Book excerpt: Modeling schizophrenia in animals represents a formidable challenge because the most characteristic symptoms of this disorder, auditory hallucinations and delusional beliefs, cannot be directly modeled in animals. Furthermore, limited current understanding of the etiology and pathophysiology underlying the clinical features of this disorder greatly hinders the ability to develop valid models based on those aspects. To circumvent these issues, investigators have attempted to recreate the leading candidate biochemical, neuroanatomical, and genetic abnormalities for this disorder in animals using pharmacological, developmental, neurotoxic, genetic engineering, and other methods. The validity of these animal preparations as animal models for schizophrenia is generally based upon the extent to which they induce measurable behavioral changes that are homologous or analogous to the core clinical features or the information-processing endophenotypes (presymptom phenotypes) that are seen in schizophrenia. The utility of any animal model to serve as a screen for novel treatments is considered related to the extent to which the schizophrenia-relevant behaviors it exhibits can be ameliorated by drugs with known efficacy in schizophrenia but not by drugs known to be therapeutic for brain disorders other than schizophrenia.

From reviewing the rich and diverse literature on vasopressin, it became evident that the rapid advancements in this field made it difficult to syn thesize the information gathered from divergent scientific disciplines into a coherent view of the biological role of vasopressin. We perceived the need for a series of critical reviews delineating this recent progress. Over the past decade, major advances have been made in studies of the anatomy, physiology, phar macology, molecular biology, and behavioral activities of vasopressin. This is, in no small measure, due to the finding that vasopressin can no longer be regarded solely as a neurohypophysial hormone. Our present knowledge is that vasopressin is synthesized in also has an axonal messenger role in the nervous system and of the brain, although the functions of vasopressin in these periph sites outside of vasopressin eral sites are not well understood. In order to prepare an overview concentrating on recent studies in vertebrates, authors were selected based on their expertise and asked to review their research area, including the work from other laboratories. Since that. time experimental approaches have diversified and the research enterprise has attracted the interest of a broad international community of scientists. Many aspects of both thirst and sodium appetite are being studied. The most prominent of these are: 1) phylogenetic and ontogenetic aspects of the phenomena of drink ing behavior, 2) the mechanisms of a variety of dipsogenic and antidipsogenic treatments, both drugs and hormones, 3) the biological controls of drinking and their interaction with the regulation of blood volume and blood pressure, 4) the peripheral signals of drinking including the role of the baro- and volume-receptors, 5) the receptor systems within the brain and the neuroanatomical circuitry for thirst and sodium appetite, and 6) the possible roles of brain sodium and of the hormones of sodium conservation ln the arousal of sodium appetite.
