Available in PDF, EPUB and Kindle. Book excerpt: Macrophages are the sentinels of the immune system whose role has evolved beyond providing aseptic conditions to homeostasis, immune regulation, development, and behaviour. These cells have varied ontogenetic origins which reflects in their phenotypic and functional heterogeneity. Macrophage functions are fine-tuned by exogenous and endogenous signals and once tweaked, the information is included in their genetic makeup, albeit not indefinitely. Subversion of the macrophage functions is the hallmark of many pathogenic organisms and modulation of macrophage activity is pivotal to many therapeutic strategies. Fascinating and rapid developments in this field have necessitated the maintenance of currency of knowledge. This book provides a current account of information on varied topics in macrophage biology. Literature surveys have been presented in a captivating and lucid language. The contributing authors have also provided brief accounts of their own research. Every chapter provides a future perspective of what more could be achieved in the context of the current knowledge. The book will be of interest to students and researchers in microbiology, immunobiology, translational research, pathology, and related fields.

Book excerpt: Genetic alterations in cancer, in addition to being the fundamental drivers of tumorigenesis, can give rise to a variety of metabolic adaptations that allow cancer cells to survive and proliferate in diverse tumor microenvironments. This metabolic flexibility is different from normal cellular metabolic processes and leads to heterogeneity in cancer metabolism within the same cancer type or even within the same tumor. In this book, we delve into the complexity and diversity of cancer metabolism, and highlight how understanding the heterogeneity of cancer metabolism is fundamental to the development of effective metabolism-based therapeutic strategies. Deciphering how cancer cells utilize various nutrient resources will enable clinicians and researchers to pair specific chemotherapeutic agents with patients who are most likely to respond with positive outcomes, allowing for more cost-effective and personalized cancer therapeutic strategies.

Book excerpt: This volume – for pharmacologists, systems biologists, philosophers and historians of medicine – points to investigate new avenues in pharmacology research, by providing a full assessment of the premises underlying a radical shift in the pharmacology paradigm. The pharmaceutical industry is currently facing unparalleled challenges in developing innovative drugs. While drug-developing scientists in the 1990s mostly welcomed the transformation into a target-based approach, two decades of experience shows that this model is failing to boost both drug discovery and efficiency. Selected targets were often not druggable and with poor disease linkage, leading to either high toxicity or poor efficacy. Therefore, a profound rethinking of the current paradigm is needed. Advances in systems biology are revealing a phenotypic robustness and a network structure that strongly suggest that exquisitely selective compounds, compared with multitarget drugs, may exhibit lower than desired clinical efficacy. This appreciation of the role of polypharmacology has significant implications for tackling the two major sources of attrition in drug development, efficacy and toxicity. Integrating network biology and polypharmacology holds the promise of expanding the current opportunity space for druggable targets.

Book excerpt: A link between inflammation and cancer has been established many years ago, yet it is only recently that the potential significance of this connection has become apparent. Although several examples of chronic inflammatory conditions, often induced by persistent irritation and/or infection, developing into cancer have been known for some time, there has been a notable resistance to contemplate the possibility that this association may apply in a causative way to other cancers. Examples for such progression from chronic inflammation to cancer are colon carcinoma developing with increased frequency in patients with ulcerative colitis, and the increased incidence of bladder cancer in patients suffering from chronic Schistosoma infection. Inflammation and cancer have been recognized to be linked in another context for many years, i.e., with regards to pathologies resembling chronic lacerations or 'wounds that do not heal.' More recently, the immunology of wound healing has given us clues as to the mechanistic link between inflammation and cancer, in as much as wounds and chronic inflammation turn off local cell-mediated immune responses and switch on growth factor release as well the growth of new blood vessels - angiogenesis. Both of these are features of most types of tumours, which suggest that tumours may require an immunologically shielded milieu and a growth factor-rich environment.

This book was released on 2018-01-29 with total page 326 pages. Available in PDF, EPUB and Kindle. Book excerpt: In the age of digitization our society is transformed into a new state. In particular, machine intelligence dramatically elevates our capability to create and digest information. Naturally, healthcare is also impacted by this trend and will even be more transformed into a informatic driven discipline in the future. In the most important area of histo-pathology, the interpretation of tissue slices from cancer patients, informatics will have an early and huge impact on treatment decisions and probably will act as the leading discipline for this transformation in medicine. Tissue Phenomics provides a comprehensive methodology aiming at the discovery of the most accurate tissue-based decision support algorithm by close integration of assay development, image analysis and bioinformatics, and optimization feedback loops. In this book the methodology of Tissue Phenomics and its benefits and wealth’s are described. The different components of Tissue Phenomics are explained in the different chapters. In the chapters 2 to 4 of this book different authors describe various approaches on how to convert the wealth of tissue slide pixel data into mineable information using knowledge-based and data-driven image analysis methods. Subsequently, the datafication of images and the bioinformatics part plays a crucial role in generating prognostic and predictive models for disease progression. The integration of other data sources such as genomics, radiomics and patient related information is also important and is described as well. As discussed in chapters 5 and 6, these models may classify patients in distinct groups such as those responding to a given therapy. Since Tissue Phenomics provides a huge set of potentially prognostic features (phenes), one focus of both chapters is robust feature selection methods by advanced Monte-Carlo cross validation algorithms. In chapter 7 we discuss multiple application examples of Tissue Phenomics in academic and commercial settings and its tremendous impact to advances in biomedical sciences. Building on the successes in research, chapters 8 and 9 discuss applications in clinical environments and provide a flavor to the future envision in chapter 10, where tissue datafication and subsequent patient profiling is part of every routine examination, with the goal to best match patients with the most successful therapy, as predicted by tissue phenes.

Available in PDF, EPUB and Kindle. Book excerpt: This book is focused on the analysis of the role played by immune cell components in the angiogenic process associated with inflammation and tumor growth. Both innate and adaptive immune cells are involved in the mechanisms of endothelial cell proliferation, migration and activation, through the production and release of a large spectrum of pro-angiogenic mediators. These may create the specific microenvironment that favors an increased rate of tissue vascularization. The link between chronic inflammation and tumorigenesis was first proposed by Rudolf Virchow in 1863 after the observation that infiltrating leukocytes are a hallmark of tumors and first established a causative connection between the lymph reticular infiltrate at sites of chronic inflammation and the development of cancer. Tumors were described as wounds that never heal and surgeons have long described the tendency of tumors to recur in healing resection margin and it has been reported that wound healing environment provides an opportunistic matrix for tumor growth. As angiogenesis is the result of a net balance between the activities exerted by positive and negative regulators, this book will also provide information on some anti-angiogenic properties of immune cells that may be utilized for a potential pharmacological use as anti-angiogenic agents in inflammation as well as in cancer. The work is written for researchers in the field and also for graduate students which approach this matter.
