and effectively metabolize fat (ketones and free fatty acids) as your primary source of fuel, rather than glucose from carbs, starches, and sugars. Because fat is so satisfying, you naturally wind up eating less—without feelings of hunger or deprivation. And natural dietary fat is ultimately the key to feeling better, looking better, thinking clearer, and living longer. Discover all this and more healthy tips in one of the most groundbreaking nutritional books in recent time.

Book excerpt: This volume presents a collection of protocols that describe methodologies to study thermogenic fat biology from various angles. This book is divided into 2 parts. Part 1 focuses on establishing in vitro culture systems. The chapters in this section introduce techniques on how to isolate, culture, and differentiate primary fat cells from both laboratory mice and humans. This part also presents flow cytometry methods to isolate various subpopulations of precursors within the stromal vascular fraction of the adipose tissue, which contains both preadipocytes and immune cells. Part 2 introduces multiple means to genetically manipulate and evaluate brown and beige fat in vivo. The chapters in this section explore methods on bioenergetics analyses both in vitro and in vivo. They also cover how to evaluate thermogenic fat contents and activity in humans, how to culture these cells though interdisciplinary approaches, and how to use thermogenic fat cell lines to carry out drug screens. Written in the highly successful Methods in Molecular Biology series format, chapters include introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and tips on troubleshooting and avoiding known pitfalls. Thorough and cutting-edge, Thermogenic Fat: Methods and Protocols is a valuable resource for both experts and novices who are interested in learning assays and investigating brown and beige fat functions.

These adipose tissue macrophages (ATMs) have an important role in regulating homeostasis and contributing to the metabolically harmful chronic inflammation seen in some obese individuals. Macrophages are cells exhibiting plasticity, that is to alter their polarization between pro- and anti-inflammatory phenotypes, in response to stimuli in their microenvironments. There has been a deal of evidence supporting the heterogeneity of these ATM populations, however, research into their differential functionality, developmental origin, and phenotype warrants further research to reach a consensus. Further questions remain regarding the nature of this macrophage differentiation and plasticity, and whether it is innately determined or is induced by the specific adipose tissue microenvironment. Differences in the phenotypes of metabolically unhealthy obese (MUO) and the metabolically healthy obese (MHO) individuals suggest that there are differences in both tissues and overall systemic function. The divergent phenotypes seen in MHO and MUO individuals provides evidence that differences in adipose tissue function are determined by local inflammatory responses driven by macrophage plasticity towards pro-inflammatory phenotypes.

Available in PDF, EPUB and Kindle. Book excerpt: We teach our students of behavioural science that one first defines a research problem, and then the most appropriate animal is selected to investigate hypotheses. The reverse order of events is improper: a particular class of animals should not be studied for its own sake. In the case of the Pinnipeds (seals, sea lions, fur seals and walruses) the organism and the problem are essentially the same. The research questions presented in this volume in one way or another relate to survival in two worlds, the ocean for foraging, and the terrain at its edge or frozen above it for breeding. The evolution of Pinniped behaviour and the mechanisms which underlie it are a consequence of having to cope with two seemingly incompatible sets of environmental constraints. The physiological adaptations for concomitant functioning in two media with very different physical characteristics have produced correlated behav ioural modifications. The energetic demands of reproduction and foraging are idiosyncratic because each activity occurs on opposite sides of the air/water interface. As a result, the mating system must reconcile aquatic design for such functions as locomotion and thermoregulation, with the terrestrial requirements for successful pupping. Similarly, the ecology of this dual habitat prescribes the rules governing the behaviour of the neonate and its interactions with its mother.
