Book excerpt: Tomato spotted wilt virus (TSWV) is an emerging pathogen with wide host range and one of the most important viruses of plants. Information regarding processing of negative single stranded RNA viruses such as TSWV in the RNA silencing pathway remains limited. In nature TSWV is only transmitted by thrips as vectors and since infection occurs in both thrips and plants, an experimental system to transmit using thrips and the detection of TSWV were established. In order to understand the processing of TSWV in the RNA silencing pathway, Arabidopsis thaliana as a model plant was used in the genetic analysis against TSWV. Core components of the RNA silencing machinery tested were Dicer-like proteins(DCLs), RNA dependent RNA Polymerases (RDRs), and Argonaute proteins (AGOs). Results suggest DCL4 and DCL2 proteins are involved in the recognition and processing of TSWV. RDR1 is needed for the amplification of siRNAs derived from TSWV. AGOs protein levels remain unaffected after TSWV inoculation compared to mock inoculated plants suggesting that degradation by TSWV does not occur. It is well known that plant viruses encode suppressors of RNA silencing to facilitate plant infection and TSWV encodes the Non-Structural-small protein (NSs) whose mechanism of suppression has not been elucidated. NSs protein from TSWV was cloned from viral RNA and tagged with a 6HIS-3xFLAG tag at the C terminus for detection (NSs-HF). In transient assays activity of NSs as a suppressor of RNA silencing was corroborated by preventing silencing of green fluorescent protein (GFP) at three days post infiltration (dpi). Infectious clones of TSWV are not available and to determine the mechanism of suppression, NSs-HF was introduced in a previously described inactive model virus Turnip mosaic virus (TuMV-GFP-AS9) which has an inactive suppressor a does not infect wild type plants. The rescue of pathogenicity of this virus was observed after GFP detection under UV light and the presence of the NSs protein by western blotting. These experiments establish a foundation to study TSWV within the RNA silencing pathway and are the basis for future experiments to understand the mechanism of suppression of RNA silencing by TSWV.

Book excerpt: Tomato spotted wilt virus (TSWV) is the type species of the genus Tospovirus in the family Bunyaviridae and is exclusively transmitted by thrips in nature. To determine the viral RNA segment(s) responsible for thrips transmissibility, a viral genetic system that included reassortment and segment-specific RFLP analysis was developed. A complete set of reassortants between TSWV-RG2, a thrips transmissible isolate, and TSWV-D, a thrips nontransmissible isolate was generated. Thrips transmission assays of the reassortants showed that reassortants containing the M RNA from TSWV-D, a thrips nontransmissible isolate, were not transmissible, whereas reassortants with the M RNA from TSWV-RG2, a thrips transmissible isolate, were transmissible, irrespective of the origin of the L and S RNA. This is compelling evidence that thrips transmissibility of TSWV resides on the M RNA. Initial attempts to delineate specific regions of the M RNA associated with thrips transmission and characterize genetic changes on the M RNA of the TSWV following repeated mechanical or thrips transmission were made by comparing the nucleotide sequence of the M RNAs of the 3 poorly-transmissible TSWV isolates and transmissibility-restored isolates. After 24 transmission cycles by mechanical means or by thrips, whole M RNA sequences were determined by RT-PCR and sequencing. The mean mutation frequency of the NSm open reading frame (ORF) for the three isolates per passage was 9.2 X 10−5 nucleotide substitutions per base per mechanical or thrips passage. A similar pattern was observed in GN/GC ORF with 1.1 X 10−4 and 1.3 X 10−4 nucleotide substitutions per base per mechanical or thrips passage, respectively. Interestingly, two NS mutations and one synonymous S mutation in the G[subscript N]/G[subscript C] ORF were conserved in the three isolates after the series of mechanical transmission. Two NS mutations consist of a substitution of C86 to U (C86U) changing amino acid.

Book excerpt: Tomato spotted wilt virus (TSWV) is an important agronomic and horticultural pest in NC and other parts of the world. Many have speculated that management of weedy species that host the virus may aid in decreasing TSWV occurrence on a farm. In May of 2003 and 2004, weedy species were sampled around six fields, two fields per replication, at the Center for Environmental Farming Systems (CEFS) in Goldsboro, NC, using a 15 m x 15 m grid to map the overwintering pattern of TSWV on the farm. Weeds were tested for TSWV infection and weed density ratings were assigned in each grid cell. Indicator plants (tomato and peanut) were used to map TSWV occurrence within the fields. Thrips movement was monitored with sticky traps to reveal spatial patterns of movement across the farm and, along with indictor plants, were evaluated biweekly through the summers of 2003 and 2004 to monitor temporal movement/occurrence. Plots were located at field edges and in field centers to detect the differences between trivial movement of infected thrips and movement up to 60 m from bordering weedy hosts. TSWV occurrence was equally likely to be found in the center versus the edge plots, revealing that complete eradication of host species up to 60 m from a field may not aid in decreasing TSWV in a field. TSWV occurrence in and around the three replications that were spread across the farm revealed an effect of replication in 2003 when more virus was found in the southern-most replication. Ranunculus sardous (hairy buttercup) tested positive most often compared to all other weed species tested and was also found in the highest density surrounding the southern-most replication.

Book excerpt: Tomato spotted wilt virus (TSWV) is an economically important plant virus dependent on insects (thrips) for transmission to plant hosts. Like many animal-infecting viruses, TSWV replicates in the cells of its insect vector. The virus is an emergent disease threatening food and fiber crops worldwide. The aim of this work was to develop novel control strategies against TSWV through a better understanding of the virus-vector interaction. Previously, the TSWV GN protein was shown to be the viral attachment protein, a molecule mediating attachment of virus particles to the midgut epithelial cells of vector thrips. The specific goals of my research were to further examine the utility of disrupting the virus-vector interaction for effective virus control by exploiting GN properties, and to track the route of TSWV in thrips using confocal microscopy. To achieve these goals, I expressed soluble and insoluble forms of GN fused to green fluorescent protein (GFP) transiently and transgenically and examined their cellular localization in planta. GN::GFP recombinant protein localized to Golgi stacks throughout the cells as indicated by a punctate pattern or co-localization to a Golgi marker. In contrast, the soluble form of GN, GN-S::GFP, localized to the ER and apparently also to the cytoplasm. Virus acquisition and transmission assays with GN-S::GFP transgenic tomato plants demonstrated that transmission of TSWV by F. occidentalis was reduced by 35 to 100%. These results indicated that transgenic expression of GN-S in tomato plants may have the potential to prevent secondary spread of the virus. Novel features of the morphology of principal (PSGs) and tubular salivary glands (TSGs) of the insect vector F. occidentalis and of their infection with TSWV were described. The virus colonized different cell types and regions within the PSGs with variable intensity and distribution; and accumulated at the lumen of individual cells. The TSGs of F. occidentalis are proposed as a route for TSWV infection into the PSGs. The transgenic plants and the new knowledge of the virus vector interaction are promising tools to control TSWV and a model approach for the control of other vector-borne viruses.
