Available in PDF, EPUB and Kindle. Book excerpt: When Evita opened on Broadway during the 1979-1980 season, it was (as one of its songs said) "High Flying Adored." But in the 1970-71 season, the producers of Lolita, My Love saw their show (as one of its songs said) "Going, Gone, Gone" after its torturous Philadelphia and Boston tryouts. It didn't even try to brave Broadway, although the bookwriter-lyricist of My Fair Lady had written it. It happens every season. Broadway has one, two, or a few hit musicals, but many, many more flops. Here's a look at the extreme cases from each season of the past half-century. The musicals that everyone knew would be hits - The Sound of Music, The Phantom of the Opera, The Producers - and were. The tuners that sounded terrible from the moment they were announced - Via Galactica, The Civil War, Lestat - and turned out to be even worse than anyone expected. The shows that were destined to succeed - Breakfast at Tiffany's, 1600 Pennsylvania Avenue, Merrily We Roll Along - but didn't. The ones that didn't have a chance - Man of La Mancha, 1776, Grease - but went on to household-name status. Yes, Broadway is the oldest established permanent non-floating crap game in New York, and Peter Filichia takes a look at 100 shows that met either the most glorious or the most ignominious fates.

Available in PDF, EPUB and Kindle. Book excerpt: Fragment-based drug discovery (FBDD) is a new paradigm in drug discovery that utilizes very small molecules - fragments of larger molecules. It is a faster, cheaper, smarter way to do drug discovery, as shown by the number of pharmaceutical companies that have embraced this approach and the biotechnology companies who use fragments as their sole source of drug discovery. Fragment-Based Drug Discovery: A Practical Approach is a guide to the techniques and practice of using fragments in drug screening. The emphasis is on practical guidance, with procedures, case studies, practical tips, and contributions from industry. Topics covered include: an introduction to fragment based drug discovery, why using fragments is a more efficient process than predominant models, and what it means to have a successful FBDD effort. setting up an FBDD project library building and production NMR in fragment screening and follow up application of protein-ligand NOE matching to the rapid evaluation of fragment binding poses target immobilized NMR screening: validation and extension to membrane proteins in situ fragment-based medicinal chemistry: screening by mass spectrometry computational approaches to fragment and substructure discovery and evaluation virtual fragment scanning: current trends, applications and web based tools fragment-based lead discovery using covalent capture methods case study from industry: the identification of high affinity beta-secretase inhibitors using fragment-based lead generation With contributions from industry experts who have successfully set up an industrial fragment-based research program, Fragment-Based Drug Discovery: A Practical Approach offers essential advice to anyone embarking on drug discovery using fragments and those looking for a new approach to screening for drugs.

Available in PDF, EPUB and Kindle. Book excerpt: The book aims to introduce the reader to the emerging field of Evolutionary Systems Biology, which approaches classical systems biology questions within an evolutionary framework. An evolutionary approach might allow understanding the significance of observed diversity, uncover “evolutionary design principles” and extend predictions made in model organisms to others. In addition, evolutionary systems biology can generate new insights into the adaptive landscape by combining molecular systems biology models and evolutionary simulations. This insight can enable the development of more detailed mechanistic evolutionary hypotheses.

Available in PDF, EPUB and Kindle. Book excerpt: When Bill James published his original Historical Baseball Abstract in 1985, he produced an immediate classic, hailed by the Chicago Tribune as the “holy book of baseball.” Now, baseball's beloved “Sultan of Stats” (The Boston Globe) is back with a fully revised and updated edition for the new millennium. Like the original, The New Bill James Historical Baseball Abstract is really several books in one. The Game provides a century's worth of American baseball history, told one decade at a time, with energetic facts and figures about How, Where, and by Whom the game was played. In The Players, you'll find listings of the top 100 players at each position in the major leagues, along with James's signature stats-based ratings method called “Win Shares,” a way of quantifying individual performance and calculating the offensive and defensive contributions of catchers, pitchers, infielders, and outfielders. And there's more: the Reference section covers Win Shares for each season and each player, and even offers a Win Share team comparison. A must-have for baseball fans and historians alike, The New Bill James Historical Baseball Abstract is as essential, entertaining, and enlightening as the sport itself.

Book excerpt: “This fantasy about a drug that gives users a perfect week, then sudden death, is compelling fiction—and nearly a masterpiece.” —The Guardian A new drug is on the street. Everyone’s buzzing about it. Take the hit. Live the most intense week of your life. Then die. It’s the ultimate high at the ultimate price. Adam thinks it over. He’s poor, and doesn’t see that changing. Lizzie, his girlfriend, can’t make up her mind about sleeping with him, so he can’t get laid. His brother Jess is missing. And Manchester is in chaos, controlled by drug dealers and besieged by a group of homegrown terrorists who call themselves the Zealots. Wouldn’t one amazing week be better than this endless, penniless misery? After Adam downs one of the Death pills, he’s about to find out. “A boundary pushing thriller. . . . Amid violent action, existential anguish, and the heightened appreciation for life that death can bring, Burgess has created a premise that readers will find hard to forget.” —Publishers Weekly “Viscerally exciting and emotionally engaging. . . . A clear winner from Burgess.” —Booklist “An exciting, dark story of sex, drugs, and revolution that is sure to grip readers.” —School Library Journal

Book excerpt: The strengths and weaknesses of human memory have fascinated people for hundreds of years, so it is not surprising that memory research has remained one of the most flourishing areas in science. During the last decade, however, a genuine science of memory has emerged, resulting in research and theories that are rich, complex, and far reaching in their implications. Endel Tulving and Fergus Craik, both leaders in memory research, have created this highly accessible guide to their field. In each chapter, eminent researchers provide insights into their particular areas of expertise in memory research. Together, the chapters in this handbook lay out the theories and presents the evidence on which they are based, highlights the important new discoveries, and defines their consequences for professionals and students in psychology, neuroscience, clinical medicine, law, and engineering.
