Solid-phase micro-extraction (SPME) methods have been developed as a chemical analysis tool offering fast, simple, non-invasive sampling without the use of solvents. In this study SPME devices were tested for applicability for in-planta detection of chlorinated solvents. To evaluate the use of SPME for VOCs in-planta a number of integrated studies were undertaken. Uptake profiles were developed for nine chlorinated solvents in a liquid polydimethylsiloxane (PDMS) matrix with 100 [mu]m Carboxene SPME fibers. Timeweighted average (TWA) sampling was conducted by exposing the SPME fiber to the chemical mixtures using three retraction lengths. Linear uptake profiles were demonstrated for 25 of the 27 of the sampling conditions. A storage experiment was conducted to determine sample retention on the SPME fiber for transport prior to analysis. It was demonstrated that all chemicals except dichloromethane are retained on the fiber for up to 24 hours. Field sampling with SPME devices was conducted at a known chlorinated solvent contaminated site using a newly designed in-planta sampler. The exceptional sensitivity of the SPME platform, combined with its adaptability to traditional GC-MS systems and its ability to extract samples with minimal work-up, make it appropriate to applications in forensic laboratories. In a series of research projects, solid phase micro-extraction was employed for the analysis of explosives, commercial tire treatments, and entomological specimens. In the first project, the volatile organic compounds emanating from two brands of pseudo-explosive training aids for use in detector dog imprinting were determined by SPME-GC-MS, and the efficacy of these training materials was tested in live canine trials. In the second project, the headspace above various plasticizers was analyzed comparative to that of Composition C-4 in order to draw conclusions about the odor compound, 2- ethyl-1-hexnaol, with an eye toward the design of future training aids. In the third, automobile tires which had participated in professional race events were analyzed for the presence of illicit tire treatments, and in the fourth, a novel SPME-GC-MS method was developed for the analysis of blowfly (Diptera) liquid extracts. In the fifth and final project, the new method was put to the task of performing a chemotaxonomic analysis on pupa specimens, seeking to chemically characterize them according to their age, generation, and species.

Recently, the sample preparation in the analysis of aqueous samples has been achieved by solid phase microextraction (SPME). This method has been shown to be fast, inexpensive, solventless, portable and automatable. SPME has several advantages over conventional liquid-liquid extraction. SPME has been shown to be a quantitative technique for volatile and semivolatile compounds from gaseous and liquid samples. The quantization by SPME is also linear over four orders of magnitude. As this method can reach a detection limit of 15 ppt (parts per trillion) for several compounds and can be easily automated, the analysis of volatile organic compounds (VOCs) in the environment, in particular, water samples, is highly advantageous by this method. This study will describe the analysis of volatile organics, BTEX, and halogenated organics in water. The lowest limit of detection, linearity, and other parameters will be discussed. The analysis of a soil sample taken near an oil storage tank for VOCs by SPME will be described. A comparison between different fibers (different types of coating materials and film thickness) using different columns will be described in this work.

Book excerpt: A prototype rapid, high volume air sampling device based on Solid Phase Microextraction (SPME) has been developed for the collection of trace level volatile organic compounds (VOCs). The High Surface Area-Solid Phase Microextraction (HSASPME) device contains ten times more polymer than traditional SPME fibers and is uniquely designed to optimize compound uptake at higher flow rates. This study evaluated the extraction efficiency at six air sampling flow rates ranging from 0.1 L/min to 10 L/min and compared total compound extraction at the two extreme flow rates. A 10 ppb(v) concentration of 39 volatile organic compounds was used. Carboxen/Poly(dimethylsiloxane) and Poly(dimethylsiloxane) polymer coatings were evaluated using an Agilent 6890N/5973, a resistively heated Low Thermal Mass Gas Chromatograph column and an Entech 7100 Preconcentrator. Larger extraction efficiencies were observed at lower flow rates, but the higher flow rates proved superior in total compound extraction per unit time. Across the range of compounds, the HSASPME device achieved an average 8-fold increase in compound uptake at a flow rate of 10 L/min as compared to 0.1 L/min.
