This method of elastic scattering is used in various branches of science and technology, includ ing condensed matter physics, molecular biology and biophysics, polymer science, and metallurgy. Many small-angle scattering studies are of value for pure science and practical applications. It is well known that the most general and informative method for investigating the spatial structure of matter is based on wave-diffraction phenomena. In diffraction experiments a primary beam of radiation influences a studied object, and the scattering pattern is analyzed. In principle, this analysis allows one to obtain information on the structure of a substance with a spatial resolution determined by the wavelength of the radiation. Diffraction methods are used for studying matter on all scales, from elementary particles to macro-objects. The use of X rays, neutrons, and electron beams, with wavelengths of about 1 A, permits the study of the condensed state of matter, solids and liquids, down to atomic resolution. Determination of the atomic structure of crystals, i.e., the arrangement of atoms in a unit cell, is an important example of this line of investigation.

Among these are fluorescent core-shell silica nanoparticles synthesized in water known as Cornell prime dots (C' dots), which are composed of a silica core and a poly(ethylene glycol) (PEG) shell. These C' dots are poised to become promising diagnostic and therapeutic tools in medicine, particularly for cancer applications. While spherical C' dots have already been in FDA-approved human clinical trials, exploration has expanded into different topologies of silica nanoparticles such as rings and cages, providing higher aspect ratios and higher surface area to volume ratios. For clinical translation of such nanostructures, quantitative assessments of their size and size dispersity remain parameters for further investigation and are critical to understanding and evaluating their effectiveness and safety in nanomedicine applications. Here, solution small-angle X-ray scattering was employed to characterize 3D structural details of ring-type silica nanostructures and their size and size dispersity. A hollow cylinder model provided consistent structural and dispersity parameters that were supported by transmission electron microscopy investigations.

However, the synthesis and characterization of alloy nanoparticles are still posing significant challenges, leading to an increasing demand for in situ characterization techniques. Small-angle X-ray scattering (SAXS) is a powerful method for structural analysis of nanoparticles. As the SAXS signal is essentially the Fourier transform of the electron distribution, it provides structural information for the entire ensemble of nanoparticles. The development of SAXS has been facilitated by significant advances in synchrotron X-ray sources and data processing methods, leading to the development of the 3D-SAXS method, which enables the reconstruction of the 3D structures of particles from SAXS profiles.Although SAXS has the potential to be a powerful tool for investigating the internal structures of alloy nanoparticles, its application is hindered by the challenges posed by polydispersity, which can cause smearing effects that complicate the geometry recovery process. This dissertation presents a novel approach to overcome the problem of polydispersity in SAXS data analysis, thus demonstrating the utility of SAXS in investigating the internal electron density distributions of alloy nanoparticles. In Chapter 2, the SharPy algorithm is introduced as a size-refocusing program that reduces the smearing effect caused by polydispersity in SAXS data. SharPy is based on a penalized iterative regression approach to fit the pair distance distribution function (PDDF) with an estimated size distribution. It can provide detailed information about the shape of nanoparticles from the smeared SAXS signal under various scenarios and conditions. Chapter 3 investigates the simulated SAXS profiles of AuAg core-shell nanoparticles with varying size distribution, core-shell ratio, and degrees of alloying. It demonstrates the capability of SAXS in observing the electron density distribution of AuAg core-shell structures. These findings provide insights into the potential of SAXS as a reliable method for investigating the internal structures of alloy nanoparticles. Chapter 4 focuses on synthesizing and characterizing AuAg nanoparticles. Their SAXS profiles and PDDF analysis demonstrate that SAXS can distinguish between homogeneous and core-shell nanoparticle structures. In this chapter, the SharPy algorithm is first-time applied to real experimental data, demonstrating its ability to reveal the core-shell structure from a polydisperse nanoparticle system. Chapter 5 investigates the evolution of alloying AuAg nanoparticles through a combination of SAXS/PDDF analysis, 3D reconstruction, and molecular dynamics (MD) simulation. The study presents the 3D electron density distribution of alloying AuAg nanoparticles. The 3D reconstruction with electron density mapping provides a straightforward visualization of the electron density distribution pattern of the alloying AuAg nanoparticles. The success of the SAXS experiment lies in the development of the 3D-SAXS pipeline, which involves the use of SharPy and 3D reconstruction programs, making 3D SAXS a promising alternative to electron microscopy for visualizing the morphology of nanoparticle systems.

The goal is to form materials with improved properties than their classical ancestors with just a fractional amount of raw material. Another key feature of nanoparticles is their different, and sometimes unexpected, behavior concerning reactivity, compared with their bulk materials. Because of this, nanoparticles have a wide range of applications, especially in the field of catalysis. Here, characteristics of nanoparticles - more edges, corners, defects or oxygen vacancies are used to obtain a high performance of the catalysts. Nanoscaled particles also exhibit larger surface area and higher metal dispersion, which further contributes to the catalytic possibilities. To gain such particles, two different pathways are given: first, there is the so-called top down pathway, considered as further developments of micro technology, where physical preparation methods like lithography are used. The second way is the bottom up method where self-assembling systems, formed by surfactants, are used. Concerning gold nanoparticles, it is reported that the use of C16TAB at specified conditions, gives gold nanorods with a sharp size-distribution because the direction of growth is predetermined. Being a cationic surfactant, C16TAB affects the electrochemical potentials and introduces bromide-ions as an additional species to the reaction. To achieve gold nanoparticles from aqueous HAuCl4-solutions, the above-mentioned method needs a separate reducing agent such as ascorbic acid (Asc0), NaBH4 or N2H4. A way of synthesizing spherical gold nanoparticles is the use of Nd:YAG laser with a salt induced agglomeration. By modifying the formulation of the salt solution, different sizes are obtained. This way of synthesis, a combination of physical top-down and self-assembling bottom-up processes, can be modified by adding surfactants, like PEG, to optimize size distribution and physical characteristics, like UV-Vis absorption. This method is an elegant way of synthesis; however, problems may occur by functionalizing the particles, because of a high salt content. Here, a high influence of purity, concentration and composition to the size and shape of gold nanoparticles might be given. In this book, following the presentation of the basics of scattering from isotropic macromolecular solutions, modern instrumentation, experimental practice and advanced analysis techniques are explained. Advantages of X-rays (rapid data collection, small sample volumes) and of neutrons (contrast variation by hydrogen/deuterium exchange) are specifically highlighted. Examples of applications of the technique to different macromolecular systems are considered with specific emphasis on the synergistic use of SAXS/SANS with other structural, biophysical and computational techniques.
