Book excerpt: The prodrug approach is a promising and well established strategy for the development of new entities that possess superior efficacy, selectivity and reduced toxicity. Hence an optimised therapeutic outcome can be accomplished using this approach. Prodrug design is becoming more elaborate in the development of efficient and selective drug delivery systems. The targeted prodrug approach, in combination with gene delivery and controlled expression of enzymes and carrier proteins, is a promising strategy for precise and efficient drug delivery and enhancement of the therapeutic effect. This book describes in details all prodrug approaches and examples of prodrugs that succeeded to enter the market. There are two major prodrug design approaches that are considered as widely used among all other approaches: the targeted drug design approach by which prodrugs can be designed to target specific enzymes or carriers by considering enzyme-substrate specificity or carrier-substrate specificity in order to overcome various undesirable drug properties. Examples for such approach is the antibody-directed enzyme prodrug therapy (ADEPT), gene-directed enzyme prodrug therapy (GDEPT), virus-directed enzyme prodrug therapy (VDEPT) and GDEPT. In addition, this book describes in details a novel prodrug chemical approach which is based on intramolecular reactions that were utilised to understand how enzymes exert their high catalysis. The information gained from the experimental and theoretical calculations on these enzyme models was used to design efficient chemical moieties to be utilised as prodrug linkers with the potential to release the corresponding parent drugs in a slow or fast release manner. Several prodrugs for commonly used drugs suffer from low bioavailability or/and bitter sensation were designed using quantum mechanics methods (DFT and ab initio) and recently a large number among these prodrugs were synthesised. Examples of such prodrugs are presented in the different chapters of the book.

This book was released on 2012-12-06 with total page 391 pages. Available in PDF, EPUB and Kindle. Book excerpt: The evident rapid expansion of scientific work and intense interest in both experimental and clinical aspects of new drug delivery systems provided strong motivation for planning this symposium. In designing the program, speakers were identified for their particular expertise in a wide range of topics such as dermal delivery systems, pro-drugs, oral prolonged release, rate-controlled drug delivery, the pharmacokinetics of drug release systems, the synthesis of polymeric drug carriers and the refinement of drug delivery pumps. Because of the considerable involvement of diverse scientists from laboratories around the world where investigations relevant to the topic are now being pursued, a deliberate effort was made to invite international leaders in the field to share their knowledge and experimental outcomes. Thus, plenary papers and panel discussions were offered by organic chemists, bioengineers, patholo gists, material scientists, physical chemists, and pharmacokineti cists from academic and industrial laboratories in some dozen countries. This book which records the presentations offered at the sym posium covers a broad array of topics ranging from general overviews of the physicochemical concepts and analytical methodology which underpin the refinement of drug delivery systems and the tissue responses associated with the use of such systems through detailed discussions of a variety of current approaches employed in the development of new systems.

Available in PDF, EPUB and Kindle. Book excerpt: The application of drug delivery is a valuable, cost-effective lifecycle management resource. By endowing drugs with new and innovative therapeutic benefits, drug delivery systems extend products’ profitable lifecycle, giving pharmaceutical companies competitive and financial advantages, and providing patients with improved medications. Formulation development is now being used to create new dosage forms for existing products, which not only reduces the time and expense involved in new drug development, but also helps with regard to patent protection and bypassing existing patents. Today’s culture demands convenience, a major factor determining adherence to drug therapy. Over the past few years, patient convenience-oriented research in the field of drug delivery has yielded a range of innovative drug-delivery options. As a result, various drug-delivery systems, including medicated chewing gums, oral dispersible tablets, medicated lozenges and lollipops, have now hit the market and are very popular. These dosage forms offer a highly convenient way to dose medications, not only for special population groups with swallowing difficulties, such as children and the elderly, but for the general populace as well. This book provides valuable insights into a number of formulation design approaches that are currently being used, or could be used, to provide new benefits from existing drug molecules.
