Available in PDF, EPUB and Kindle. Book excerpt: The process of inflammation, which causes the swelling and redness around a wound, is a vital part of the body's system for fighting off infections. When the body is hurt, the immune system produces chemical signals telling cells to multiply without dying, allowing skin to close over a gash, for example. Other chemicals spur the growth of new blood vessels to feed the recovering tissue. Scientists have linked inflammation to cancer and recently to heart disease in several ways. Doctors suspect that long-term inflammation or infection is involved in up to 20 per cent of cancers, including those of the oesophagus, colon, skin, stomach, liver, bladder, breast and some kinds of lymphoma. C-reactive protein (CRP) is one of the acute phase proteins that increase during systemic inflammation. It's been suggested that testing CRP levels in the blood may be a new way to assess cardiovascular disease risk. A high sensitivity assay for CRP test (hs-CRP) is now widely available. This new book presents recent leading-edge research from around the world.

Edited by the undisputed pioneers of this area of research, this volume is the first devoted to its topic. It contains thirteen chapters, each written by leaders in their respective fields, that summarize the state of the art in developing novel, gene based treatments for inflammatory diseases. As well as providing an introduction to the basic concepts of gene therapy and the use of naked DNA approaches, the book describes the advances that have been made in applying them to arthritis, lupus, multiple sclerosis, diabetes, Sjogren`s syndrome and transplantation. One chapter is devoted to discussing the first human clinical trials that apply gene therapy to the treatment of an inflammatory disease. As well as providing novel therapeutic approaches, gene therapy facilitates the development of new and improved animal models of disease; a chapter describing these advances is also included. As an up-to-date, timely book written by th

Available in PDF, EPUB and Kindle. Book excerpt: The purpose of this volume in the series Progress in Inflammation Research is to pro vide the biomedical researcher with a description of the state of the art of the devel opment and use of animal models of diseases with components of inflammation. Par ticularly highlighted are those models which can serve as in vivo correlates of diseases most commonly targeted for therapeutic intervention. The format is designed with the laboratory in mind; thus it provides detailed descriptions of the methodologies and uses of the most significant models. Also, new approaches to the development of future models in selected therapeutic areas have been highlighted. While emphasis is on the newest models, new information broadening our understanding of several well-known models of proven clinical utility is included. In addition, we have pro vided coverage of transgenic and gene transfer technologies which will undoubtedly serve as tools for many future approaches. Provocative comments on the cutting edge and future directions are meant to stimulate new thinking. Of course, it is important to recognize that the experimental use of animals for human benefit carries with it a solemn responsibility for the welfare of these animals. The reader is referred to the section on current regulations governing animal use which addresses this concern. To fulfill our purpose, the content is organized according to therapeutic areas with the associated models arranged in subcategories of each therapeutic area.

Book excerpt: As our understanding of immune mediated chronic inflammatory diseases (IMIDs) grows, it becomes more and more clear that these conditions result from the convergence of a multitude of pathogenic mechanisms whose relative individual contribution is different in different patient subsets. Promising new technologies have been conceived that address the hypotheses that targeting multiple pathways simultaneously, selectively delivering therapeutics to areas of inflammation and/or resetting the immune system, could take efficacy to new levels. However, we have long waited for the arrival of some of these technologies to the bedside, or even far enough in the drug development process in spite of the initial enthusiasm. Some of the examples covered in this book include bispecific antibodies and genomic medicines, microparticles and targeted delivery of drugs to inflamed vasculature. Most published reviews and book chapters on novel therapies for inflammatory diseases describe positive attributes of molecules or technologies under investigation and the rationale for developing them into therapeutics. The originality and potential value of this book is not in the description of these targets or technologies from the point of view of their structure or mechanism of action exclusively, but rather, in making an effort to critically address the question of what is needed to move these technologies into the clinic. Has the technology not made it past the preclinical stage and why? Has it already been tested in humans and failed? What are the potential reasons behind those failures? What do experts in each field believe can be done better to increase the probabilities of success? In addition, the authors address the competitive landscape and summarize clinical studies that have failed in the respective area. They talk about the patient populations that would be required for the successful conduction of a clinical trial to test certain molecules, and they proactively share their views regarding both the potential and the drawbacks of targets or methodologies.

Available in PDF, EPUB and Kindle. Book excerpt: How are cancer and inflammation interrelated mechanistically and clinically? Though extensive literature exists on the topic "Cancer and Inflammation", there are relatively few texts that have truly integrated the two in spite of the many common mechanisms shared by their processes. Certainly, areas such as cytokines, growth factors, proliferation, signal transduction and angiogenesis, for example, are found in both. Yet, the dynamics of how these common mechanisms are maybe interrelated in the pathologies of the two is not widely covered. Such coverage, as presented in this volume, may help further understanding and bring new approaches to therapeutics. The first section of the book discusses inflammatory mechanisms, studied in cellular and animal studies. The second part concentrates on clinical studies with antiinflammatory drugs in cancer treatment. The volume is written for biomedical researchers in the health care industry and in academia who are working in these areas.

Available in PDF, EPUB and Kindle. Book excerpt: Regulatory T-cells are essential components of the immune system, and several different subsets of regulatory T-cells have been described. Considerable regulatory function has been attributed to the CD4+CD25+ T-cell subset. These cells act by suppressing adaptive and possibly innate immune responses thereby maintaining or restoring the balance between immunity and tolerance. The suppressive effects of CD4+CD25+ regulatory T-cells are cell-contact dependent. Recent developments and viewpoints in the field of CD4+CD25+ regulatory T-cells as well as the potential use of regulatory T-cells in immunotherapy of inflammatory diseases are discussed in this volume. By linking data from experimental models with recent findings from the clinic, this book will be of interest to immunologists and other biomedical researchers as well as clinicians interested in the regulation and manipulation of the immune response during inflammatory disease.
