The eye is a particularly valuable experimental system not only because its tissues provide examples of fundamental processes, but also because it is a prominent and easily accessible structure at very early embryonic ages. In order to provide an open forum for investigators working on all aspects of ocular development, a series of symposia on ocular and visual de velopment was initiated in 1973. A second purpose has been to foster communication between the basic research worker and the clinical community. It is our feeling that much can be learned on both sides from this interaction. The idea for an informal meeting allowing a maximum exchange of ideas originated with Dr. Leon Candeub, who supplied the necessary driving force that made the series a reality.

This volume provides a timely glimpse into the methods that have been developed to instigate, and the mechanisms that have been identified to drive, the process of nuclear reprogramming, chronicling how the field has developed over the last 50-60 years. Since the early 1950s a small number of notable experiments have provided significant impetus to the field, primarily the demonstration of reprogramming ability, first by the complex cytoplasmic milieu that constitutes the amphibian egg, then that of the mammalian egg, and finally that of the mammalian embryonic stem cell. Most recently, the demonstration that a limited pool of defined molecules is capable of reprogramming a multitude of cell types has provided massive impetus and facilitated transition towards realistic therapeutic application. We have therefore reproduced some of the key articles that elegantly document these dramatic stages of development of the field in an inclusive appendix to the book, for the benefit of readers keen to investigate the history of how the field of stem cell biology has evolved. Owing to the ever broadening nature of this field, and the incredible rate at which it is evolving, the main content of this volume focuses on areas that have shown significant movement in recent years, are most likely to translate into personalized therapeutic application, and thus provide greatest potential for significant impact on human health in the not too distant future. We recognize that research into many other disease states and cell types are all equally worthy of discussion. We would therefore like to acknowledge those researchers involved whose work we have not been able to include in this volume. Book excerpt: Many modern geneticists attempt to elucidate the molecular basis of phenotype by utilizing a battery of techniques derived from physical chemistry on subcellular components isolated from various species of organisms. Volume 5 of the Handbook of Genetics provides explanations of the advantages and shortcomings of some of these revolutionary tech niques, and the nonspecialist is alerted to key research papers, reviews, and reference works. Much of the text deals with the structure and func tioning of the molecules bearing genetic information which reside in the nucleus and with the processing of this information by the ribosomes resid ing in the cytoplasm of eukaryotic cells. The mitochondria, which also live in the cytoplasm of the cells of all eukaryotes, now appear to be separate little creatures. These, as Lynn Margulis pointed out in Volume 1, are the colonial posterity of migrant prokaryotes, probably primitive bacteria that swam into the ancestral precursors of all eukaryotic cells and remained as symbionts. They have maintained themselves and their ways ever since, replicating their own DNA and transcribing an RNA quite different from that of their hosts. In a similar manner, the chloroplasts in all plants are self-replicating organelles presumably derived from the blue-green algae, with their own nucleic acids and ribosomes. Four chapters are devoted to the nucleic acids and the ribosomal components of both classes of these semi-independent lodgers. Finally, data from various sources on genetic variants of enzymes are tabulated for ready reference, and an evaluation of this information is attempted.
