Studies of the cerebellum are now a central focus in neuroscience. During the last four decades, many laboratories worldwide have dedicated their research activities to understanding the roles of the cerebellum in motor control, cognitive processes and biology of mental processes, behavioral symptoms, and emotion. It is now accepted that the cerebellum acts as a cognitive operator in learning, perception, and attention. Moreover, major improvements in our assessment of in vivo cerebellar architecture using imaging techniques have occurred. A typical example is the accurate description of cerebellar anatomy during fetal development with MRI, a progress which has direct impacts on patient care. These advances have been associated with discoveries of new clinical disorders, in particular in the field of genetic ataxias. More than 20 new genes have been identified these last 10 years. Only for dominant ataxias, more than 30 diseases have now been unravelled. The number of ataxic disorders will increase with aging, the cerebellum being the structure of the brain with the most important loss of neurons with age. More than 300 different cerebellar disorders are encountered during daily practice, but we are missing a single source of information explaining their pathogenesis. Despite the immense amount of knowledge acquired about the cerebellar circuitry these last years, a large book covering the neuroscience of the cerebellum is missing. The goal of this endeavour is to bring up to date information relevant for basic science and also for clinical activities. Book excerpt: In this dissertation, I revisit Kawato's proposal, that the cerebellum predictively modulates descending motor commands, to achieve smooth and coordinated motion, by acquiring an inverse model of the biomechanical plant, and extend it by describing a novel and precise mapping between a cerebellar multizonal microcomplex and a joint's inverse model. This mapping renders two novel predictions: First, inferior olive's oscillations mirror the biomechanical joint's oscillations. Second, deep cerebellar neurons implement a gain factor, set by Purkinje cell inhibition, on inferior olive's signals to mirror the spinal cord's gain. I use biophysical modeling to show that oscillations within the inferior olive match the range of natural frequencies and damping ratios of biophysical joints, and that deep cerebellar neurons enable a multiplicative interaction between the Purkinje and the olivary pathways. Furthermore, I determine the effects of current injection into the inferior olive and the deep cerebellar nuclei and use these results within a control theory model to predict that experimentally disturbing the inferior olive will introduce motor output ringing, while disturbing the deep nuclei will also scale motor output. In both cases, manipulating the inverse model implemented by microzonal microcomplex will unmask the joint's natural dynamics as observed by motor ringing at the joint's natural frequency.

Numerous experimental anatomic and electrophysiologic studies have been undertaken, and much new information has been brought forward. Many apparently discordant observations have been reported however, and on many points the data obtained by the use of different methbds and approaches appear to be diffi cult to reconcile. Much of the interest in the olivocerebellar projection concerns the topographical localization within the projection. Particularly as a result of research in recent years the pattern of localization has turned out to be far more complex than previously be lieved. It was found useful, therefore, to attempt a review of the subject in the hope that a critical analysis of available observations might make it possible to obtain an integrated picture of the olivocerebellar projection and perhaps fmd some basic principles in the organization of this fiber system. As will be seen, our attempt has been only partly successful. There are still riddles that remain to be solved. In the present review attention will be focused on problems related to the locali zation within the olivocerebellar projection, particularly its anatomic aspects. An extensive review of the physiology of the inferior olive has been published recently by Armstrong (1974), who considers some anatomicophysiologic correlations as well. Physiologic fmdings will be referred to here mainly in relation to our main theme.
