Book excerpt: Time-of-flight secondary ion mass spectrometry (ToF-SIMS) is a powerful technique for the analysis of organic surfaces and interfaces for many innovative technologies. However, despite recent developments, there are still many issues and challenges hindering the robust, validated use of ToF-SIMS for quantitative measurement. These include: the lack of metrology and fundamental understanding for the use of novel cluster primary ion beams such as C60n+ and Ar2000 +; the need for validated and robust measurement protocols for difficult samples, such as those with significant micron scale surface topography; the lack of guidance on novel data analysis methods including multivariate analysis which have the potential to simplify many time-consuming and intensive analyses in industry; and the need to establish best practice to improve the accuracy of measurements. This thesis describes research undertaken to address the above challenges. Sample topography and field effects were evaluated experimentally using model conducting and insulating fibres and compared with computer simulations to provide recommendation to diagnose and reduce the effects. Two popular multivariate methods, principal component analysis (PCA) and multivariate curve resolution (MCR), were explored using mixed organic systems consisting of a simple polymer blend and complex hair fibres treated with a multi-component formulation to evaluate different multivariate and data preprocessing methods for the optimal identification, localisation and quantification of the chemical components. Finally, cluster ion beams C60 n+ and ArSOO-2S00 + were evaluated on an inorganic surface and an organic delta layer reference material respectively to elucidate the fundamental metrology of cluster ion sputtering and pave the way for their use in organic depth profiling. These studies provide the essential metrological foundation to address frontier issues in surface and nanoanalysis and extend the measurement capabilities ofToF-SIMS.

Book excerpt: Time of Flight Secondary Ion Mass Spectrometry (ToF SIMS) is a powerful tool for advanced surface analysis. It produces large data sets, which consist of mass spectra at each pixel at an imaged area. Many ToF SIMS instruments use a type of detector that suffers from two problems that lead to non-linearity in the measured sample properties: detector saturation and dead time. Linearity in this type of system is defined as the proportionality of the measured and true ion counts. Non-linearity can influence the interpretation of the data with methods such as multivariate analysis. "Detector saturation" happens when more than one ion arrives at the detector in the time interval related to one specific channel but the detector records only a single count. "Dead time" is when one event happens at a certain channel and the detector become insensitive to subsequent ions arriving within the dead time window. These problems both lead to under-counting of ions. In this thesis, we mainly focus on correcting for the dead time effects. Using extensive simulations, we first characterize the adverse effects of dead time on the output and evaluate quality of existing ways to "correct" for dead time effects. Then, we propose a novel method using the Maximum Likelihood Estimation (MLE) to estimate the true spectrum for the measured data. Specifically, we incorporate the statistical distribution of the dead time affected data in MLE, which leads to a new method for dead time correction.

Available in PDF, EPUB and Kindle. Book excerpt: Time of flight secondary ion mass spectrometry, TOF-SIMS, is a highly surface sensitive analytical technique that provides information about composition with submicron lateral resolution. For select materials, TOF-SIMS provides unparalleled sensitivity along with excellent reproducibility, and as a mass spectrometric technique, it also provides excellent specificity. Of the analytical methods available, it is among the most surface sensitive, but the physical principles that underlie it are also the least understood. This volume describes the instrumentation, the physical principles behind the technique to the extent they are understood, and provides a practical approach for the interpretation of TOF-SIMS data. The use of advanced data processing methods such as multivariate statistics are described in a readily approachable manner. Given a basic background in undergraduate chemistry and physics, the book will be of use to any student with an interest in the technique.

Book excerpt: The first application of Time-of-Flight Secondary Ion Mass Spectroscopy (ToF-SIMS) to an aged plutonium surface has resulted in a rich set of surface chemistry data, as well as some unexpected results. FY15 was highlighted by not only the first mapping of hydrogen-containing features within the metal, but also a prove-in series of experiments using the system's Sieverts Reaction Cell. These experiments involved successfully heating the sample to ~450 oC for nearly 24 hours while the sample was dosed several times with hydrogen, followed by an in situ ToF-SIMS analysis. During this year, the data allowed for better and more consistent identification of the myriad peaks that result from the SIMS sputter process. In collaboration with the AWE (U.K), the system was also fully aligned for sputter depth profiling for future experiments.

The complexity of the metabolome is outstanding, requiring the use of complementary analytical platforms and methods for its quantitative or qualitative profiling. In alignment with the selected analytical approach and the study aim, sample collection and preparation are critical steps that must be carefully selected and optimized to generate high-quality metabolomic data. This book showcases some of the most recent developments in the field of sample preparation for metabolomics studies. Novel technologies presented include electromembrane extraction of polar metabolites from plasma samples and guidelines for the preparation of biospecimens for the analysis with high-resolution μ magic-angle spinning nuclear magnetic resonance (HR-μMAS NMR). In the following chapters, the spotlight is on sample preparation approaches that have been optimized for diverse bioanalytical applications, including the analysis of cell lines, bacteria, single spheroids, extracellular vesicles, human milk, plant natural products and forest trees.

Available in PDF, EPUB and Kindle. Book excerpt: Applying screened trapezoidal potential barriers in the drift tube of a time-of-flight mass spectrometer produces flighttime shifts that can be used to separate and identify products of ion dissociation processes occurring between the ion source and the potential barrier. The flight-time shift is a function of the retarder voltage and mass-to-charge ratios of the fragment and parent ions, expressed by the flight-time shift equation. When the retarder voltage is known, analytic solu tions of this equation allow the retarded ions (fragment or parent) to be identified from the measured flight-time shifts. When the mass-to-charge ratios of these ions are known, similar solutions allow the retarder voltage that will produce a given flight-time shift to be predicted. For part IV see AD-429 438. This book was released on 2002 with total page pages. Available in PDF, EPUB and Kindle. Book excerpt: The present thesis deals with the impact ionization processes appearing at hypervelocity impacts of microparticles on solid surfaces. With a newly developed dust particle source, applied to the Heidelberg Dust Accelerator facility, aluminium, carbon, sodium contaminated carbon, iron and latex projectiles were shot on the micrometeorite detectors CDA and CIDA. The projectile masses cover a range of 10^-18 to 10^-12 kg, the projectile densities a range of 1100 - 7900 kg/m^3 and the impact speeds a range of 2 - 70 km/s. The resulting data show different impact ionization processes depending on the impact speeds: for low speeds (v 6 km/s) dominates surface ionization of target contaminants, for high speeds (v 18 km/s) volume ionization of the target and projectile material is dominating. The charge yield in the intermediate impact speed regime shows a reduced increase due to energy consumption by melting and vaporization processes. Time-of-flight mass spectroscopy, provided by both instruments, enabled the investigation of the chemical composition of the impact plasma depending on the projectile type and on the impact speed. Alkaline ions (Na, K) dominate the mass spectra at low impact speeds. For higher impact speeds appear target ions (Rh), projectile material related ions and hydrogen ions (H). The systematics of characteristic atomic and molecular ion species in the mass spectra helps to get clues on the chemical nature of unknown projectiles in space. An improved model on specific ion yields can be given. Furthermore a lot of results which are important for the instrument calibration and the understanding of flight data, like global charge yields, rise times and sensitivities were obtained. First measurements on the inner instrument wall region are provided.
