Book excerpt: Viral vectors are superior tools for gene therapy and as a genetic vaccine platform because viruses have evolved to efficiently infect and transfer their genomes to cells. Several impressive successes in viral vector-based gene therapies have been reported in humans, including restoration of vision in patients with Leber’s congenital amaurosis by retinal gene transfer and cures for severe immune deficiencies by gene transfer to hematopoietic stem cells. However, the mammalian immune system has evolved in parallel to fend off invading pathogens such as viruses. Innate and antigen-specific adaptive immune responses against viral vectors and therapeutic transgene products pose serious hurdles for successful gene therapy. Pre-existing immunity in humans, resulting from prior exposure to the parent virus that forms the basis for the gene transfer vehicle may be derived from, often prevents efficient gene transfer. This problem also reduces our ability to use certain vectors for genetic vaccination or in anti-cancer therapy. For these reasons, the gene transfer community has been extensively studying the mechanisms of immune responses against viral vectors and has started to develop strategies and protocols to block or circumvent such responses. Choice, design and engineering of a vector as well as the route of administration/target tissue can be optimized/ altered to minimize immune responses or evade pre-existing immunity. Immune suppression and modulation strategies are being developed in order to minimize inflammation, prevent antibody or T cell responses against vectors, and to promote tolerance to therapeutic gene products. Combinations of these approaches will likely facilitate clinical applications of gene therapy for many target diseases and also aid in vaccine development.

Design, capability and their evolution are introduced into current theories of trade to explain the reality of international trade in the early twenty-first century and the possibility of design-based comparative advantage is explored. Toward that end, the concepts of design, architecture, organizational capability and productivity are introduced, as are their interactions and evolution. The author starts from the fact that firms’ selection of design locations precedes that of production locations and that a new product’s initial production location is usually the same as its design location. In other words, design matters in explaining today’s trade phenomena. Thus, this book analyzes product design and its evolution in the context of the comparative advantage theory. The author argues that the concept of Ricardo’s comparative advantage must be reinterpreted in a more dynamic way than in the past, with changing labor input coefficients treated as variables and driven by international capability-building competition between factories. Book excerpt: The book expresses the conviction that the art of creating tools – Greek techne – changes its character together with the change of civilization epochs and co-determines such changes. This does not mean that tools typical for a civilization epoch determine it completely, but they change our way of perceiving and interpreting the world. There might have been many such epochs in the history of human civilization (much more than the three waves of agricultural, industrial and information civilization). This is expressed by the title Technen of the book, where n denotes a subsequent civilization epoch. During last fifty years we observed a decomposition of the old episteme (understood as a way of creating and interpreting knowledge characteristic for a given civilization epoch) of modernism, which was an episteme typical for industrial civilization. Today, the world is differently understood by the representatives of three different cultural spheres: of strict and natural sciences; of human and social sciences (especially by their part inclined towards postmodernism) and technical sciences that have a different episteme than even that of strict and natural sciences. Thus, we observe today not two cultures, but three different episteme. The book consists of four parts. First contains basic epistemological observations, second is devoted to selected elements of recent history of information technologies, third contains more detailed epistemological and general discussions, fourth specifies conclusions. The book is written from the cognitive perspective of technical sciences, with a full awareness – and discussion – of its differences from the cognitive perspective of strict sciences or human and social sciences. The main thesis of the book is that informational revolution will probably lead to a formation of a new episteme. The book includes discussions of many issues related to such general perspective, such as what is technology proper; what is intuition from a perspective of technology and of evolutionary naturalism; what are the reasons for and how large are the delays between a fundamental invention and its broad social utilization; what is the fundamental logical error (using paradoxes that are not real, only apparent) of the tradition of sceptical philosophy; what are rational foundations and examples of emergence of order out of chaos; whether civilization development based on two positive feedbacks between science, technology and the market might lead inevitably to a self-destruction of human civilization; etc.
