Hence, this study aims to determine the transcripts/exons that are regulated by stress hormones in the hippocampus. RESULTS: Hypophysectomy was confirmed by weight reduction in experimental animals. Serum corticosterone was also reduced upon hypophysectomy which was rescued by administration of CORT or ACTH. Using RNA-Seq, we identified transcripts/exons that are changed upon hypophysectomy and successfully rescued by CORT or ACTH. Specifically, edgeR and DEXSeq identified 32 genes with >2 fold changes and 889 exons with >1.5 fold changes respectively that were changed upon hypophysectomy and rescued by CORT or ACTH administration. RT-PCR validated the regulation of several target exons by stress hormones. We further identified a decline and an increase in the transcripts containing the target exon of the Sept8 gene in a time-course dependent manner upon hypophysectomy and in normal groups respectively. CONCLUSION: These findings demonstrate a specific exon that is regulated by stress hormones in the hippocampus.

A key aspect of genome complexity can be attributed to the process of alternative splicing of precursor mRNA (or pre-mRNA), during which introns are removed and exons are selectively spliced together. This highly regulated process generates different mature mRNA transcripts from a single gene and is widespread throughout the eukaryotic evolution. The majority of the genes expressed in the mammalian central nervous system undergo extensive alternative splicing. Some genes can generate more than a thousand isoforms resulting in diverse proteoforms that can differ in their function, binding preference, catalytic activity, and localization. Perturbation in alternative splicing has been linked to many neurological disorders. Despite its significance, we still lack a comprehensive understanding of its role in neuronal function. Recent studies suggest that alternative splicing is a regulator of tissue identity and development. This is particularly relevant to the brain where developmental regulation and heterogenous cell population play a crucial role in shaping its function. For a long time, technological limitations in detecting and measuring alternatively spliced transcripts hindered the progress in understanding the biology of alternative splicing. Recent advances in long-read and native mRNA sequencing and robust computational tools allow us to capture and quantify in their full-length form. Public transcriptomics datasets can be utilized for the meta-analysis of alternative splicing landscapes of various experimental conditions and disease samples. In addition, protein isoform detection complements mRNA isoform levels. Given its biological significance and recent advances, the objective of this Research Topic is to assemble current knowledge, views, and challenges in the studies of alternative splicing in brain function.

Book excerpt: The skin, the body’s largest organ, is strategically located at the interface with the external environment where it detects, integrates and responds to a diverse range of stressors, including solar radiation. It has already been established that the skin is an important peripheral neuroendocrine-immune organ that is closely networked with central regulatory systems. These capabilities contribute to the maintenance of peripheral homeostasis. Specifically, epidermal and dermal cells produce and respond to classical stress neurotransmitters, neuropeptides and hormones, production which is stimulated by ultraviolet radiation (UVR), biological factors (infectious and non-infectious) and other physical and chemical agents. Examples of local biologically active products are cytokines, biogenic amines (catecholamines, histamine, serotonin and N-acetyl-serotonin), melatonin, acetylocholine, neuropeptides including pituitary (proopiomelanocortin-derived ACTH, b-endorphin or MSH peptides, thyroid stimulating hormone) and hypothalamic (corticotropin-releasing factor and related urocortins, thyroid-releasing hormone) hormones, as well as enkephalins and dynorphins, thyroid hormones, steroids (glucocorticoids, mineralocorticoids, sex hormones, 7-δ steroids), secosteroids, opioids and endocannabinoids. The production of these molecules is hierarchical, organized along the algorithms of classical neuroendocrine axes such as the hypothalamic pituitary adrenal axis (HPA), hypothalamic-thyroid axis (HPT), serotoninergic, melatoninergic, catecholaminergic, cholinergic, steroid/secosteroidogenic, opioid and endocannabinoid systems. Disruptions of these axes or of communication between them may lead to skin and/or systemic diseases. These local neuroendocrine networks also serve to limit the effect of noxious environmental agents to preserve local and consequently global homeostasis. Moreover, the skin-derived factors/systems can also activate cutaneous nerve endings to alert the brain to changes in the epidermal or dermal environments, or alternatively to activate other coordinating centers by direct (spinal cord) neurotransmission without brain involvement. Furthermore, rapid and reciprocal communications between epidermal and dermal and adnexal compartments are also mediated by neurotransmission including antidromic modes of conduction. Lastly, skin cells and the skin as an organ coordinate and/or regulate not only peripheral but also global homeostasis.

In humans, drug and alcohol use disorders (i.e., abuse and dependence) are defined by clusters of behavioral symptoms that can be modeled to various degrees in animals. Hallmark behavioral symptoms associated with drug and alcohol dependence are compulsive drug use, loss of control during episodes of drug use, the emergence of a negative emotional state in the absence of the drug, and chronic relapse vulnerability during drug abstinence. The transition to drug dependence is defined by neuroadaptations in brain circuits that, in the absence of drugs, mediate a variety of critical behavioral and physiological processes including natural reward, positive and negative emotional states, nociception, and feeding. Chronic drug exposure during the transition to dependence spurs (1) within-systems changes in neural circuits that contribute to the acute rewarding effects of the drug and (2) recruitment of brain stress systems (neuroendocrine and extra-hypothalamic). There are substantial genetic contributions to the propensity to use and abuse drugs, and drug abuse is highly co-morbid with various other psychiatric conditions (e.g., anxiety disorders, major depressive disorder) that may precede or follow the development of drug use problems. Across drugs of abuse, there are overlapping and dissociable aspects of the behavioral and neural changes that define the transition to dependence. Even within a single drug, people abuse drugs for a variety of reasons. The picture is further complicated by the fact that humans often abuse more than one drug concurrently. Even in the face of these challenges, pre-clinical and clinical research is making exponential gains into understanding the neurobiology of drug addiction. With the advent of new technologies and their combination with traditional approaches, the field is able to ask and answer addiction-related research questions in increasingly sophisticated ways. Here, we hope to assemble a collection of articles that provide an up-to-the-moment snapshot of the prevailing empirical, theoretical and technical directions in the addiction research field. We encourage submissions from all investigators working to understand the neurobiology of addiction, especially as it pertains to reward and stress pathways in the brain.
