Available in PDF, EPUB and Kindle. Book excerpt: Lipoxygenases and Their Products provides a cogent summary of the advances in understanding lipoxygenases and their products, emphasizing their pharmacology and pathophysiology; regulation and structure; physiological role; and receptors and receptor antagonists. This text also reviews the literature where specific receptor antagonists (as opposed to enzymatic inhibitors) have been used to characterize the pharmacologic action and the pathophysiologic roles of some of lipoxygenase products of arachidonic acid metabolism. This book is organized into 11 chapters and begins with an overview of the biochemical, pharmacological, and pathophysiological aspects of 5-lipoxygenase products of arachidonic acid metabolism in the respiratory, cardiovascular, renal, gastrointestinal, and central nervous systems. The discussion then shifts to human 5-lipoxygenase, emphasizing this enzyme purification, stimulatory factors, and membrane translocation. The reader is methodically introduced to 5-lipoxygenase activity in rats and pigs; the potential and therapeutic value of 5-lipoxygenase inhibitors; the distribution of 15-lipoxygenase in cells or tissues; and products formed through the 15-lipoxygenase pathway. This book also considers lipoxins and third-generation peptidoleukotriene receptor antagonists, and then concludes with a summary of the physiological and pathophysiological effects of leukotrienes and lipoxins in a variety of organs. This book is of interest to scientists interested in arachidonic acid and its metabolites, to scientists engaged in drug discovery-and-development activities in academia and the pharmaceutical industry, and to clinicians interested in diseases associated with leukotrienes and related products.

Book excerpt: It is only during the last decade that the functions of sinusoidal endothelial cells, Kupffer cells, hepatic stellate cells, pit cells and other intrahepatic lymphocytes have been better understood. The development of methods for isolation and co-culturing various types of liver cells has established that they communicate and cooperate via secretion of various intercellular mediators. This monograph summarizes multiple data that suggest the important role of cellular cross-talk for the functions of both normal and diseased liver. Special features of the book include concise presentation of the majority of detailed data in 19 tables. Original schemes allow for the clear illustration of complicated intercellular relationships. This is the first ever presentation of the newly emerging field of liver biology, which is important for hepatic function in health and disease and opens new avenues for therapeutic interventions.

Book excerpt: Prostaglandins, Leukotrienes, and Cancer is a multi-volume series that will focus on an emerging area of cancer research. In 1968, R.H. Williams first reported that elevated prostaglandin levels are present in human medullary car cinoma. Since that time, the concept that arachidonic acid metabolites may be in volved in cancer has expanded to include every aspect of the disease from cell transformation through metastasis. Prostaglandins and leukotrienes are generic terms used to describe a family of bioactive lipids produced from unsaturated fatty acids (principally from arachidonic acid) via the cyclooxygenase and lipoxygenase pathways, respec tively. Cyclooxygenase products consist of diverse products such as prosta glandin E, (POE,), prostacyclin (POI2) and thromboxane A2 (TXA2), whereas lipoxygenase products consist of hydroperoxy fatty acids and mono-, di- and tri-hydroxy acids including leukotrienes. The precursor fatty acids for the cyclooxygenase and lip oxygenase pathways are present in cellular phospholipids. This finding established an important control point in their biosynthesis-the release of substrate. This occurs in response to numerous stimuli that act at the cell surface. Dr. Bengt Samuelsson's extensive study of the metabolism of pros taglandins indicated that they are rapidly inactivated on a single pass through pulmonary circulation. Thus, they cannot act as circulating hormones and appear to be made on demand in or in the vicinity of target tissues leading to the concept that prostaglandins are local hormones or autocoids.

Available in PDF, EPUB and Kindle. Book excerpt: The endothelium, a monolayer of endothelial cells, constitutes the inner cellular lining of the blood vessels (arteries, veins and capillaries) and the lymphatic system, and therefore is in direct contact with the blood/lymph and the circulating cells. The endothelium is a major player in the control of blood fluidity, platelet aggregation and vascular tone, a major actor in the regulation of immunology, inflammation and angiogenesis, and an important metabolizing and an endocrine organ. Endothelial cells controls vascular tone, and thereby blood flow, by synthesizing and releasing relaxing and contracting factors such as nitric oxide, metabolites of arachidonic acid via the cyclooxygenases, lipoxygenases and cytochrome P450 pathways, various peptides (endothelin, urotensin, CNP, adrenomedullin, etc.), adenosine, purines, reactive oxygen species and so on. Additionally, endothelial ectoenzymes are required steps in the generation of vasoactive hormones such as angiotensin II. An endothelial dysfunction linked to an imbalance in the synthesis and/or the release of these various endothelial factors may explain the initiation of cardiovascular pathologies (from hypertension to atherosclerosis) or their development and perpetuation. Book excerpt: Arachidonic acid (AA) is an -6 polyunsaturated fatty acid found in the phospholipids of the membranes of the human bodys cells, and is abundant in the brain, muscles, and liver. This fatty acid is particularly obtained from meat products including chicken, beef, pork, and fishes. An interesting source of AA is through its accumulation in a green microalga, Myrmecia incisa, enhanced by nitrogen starvation. The general functions of AA and its metabolites including its association with coronary heart disease, oxidative stress and cancer, metabolic syndrome, insulin resistance, and its endocrine response to stress are also discussed.

This book was released on 1999-05-13 with total page 722 pages. Available in PDF, EPUB and Kindle. Book excerpt: Every aspect of immune function and host defense is dependent upon a proper supply and balance of nutrients. Severe malnutrition can cause significant alteration in immune response, but even subclinical deficits may be associated with an impaired immune response, and an increased risk of infection. Infectious diseases have accounted for more off-duty days during major wars than combat wounds or nonbattle injuries. Combined stressors may reduce the normal ability of soldiers to resist pathogens, increase their susceptibility to biological warfare agents, and reduce the effectiveness of vaccines intended to protect them. There is also a concern with the inappropriate use of dietary supplements. This book, one of a series, examines the impact of various types of stressors and the role of specific dietary nutrients in maintaining immune function of military personnel in the field. It reviews the impact of compromised nutrition status on immune function; the interaction of health, exercise, and stress (both physical and psychological) in immune function; and the role of nutritional supplements and newer biotechnology methods reported to enhance immune function. The first part of the book contains the committee's workshop summary and evaluation of ongoing research by Army scientists on immune status in special forces troops, responses to the Army's questions, conclusions, and recommendations. The rest of the book contains papers contributed by workshop speakers, grouped under such broad topics as an introduction to what is known about immune function, the assessment of immune function, the effect of nutrition, and the relation between the many and varied stresses encountered by military personnel and their effect on health.

Available in PDF, EPUB and Kindle. Book excerpt: Lipids have been in clinical use as components of intravenous nutrition for over 50 years. Over the last 15 years, new and improved lipids that include olive oil and/or fish oil have replaced the more traditional ones. These new lipids offer the opportunity to deliver high amounts of fatty acids and possess different functional properties: in particular, they can influence inflammatory processes, immune responses and hepatic metabolism. This book brings together articles written by leading international authorities in the area of intravenous lipids. Contributions discuss the latest findings in the field, ranging from pre-clinical research to the most recent clinical trials. Lipid functionality and utility in pediatric, adult surgical and critically ill patients are covered, as is the use of lipids in long-term home parenteral nutrition. Addressing a broad spectrum of topics, this publication provides a wealth of information for basic scientists, clinical researchers and clinical practitioners alike.
