Book excerpt: In MCF7 and T47D cells cotreated with 1 nM 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) plus 0.1-10 uM 3̕,4̕ -dimethoxy flavone (DMF), there was a concentration-dependent decrease in the TCDD-induced ethoxyresorufin O-deethylase (EROD) activity. Gel mobility shift assays showed that 3̕,4̕ -DMF inhibited TCDD-induced aryl hydrocarbon receptor (AhR) transformation in rat liver cytosol and blocked TCDD-induced formation of the nuclear AhR complex in MCF7 and T47D cells. The antiestrogenic activity of TCDD in estrogen-induced transactivation assays in MCF7 cells was reversed by 3̕,4̕ -DMF, confirming the AhR antagonist activity of this compound in breast cancer cells. Cotreatment of T47D and MCF7 cells with TCDD and 10 uM resveratrol inhibited induction of CYP1A1 mRNA and EROD activity. Resveratrol did not inhibit TCDD-induced AhR transformation and reporter gene activity. Actinomycin D chase experiments in T47D cells showed that the mechanism of inhibition of CYP1A1 mRNA and EROD activity is due to an increased rate of CYP1A1 mRNA degradation, suggesting that resveratrol inhibits CYP1A1 via an AhR-independent post-transcriptional pathway. Vitamin D receptor-interacting protein 150 (DRIP150) coactivated estrogen receptor [alpha] (ER [alpha])-mediated transactivation and the response was AF2-dependent in ZR75 breast cancer cells. C-and N-terminal NR-boxes (amino acids 1186-1182 and 73-69, respectively) were not necessary for coactivation of ER [alpha]. Analysis of DRIP150 deletion mutants identified a 23 amino acid sequence (811-789) required for coactivation. The 23 amino acid contained two regions at amino acids 789-794 and 795-804 which resembled [alpha] -helical motifs identified in Lanuguinosa lipase/histamine N-methyl transferase and hepatocyte nuclear factor 1, respectively. A squelching assay using specific point mutations within each [alpha] -helix showed that the NIFSEVRVYN (795-804) region was the critical sequence required for the coactivator activity of DRIP150.

The scope of the project is to examine whether or not the AhR complexed with the AhR-nuclear transporter (ARNT protein, binds to several xenobiotic responsive elements (XRE) strategically located at -539 bp (CCGTGGAA=Cyp1A1-like) and +20base pairs (bp) (GCGTG=XRE-1) from the transcription start site on exon-1A. Two additional XREs (GCGTG) have been localized at -107 bp in the intervening sequence upstream (XRE-2) and +218 bp (XRE-3) into exon-1B. Findings of the experiments conducted during the lest period include: 1) Completed testing of mutation contructs for XRE2 and investigated the effects of antagonists of the AhR and ER-alpha on regulation of BRCA-1 transcription by estrogen and the dioxin-like compound TCDD in breast cancer MCF-7 cells. 2) Characterized the mechanism of interaction between the ER-alpha and the AhR at the XRE-2 by chromatin immunoprecipitation assay (ChIP).

Book excerpt: In this study, we performed a functional mapping of the Estrogen Receptor (ER) promoter region and found a region in the 5' untranslated leader sequence that controlled ER transcription in ER-positive breast carcinoma cell lines. This region contained two binding sites for a transcription factor, Estrogen Receptor Factor-1 (ERF-1). ERF-1 was identified by gel shift assay and was present in ER-positive breast and endometrial carcinoma cells, but absent in ER-negative cell lines, thus inferring a role for ERF-1 in the regulation of ER transcription in breast carcinoma. Mutational analysis of the ERF-1 binding site showed that ERF-1 binding was correlated with transcriptional activity. ERF-1 was purified from MCF7 breast carcinoma cells using ion-exchange and DNA affinity chromatography and the cDNA was isolated from a MCF7 expression cDNA library. ERF-1 was identified as AP2gamma, a member of the AP2 transcription factor family. When binding specificity of ERF-1 was compared to AP2alpha using PCR-Assisted Binding Site selection and competitive gel shift assay, it was found that the two proteins recognized the same DNA sequence. Cotransfection of AP2alpha or AP2gamma with an ER promoter reporter construct demonstrated that activation of the ER promoter is dependent upon cell type.

A lively and varied cultural programme (classics to jazz) was also provided to maintain the interests and enthusiasms of the participants.
