The First Conference was held at the Aviya Sonesta Hotel in Eilat, Israel, on March 24-27, 1985. The record of this First Conference was published by Plenum Press in 1986 as Volume 29 in Advances in Behavioral Biology, under the title "Alzheimer's and Parkinson's Diseases: Strategies for Research and Development." We are happy that the comprehensive texts of the oral and poster presentations of the Second Conference could again be published within the framework of this series. Since the First Conference in 1985, rapid progress has been made in both basic and 'therapeutic aspects of these diseases. About 700 scientists from allover the world participated in the Second Conference, and 300 papers were presented in oral and poster sessions. Many people and organizations have helped to organize this multi disciplinary international conference and hence have contributed to the scientific quality of these two volumes. Book excerpt: Collectively, neurodegenerative diseases are characterized by chronic and progressive loss of neurons in discrete areas of the brain, producing debilitating symptoms such as dementia, loss of memory, loss of sensory or motor capability, decreased overall quality of life eventually leading to premature death. Two types of cell death are known to occur during neurodegeneration: (a) apoptosis and (b) necrosis. The necrosis is characterized by the passive cell swelling, intense mitochondrial damage with rapid loss of ATP, alterations in neural membrane permeability, high calcium influx, and disruption of ion homeostasis. This type of cell death leads to membrane lysis and release of intracellular components that induce inflammatory reactions. Necrotic cell death normally occurs at the core of injury site. In contrast, apoptosis is an active process in which caspases (a group of endoproteases with specificity for aspartate residues in protein) are stimulated. Apoptotic cell death is accompanied by cell shrinkage, dynamic membrane blebbing, chromatin condensation, DNA laddering, loss of phospholipids asymmetry, low ATP levels, and mild calcium overload. This type of cell death normally occurs in penumbral region at the ischemic injury site and in different regions in various neurodegenerative diseases.

The cause of this disease is unknown; indeed, evidence increasingly suggests that there may be multiple Alzheimer-type syndromes with different etiologies, analogous to different types of psychosis. Currently there are no means to prevent the disease, slow its progress or reverse its neurodegenerative consequences. With few exceptions, clinical trials of a variety of compounds have resulted in patient responses that are disappointing with respect to both the proportion of responders and the magnitude of the responses. Novel approaches to the treatment of Alzheimer's disease are clearly warranted. For this reason, we organized the First Suncoast Workshop on the Neurobiology of Aging in St. Petersburg, Florida, which took place from February 26-March 1, 1989. This workshop focused on novel treatments and models for Alzheimer's disease and represented a cooperative venture among academia, government and industry, both in its participants and sponsorship. The Center for the Neurobiology of Aging at the University of Florida, the National Institute on Aging and Taiho Pharmaceutical Corporation in Japan sponsored the workshop in which scientists from the North America, Europe, Japan and other parts of Asia participated.

Available in PDF, EPUB and Kindle. Book excerpt: "Alzheimer’s disease (AD) is a neurodegenerative disorder that advances through a unique cascade of characteristic neuropathological features. Distinguishing features include the accumulation of amyloid beta plaques and neurofibrillary tangles (NFTs) in the brain, accompanied by learning and memory impairments. The disease is projected to begin years-to-decades before clinical diagnosis. Accordingly, prodromal biomarkers are a necessity with early diagnosis as the goal. Neurochemical measures obtained via non-invasive brain imaging have potential to serve as early, AD biomarkers. Evidence suggests that alterations in the concentrations of certain metabolites are related to inflammation. It is known that while inflammation is ubiquitous in the progressed disease state, it is also apparent during initial stages. Of interest, a paradoxical increase in neuronal activity during early stages of AD has been noted, with a suspected link to an initial inflammatory response via proinflammatory cytokines. Early evidence suggests that alongside their role in immune mediation, cytokines play a critical part in maintaining optimal synaptic activity, and that early deviations from this role may contribute directly to downstream AD pathogenesis. Therefore, as magnetic resonance spectroscopy (MRS) has the capacity to noninvasively quantify neurochemical concentrations, it is an appropriate tool to pinpoint early fluctuations associated with inflammation and elevated cytokine activation.A greater understanding of AD requires an improved appreciation for its initial evolution. This requires, not only investigation into potential preclinical, neurochemical biomarkers, but furthermore, an examination into any parallel cognitive deficits. This project investigated prodromal AD in a transgenic (Tg) rodent model from early adulthood to middle age. It utilized in vivo proton magnetic resonance spectroscopy (1H MRS) to assess longitudinal changes in neurochemistry and the Barnes maze (BM) to evaluate spatial learning and memory. The central research objective was the longitudinal characterization of neurochemistry and behaviour in the TgF344-AD rodent model during the initial disease state, to highlight abnormalities that precede diagnosable AD.The study was conducted from approximately 4 to 10 months of age, in both TgF344-AD rats and their wildtype (WT) controls. Five evenly spaced magnetic resonance (MR) scans were acquired during this time (1 every 6 weeks), in addition to three sessions of behavioural testing (1 every 2 months). The BM was implemented to test spatial learning and memory ability, and longitudinal changes in metabolic levels were evaluated through linear mixed effects modeling.Altered neurochemistry was recorded between genotypes and across time in prodromal stages of AD. More specifically, Tg rats had significantly lower levels of glutamine (Gln), a precursor to glutamate (Glu), following disease initiation and increasing with age. In addition, with time Glu levels fell and myo-inositol (Ins) levels rose, independent of genotype. N-acetyl aspartate (NAA), reportedly reduced at later stages of AD, remained unchanged in Tg relative to WT rats. Neurochemical changes were not accompanied by a significant loss in cognitive ability, and thus appear to precede characteristic cognitive impairments.This research demonstrates that altered neurochemistry materializes in early AD and occurs before the onset of measurable behavioural deficits. A better understanding of preclinical AD requires further investigation into the relationship between neurochemistry, cognition, neuroanatomy, and the concentrations of proinflammatory mediators. Consequently, this could contribute to novel diagnostic and treatment techniques through the identification of reliable, early biomarkers"--

" More than any statistics, these words convinced me that Tropon's decision to leave the field of classic CNS pharmacology and move into the field of gerontopsychopharmacology was the right one, even though we knew that success would be uncertain and that, even if it finally comes it will be many years hence. At this point, let me add a personal comment: each one of us is judged by his or her own success. We live in a competitive society where success counts. This applies not only to the businessman, but also to the scientist, in particular, to those in industry.

Senile dementia is now widely recognized as a medical and social problem likely to reach epidemic proportions by the turn of the century. By that time it is esti mated that almost 20% of the population in most developed countries will be over the age of 65 and at a conservative estimate 1 in 10 of them will suffer from a dementing illness. Many symposia have appeared over the last few years describing the neuropathological and neurochemical deficiencies in Alzheimer's and other forms of dementia, the neuropsychological features of the disease, and attempts to treat it. In this volume, we have selected topics and authors who are beginning to question some of the earlier assumptions and to ask different questions about dementia. In the first four chapters the neuropathology and neu rochemistry of dementia are reevaluated. It is important to understand the relationship between the formation of amyloid plaques and neurofibrillary tangles, the progressive degeneration in cortex, and the neurochemical deafferentation of cortex. In particular, the possibility is considered that the most severe pathology is seen in a well-defined limbo/cortical circuitry known to be involved in mnemonic processing. The growing interest in the genetic determinants of familial Alz heimer's is reviewed. It is also recognized that detailed comparisons of the neural and psychological characteristics of the various degenerative dis eases that impair cognitive processing may be valuable.
