The general approach towards the synthesis of batrachotoxin involves preparation of the AB rings and the DE rings independently, followed by union of these two ring systems, subsequent closure of the C ring, and elaboration to the natural product. A deconjugative alkylation approach has thus far proved to be the most reliable route to the union of the AB rings to the DE rings, however other methods should be explored. Model studies and synthesis of advanced intermediates have provided a good foundation for future work towards the synthesis of batrachotoxin. Part II of this dissertation presents the studies directed toward the total synthesis of Phorboxazole B with an emphasis on efforts for uniting the side chain and macrocyclic core of the natural product by Wittig olefination. Model studies provided detailed insight into the conditions that would be optimal for union of these two advanced intermediates. While these model studies indicated a promising outcome and were even performed on highly elaborate model compounds, these conditions did not translate well in the union of the real side chain and macrocyle of Phorboxazole.

Book excerpt: The total synthesis of a highly potent cytotoxic marine natural product, phorboxazole A, is described. Both a palladium(II)-mediated and a palladium(II)-catalyzed intramolecular alkoxycarbonylation were used as key steps for the preparation of the two cis-2,6-disubstituted tetrahydropyran rings (C22-C26 and C11-C15) of phorboxazole A. The synthesis used a convergent approach involving five subunits, namely to prepare C3-C8 (195), C9-C19 (149), C20-C32 (131), C33-C41 (130) and C42-C46 (129). Assembly of two subunits, 130 and 131, employed Evans coupling protocol and provided 141 in excellent yield. The coupled product 141 was converted to aldehyde 215 in a six step sequence in which Julia-Kocienski olefination using a benzothiazole sulfone 129 was used to install the triene side chain. Allylsilylation of 149 with 195 afforded 202, which was linked by Wittig olefination with 215 to furnish 216. The C5-C9 trans-2,6-disubstituted tetrahydropyran ring was constructed from 216 in a three step sequence. Further transformations from 219 led to phorboxazole A (1) as an off-white solid.

The synthetic strategy started from a homo-Robinson annulation, followed by a ketene-olefin [2+2] cycloaddition to introduce the lactone ring. Then a Grignard-type reaction appended the isopropenyl moiety, and the synthesis of tricholomalides A and B was achieved. During the course of synthesis, the structures of tricholomalides A and B were revised. The second part describes the synthetic studies towards (+)-cortistatin A, especially the A ring functionalization. The C3 nitrogen was introduced by azide displacement, and C2 hydroxyl was built up by Luche reduction. The challenging C1 functionalization was achieved with bromine-induced methoxymethyl deprotection, and some interesting chemistry was found in this system. The synthetic endeavor set a promising stage for the total synthesis of cortistatin A.
