Book excerpt: Burley tobacco (Nicotiana tabacum L.) has historically been fertilized with high nitrogen rates. This study was conducted to evaluate the effect of nitrogen rate on yield and total tobacco specific nitrosamine (TSNA) concentration. The experiment also evaluated the use of in season nitrogen monitoring techniques and how well they were able to predict yield and total TSNA concentration. In 2007, five preplant nitrogen treatments along with two sidedress nitrogen treatments were used. These included: 0, 67, 135, 202, 270 kg/ha nitrogen preplant and 0 and 67 kg/ha nitrogen sidedress. In 2008, there were only four preplant nitrogen rates with the two sidedress nitrogen rates. The 270 kg N/ha was removed for 2008. Also in 2008, four varieties were used and they were KT 204LC, TN 90LC, TN 90HC, and NC 7. Sidedress treatments were applied at three weeks after transplanting. The in season nitrogen monitoring techniques were the pre-sidedress nitrate test (PSNT), the Cardy nitrate meter, and the CM 1000 chlorophyll meter. PSNT samples were taken before sidedress treatments were applied. Chlorophyll and Cardy meter samples were taken at sidedress, topping, and harvest in 2007. A five weeks after transplant sampling date was added in 2008. Yields were optimized at 135 kg/ha total nitrogen rates. A quadratic plateau model was used to determined critical levels for PSNT and total nitrogen predicting yield and total TSNA. Relative yield plateaued at 0.953 with 204.8 kg/ha total nitrogen applied. A critical PSNT level of 46.4 ppm soil nitrate was established for burley tobacco. TSNA levels increased up to 135 kg N/ha and reached a plateau at 257.8 kg/ha total nitrogen. Total nitrogen applied was the best yield predictor out of the methods used to evaluate nitrogen status of burley tobacco. However analysis of PSNT for different soil types might prove to be a more reliable nitrogen management tool. Cardy nitrate measurements did differ over the nitrogen rates, but there was no relationship between yield or TSNA and petiole nitrate levels during the growing season. Chlorophyll measurements did not correlate to nitrogen rates or yield.

Book excerpt: Our objectives in this study were to evaluate the state recommendations for nitrogen, determine the effects of nitrogen on tobacco specific nitrosamines (TSNA), yield and quality, determine efficient nitrogen rates and evaluate the pre sidedress nitrate test (PSNT) as an indicator of nitrogen needs in burley tobacco. This study had nine nitrogen rates, in a split plot design with four replications. Main plots consisted of preplant nitrogen: 89.6, 179.2 and 268.8 kg/ha. Sub plots consisted of sidedress nitrogen: 0, 52 and 112 kg/ha. Soil nitrate-N was determined using the PSNT, with samples taken to a depth of 0.15 m three weeks after transplanting. There were two locations in 2004, at Greeneville, TN (GR) and Glade Spring, VA (GL). Two more locations were added in 2005 and 2006, Springfield, TN (HR) and Lexington, KY (LE) along with a tobacco after tobacco (TAT) rotational study at GR and HR also in 2006. Nitrogen rates within the state recommendations gave yields statistically equal to the maximum. In several cases, yield was maximized by lower than recommended rates. The study showed an increase in yield with an increase in nitrogen above the lowest treatment. All yields, except for HR 2005, exceeded the long term, Tennessee state burley yield average. A positive yield response to sidedress nitrogen fertilizer was observed at the 89.6 kg/ha treatment, with 56 and 112 kg/ha sidedressed and the 179.2 kg/ha treatment with 56 kg/ha sidedressed. There was only one instance with 268.8 kg/ha pre plant nitrogen where there was a positive yield response, likely due to variability within the study. Total TSNA content was also increased with higher nitrogen rates. TSNA content was lower in the dryer 2005 curing season. Although not an objective of this study, and not proven, it is likely that the locations of the curing structures had some effect on TSNA content. PSNT values increased as nitrogen rates increased. From this study the PSNT does not seem to be a precise enough indicator of nitrogen needs in burley tobacco to be used in making N application decisions.

Book excerpt: Emissions were affected by termination timing x day of sampling (DOY) and residue treatment x DOY, but no strong patterns were observed. Cumulative N2O emissions were highly correlated with aboveground residue C and C/N and root C/N. Daylight hours and corn growing degree days were significant predictors of N2O emissions. Interactions between soil temperature, NH4+, and corn growing degree days only explained ~ 10% of the variation in N2O emissions. Assessment of complete denitrification or 15N tracing experiments should be explored in the context of perennial termination to identify strategies to limit N2O emissions. In Chapter 6 I used a novel approach to integrate soil health and soil metagenomes to assess differences in soil health and microbial N metabolisms between uncultivated soils and cropped soils managed with contrasting practices. Soil samples were collected from dairy farms and included samples from forest or hedgerows next to cropping soils managed with either no-till and conventional amendments or tillage and organic amendments. Soil health was assessed using chemical, physical, and biological indicators using two sets of soil health tests. Soil microbially-mediated N transformations were quantified through whole-shotgun metagenomics. Soil health, particularly related to organic C and N, were highest in uncultivated soils followed by cropped soils from no-till farms while cropped soils from tilled-organic farms had the poorest soil health. Microbial genes for organic N use were correlated with soil quality indices and were highest in uncultivated soils, while inorganic N use genes were negatively correlated with ACE protein and were highest in the cropped soils. Dairy farm soil management employing no-till increased soil health and microbial organic N metabolisms relative to tilled-organic soils. Organic N metabolisms like urea production and consumption may be appropriate biological indicators of soil health. Further exploration across a wider range of cropping systems would be necessary to fully assess the validity in urea cycling in delineating soil health.
