Book excerpt: Glycoconjugate Research, Volume I contains the proceedings of the Fourth International Symposium on Glycoconjugates, held in Woods Hole, Massachusetts, in September 1977. Contributors focus on the state of knowledge in the field of glycoconjugates ranging from polysaccharides and glycoproteins to glycolipids, proteoglycans, and all their varieties in plants, animals, and microorganisms. This text is organized into three sections and consists of 87 chapters. After an introductory chapter where the use of glycosidases for the structural analysis of complex carbohydrates is described, the discussion turns to other methods used for elucidating the structure of complex carbohydrates. The chapters that follow focus on the primary structure and conformation of glycans that are N-glycosically linked to peptide chains; isolation of monofuco-disialoganglioside from human and bovine brains; and partial chemical characterization of activated peptides isolated from a single cell suspension of rat colonic secretory cells. The last two sections explore cartilage proteoglycans and the structure-function relationships of glycoconjugates, touching on topics such as link proteins; the biochemical aspects of malignancy in human chondrosarcomas; defensive response of plants to a microbial oligosaccharide with a pheromone-like activity; and the interaction of saccharides with ricin. This book is intended for students and practitioners of chemistry and biochemistry; biology and microbiology; and physiology.

Book excerpt: A new focus on glycoscience, a field that explores the structures and functions of sugars, promises great advances in areas as diverse as medicine, energy generation, and materials science, this report finds. Glycans-also known as carbohydrates, saccharides, or simply as sugars-play central roles in many biological processes and have properties useful in an array of applications. However, glycans have received little attention from the research community due to a lack of tools to probe their often complex structures and properties. Transforming Glycoscience: A Roadmap for the Future presents a roadmap for transforming glycoscience from a field dominated by specialists to a widely studied and integrated discipline, which could lead to a more complete understanding of glycans and help solve key challenges in diverse fields.

Book excerpt: This volume provides a comprehensive understanding of the enigmatic identity of the glycome, a complex but important area of research that has been largely ignored due to its complexity. The authors thoroughly deal with almost all aspects of the glycome, i.e., elucidation of the glycan identity enigma and its role in regulation of the cellular process, and in disease etiology. The book bridges the knowledge gap in understanding the glycome, from being a cell signature to its applications in disease etiology. In addition, it details many of the major insights regarding the possible role of the glycome in various diseases as a therapeutic marker. The book systematically covers the major aspects of the glycome, including the significance of substituting the diverse monosaccharide units to glycoproteins, the role of glycans in disease pathologies, and the challenges and advances in glycobiology. The authors stress the significance and huge encoding power of carbohydrates as well as provide helpful insights in framing the bigger picture. The Glycome: Understanding the Diversity and Complexity of Glycobiology details state-of-the-art developments and emerging challenges of glycome biology, which are going to be key areas of future research, not only in the glycobiology field but also in pharmaceutics.

Proteins, nucleic acids, and polysaccharides are macromolecule--polymers formed from families of simpler subunits. Because of their size and complexity, the polymers are capable of both inter- and intramolecular interactions. These interactions confer upon the polymers distinctive three-dimensional shapes. These tertiary configurations, in turn, determine the function of the macromolecule. Computers have become so inextricably involved in empirical studies of three-dimensional macromolecular structure that mathematical modeling, or theory, and experimental approaches are interrelated aspects of a single enterprise.

Book excerpt: In the past decade, there has been an explosion of progress in understanding the roles of carbohydrates in biological systems. This explosive progress was made with the efforts in determining the roles of carbohydrates in immunology, neurobiology and many other disciplines, examining each unique system and employing new technology. This volume represents the first of three in the Methods in Enzymology series, including Glycomics (vol. 416) and Functional Glycomics (vol. 417), dedicated to disseminating information on methods in determining the biological roles of carbohydrates. These books are designed to provide an introduction of new methods to a large variety of readers who would like to participate in and contribute to the advancement of glycobiology. The methods covered include structural analysis of carbohydrates, biological and chemical synthesis of carbohydrates, expression and determination of ligands for carbohydrate-binding proteins, gene expression profiling including micro array, and generation of gene knockout mice and their phenotype analyses.

Available in PDF, EPUB and Kindle. Book excerpt: Although carbohydrates are most commonly associated with metabolism, these molecules are involved in a variety of other critical processes that range from cellular trafficking to transcriptional regulation. Alterations in carbohydrate sequence are linked to a host of diseases ranging from cancer to dementia, yet progress in structural glycobiology has been slow compared to that of proteins and DNA. The following work pushes the boundary of our understanding of carbohydrate interactions at the atomic level. First, a 3D adaptation of the Symbol Nomenclature for Glycans (SNFG) was developed as a cartoon representation for carbohydrates to improve the communication of complex structural features to both glycobiologists and non-specialists. Second, the prediction of protein-carbohydrate complexes was improved by including a scoring term within docking programs that accounts for the linkage between glycan residues. Third, the crystal structure of a silayltransferase is presented with a docked model for the sugar donor. Fourth, a significant binding preference was identified between an enzyme within the heparan sulfate biosynthetic pathway and downstream products, with implications for protein localization in vivo. Lastly, a study of the intra-molecular interactions of a glycoprotein suggests a regulatory mechanism in which glycosylation affects intrinsic protein dynamics. Collectively, these efforts contribute to our understanding of the glycan structure-function relationship and facilitate future efforts in glycobiology.

Book excerpt: By guiding the reader toward answering basic questions on the structural analysis of glycoconjugates using conventional techniques, this book studies methods such as chemical cleavage, molar composition, use of enzymes, use of lectins and typical fractionation procedures such as HPLC, GLC, TLC, and other chromatographic systems. More sophisticated techniques such as NMR or MS are described to give the reader the essential information needed to adapt procedures to his/her requirements. This book will serve as a practical manual designed for scientists not familiar with glycoconjugate methodologies, but whose research work requires contact with analytical problems or metabolic investigations in glycobiology. It is the result of twenty years of experience of summer schools held in a laboratory environment and will acquaint the reader with widely used techniques in the area rather than cataloguing an exhaustive list of methods. This will help any biochemist to develop an experiment with ease.
